表6は、「親トリガー」から由来するか、または「親トリガー」に関連している(表1を参照されたい)配列を有するヘアピン(dsRNAを形成するためにハイブリッド形成するセンス領域及びアンチセンス領域をいずれも含有する1つのRNA転写物)の形態でdsRNAトリガーを提供し、親トリガーは、Leptinotarsa decemlineataに対して殺虫力を有すると判断された(表1、表3及び表4を参照されたい)。ヘアピントリガーは、例えば、細菌細胞内または植物細胞内で発現させるために、適切なプロモーターまたは他の要素を有する発現構造体に提供されるとき、インビトロ発現またはインビボ発現に好適である。表6に開示される非限定的実施形態は、それぞれT7プロモーター(各ヘアピン配列においてヌクレオチド位置1〜17に配置される)、及びセンス領域とアンチセンス領域との間に配置される「ループ」またはスペーサーを含み、ループは、非特異的(標的遺伝子のいかなる部分にも相補的でないまたは同一でない)ヌクレオチドを含む。当業者は、所与の細胞型中での発現に好適な異なるプロモーターと異なるスペーサー配列若しくはループ配列との組み合わせで、ヘアピンのセンス領域及びアンチセンス領域が有用であること(さもなければ、センス領域及びアンチセンス領域の接合部のヌクレオチドがヘアピン内で必要な「回転」若しくは最小ループを形成する場合は全くないこと)は、すぐに理解するであろう。また、当業者は、類似の組換え型DNA構造体が、表1〜5に提供された平滑末端化dsRNAトリガーに対応するヘアピンdsRNAトリガーをコードするか、または標的遺伝子配列群に提供される標的遺伝子を標的にするように容易に設計されることは理解するであろう。

- 前記方法は、配列番号:730の少なくとも21の連続するヌクレオチドに相補的であるヌクレオチド配列を含む少なくとも1つのポリヌクレオチドを含む組成物を前記植物に局所投与することを含み:前記組成物は、キャリア剤、界面活性剤、陽イオン性脂質、オルガノシリコーン、オルガノシリコーン界面活性剤、ポリヌクレオチド除草剤分子、非ポリヌクレオチド除草剤分子、非ポリヌクレオチド殺虫剤、毒性緩和剤及び昆虫成長調整剤からなる群から選択される1つ以上の成分を更に含む、請求項1に記載の方法。
- 前記方法は、前記Leptinotarsa種を有効量の二本鎖RNA含有溶液と接触させることを含み、前記二本鎖RNAの少なくとも一本鎖は、リボソームタンパク質または前記遺伝子から転写されたRNAをコードする遺伝子の少なくとも21の連続するヌクレオチドに相補的であり、前記Leptinotarsa種は、Leptinotarsa decemlineataであり、RNA干渉が誘導され、Leptinotarsa decemlineataの死滅が発生し、前記リボソームタンパク質は、リボソームL7タンパク質または配列番号:730によってコードされるタンパク質であるか、または前記二本鎖RNAは、配列番号:989、988、1104または1105からなる群から選択される配列を含む、請求項1に記載の方法。
- 前記溶液は、オルガノシリコーン界面活性剤または陽イオン性脂質からなる群から選択される1つ以上の成分を更に含む、請求項6に記載の方法。

- 請求項1に記載の方法によって提供される、Leptinotarsa種の侵入に対する改善された耐性を有する植物、または果実、種子、若しくは前記植物の伝播可能部分。
- 前記植物は、ジャガイモ、トマト及びナスからなる群から選択される、請求項10に記載の植物。
- Leptinotarsa種を防除するための殺虫性組成物であって、前記殺虫性組成物が、
(a)配列番号:730またはそれから転写されたRNAに相補的である少なくとも21の連続するヌクレオチドを含む殺虫性有効量のポリヌクレオチド、または、
(b)配列番号:730またはそれから転写されたRNAの少なくとも21の連続するヌクレオチドに相補的である少なくとも1つのサイレンシング要素を含む殺虫性有効量の少なくとも1つのポリヌクレオチド、または、
