Claims (11)
- 화학식 V의 화합물을 상응하는 활성화 후에 적합한 용매 속에서 디-(C1-4-알킬)-포스포노아세트산과 반응시키는 단계(a) 및생성된 화학식 VI의 화합물을 화학식 의 알데히드(여기서, R3 및 R4는 동일하거나 상이할 수 있으며, 각각 직쇄 또는 측쇄 C1-C4알킬 그룹이다)와 반응시키거나, 적합한 유기 또는 무기 염기를 사용하여 상응하는 알데히드 등가물과 반응시키는 단계(b)를 포함하는, 화학식 VII의 화합물의 제조방법.화학식 V화학식 VI화학식 VII위의 화학식 V, 화학식 VI 및 화학식 VII에서,X는 메틸렌 그룹 또는 질소 원자이고,Ra는 벤질, 1-페닐에틸 또는 3-클로로-4-플루오로페닐 그룹이고,R1, R2 및 R4는 직쇄 또는 측쇄 C1-4-알킬 그룹이고, 이들은 동일하거나 상이할 수 있다.
- N4-(3-클로로-4-플루오로-페닐)-7-(테트라하이드로푸란-3-일옥시)퀴나졸린-4,6-디아민을 상응하는 활성화 후에 적합한 용매 속에서 디-(C1-4-알킬)-포스포노아세트산과 반응시키는 단계(a) 및생성된 디알킬에스테르 {[4-(3-클로로-4-플루오로-페닐아미노)-7-((S)-테트라하이드로푸란-3-일옥시)-퀴나졸린-6-일카바모일]-메틸}-포스포네이트를 적합한 유기 또는 무기 염기를 사용하여 상응하는 (디메틸아미노)-아세트알데히드-디알킬아세탈로부터 동일 반응계 내에서 제조된 알데히드와 반응시키는 단계(b)를 포함하는, 4-[(3-클로로-4-플루오로페닐)아미노]-6-{[4-(N,N-디메틸아미노)-1-옥소-2-부 텐-1-일]아미노}-7-((S)-테트라하이드로푸란-3-일옥시)-퀴나졸린의 제조방법.
- 제2항에 있어서, 단계(a)에서와 같이, 디에틸포스포노아세트산이 반응물로서 사용되는 방법.
- 제1항 또는 제2항에 있어서, 단계(b)에서, DBU(1,5-디아자비사이클로[4.3.0]논-5-엔), 수산화나트륨 또는 수산화칼륨이 염기로서 사용되는 방법.
- 제4항에 있어서, 단계(b)에서, 수산화칼륨이 염기로서 사용되는 방법.
- 제1항에 따르는 단계(a) 및 단계(b)와 함께, 생성된 4-[(3-클로로-4-플루오로페닐)아미노]-6-{[4-(N,N-디메틸아미노)-1-옥소-2-부텐-1-일]아미노}-7-((S)-테트라하이드로푸란-3-일옥시)-퀴나졸린을 가열하면서 적합한 용매 속에서 말레산과 반응시킴으로써 디말레에이트로 전환시키는 단계(c)를 포함하는, 4-[(3-클로로-4-플루오로페닐)아미노]-6-{[4-(N,N-디메틸아미노)-1-옥소-2-부텐-1-일]아미노}-7-((S)-테트라하이드로푸란-3-일옥시)-퀴나졸린의 제조방법.
- 제6항에 있어서, 에탄올 또는 이소프로판올이, 임의로 물을 첨가하여, 용매로서 사용되는 방법.
- 제6항 또는 제7항에 있어서, 적어도 2당량의 말레산이 사용되는 방법.
- 4-[(3-클로로-4-플루오로페닐)아미노]-6-{[4-(N,N-디메틸아미노)-1-옥소-2-부텐-1-일]아미노}-7-((S)-테트라하이드로푸란-3-일옥시)-퀴나졸린 디말레에이트.
- 4-[(3-클로로-4-플루오로페닐)아미노]-6-{[4-(N,N-디메틸아미노)-1-옥소-2-부텐-1-일]아미노}-7-((S)-테트라하이드로푸란-3-일옥시)-퀴나졸린 디말레에이트를 임의로 하나 이상의 불활성 담체 및/또는 희석제와 함께 포함하는 약제학적 조성물.
- 양성 또는 악성 종양의 치료 및 위장관, 담관 및 담낭 질환의 치료 뿐만 아니라, 기도 및 폐 질환의 예방 및 치료용 약제학적 조성물을 제조하기 위한, 4-[(3-클로로-4-플루오로페닐)아미노]-6-{[4-(N,N-디메틸아미노)-1-옥소-2-부텐-1-일]아미노}-7-((S)-테트라하이드로푸란-3-일옥시)-퀴나졸린 디말레에이트의 용도.