(c)配列番号:730またはそれから転写されたRNAの少なくとも21の連続するヌクレオチドに相補的である少なくとも1つのセグメントを含む、殺虫性有効量の少なくとも1つのRNA、または
(d)前記Leptinotarsa種によって摂餌または接触されると、Leptinotarsa種において死滅または成長の阻害を引き起こすRNA分子であって、ここに前記RNA分子が配列番号:730またはそれから転写されたRNAに相補的である少なくとも21の連続するヌクレオチドを含む、前記RNA分子、または
(e)前記Leptinotarsa種によって摂餌または接触されると、Leptinotarsa種において死滅または成長の阻害を引き起こす殺虫性二本鎖RNA分子であって、ここに前記殺虫性二本鎖RNA分子の少なくとも1本の鎖は、配列番号:730に相補的な21の連続するヌクレオチドを含み、または
(f)配列番号:989、988、1104、及び1105からなる群から選択される配列を含む殺虫性有効量の少なくとも1つの二本鎖RNAを含む、前記殺虫性組成物。 - 前記殺虫性組成物は、固体、液体、粉体、懸濁液、乳化剤、噴霧剤、封入形態、マイクロビーズ、キャリア微粒子、フィルム、基質、または種子処理剤、土壌潅水、植付可能配合物、及びインファロー配合物からなる群から選択される少なくとも1つの形態である、請求項12に記載の殺虫性組成物。
- ポリヌクレオチド含有組成物は、キャリア剤、界面活性剤、陽イオン性脂質、オルガノシリコーン、オルガノシリコーン界面活性剤、ポリヌクレオチド除草剤分子、非ポリヌクレオチド除草剤分子、非ポリヌクレオチド殺虫剤、毒性緩和剤及び昆虫成長調整剤からなる群から選択される少なくとも1つの成分を更に含む、請求項12に記載の殺虫性組成物。
- 前記殺虫性組成物は、前記Leptinotarsa種によって摂取されるか、またはLeptinotarsa種が接触するときにLeptinotarsa種の死滅または成長の阻害を招く殺虫性二本鎖RNA分子を含み、前記殺虫性二本鎖RNA分子は、配列番号:730の配列を有するDNAまたは前記DNAから転写されたRNAの21の連続するヌクレオチドに相補的である少なくとも1つのセグメントを含み、前記二本鎖RNA分子は、長さが少なくとも50塩基対であるか、または長さ約100〜約500塩基対である、請求項12に記載の殺虫性組成物。
- 異種プロモーターを含む組換え型DNA構造体であって、前記異種プロモーターは、
(a)配列番号:730またはそれから転写されたRNAの少なくとも21の連続するヌクレオチドに相補的である少なくとも1つのサイレンシング要素をコードしたDNA、または、
(b)配列番号:730またはそれから転写されたRNAに相補的である少なくとも21の連続するヌクレオチドを含む少なくとも1つのサイレンシング要素をコードするDNA、または、
(c)配列番号:989、988、1104及び1105またはこれらの相補体からなる群から選択されるヌクレオチド配列を含むRNAをコードするDNA、に作動可能に連結される、組換え型DNA構造体。 - 請求項16に記載の組換え型DNA構造体を含む植物染色体若しくは色素体又は組換え型植物ウイルスベクター若しくは組換え型バキュロウイルスベクター。
- ゲノム内に請求項16に記載の前記組換え型DNA構造体を有するトランスジェニックナス科植物細胞。
- 前記トランスジェニックナス科植物細胞は、コードするそのゲノム内にパタチン、植物レクチン、植物エクジステロイド、植物エクジステロイド、Bacillus thuringiensis殺虫タンパク質、Xenorhabdus殺虫タンパク質、Photorhabdus殺虫タンパク質、Bacillus laterosporous殺虫タンパク質、及びBacillus sphaericus殺虫タンパク質からなる群から選択される少なくとも1つの殺虫剤をコードするDNAを更に有する、請求項18に記載のトランスジェニックナス科植物細胞。
- 請求項18に記載のトランスジェニックナス科植物細胞を含むトランスジェニックナス科植物、果実、種子または前記トランスジェニックナス科植物の伝播部分。
