Abstract

Se divulgan moléculas de ARN satélite de Martellivirales sintéticas y partículas satélite que las contienen. También se divulgan moléculas de ARN satélite de Martellivirales sintéticas que contienen amplicones de virus de ARN heterólogos (HRV) internos. También se divulgan métodos para utilizar las moléculas de ARN satélite de Martellivirales y las partículas satélite que las contienen para cambiar fenotipos vegetales, mejorar la resistencia al estrés de las plantas y mejorar la resistencia a plagas y patógenos de las plantas. Reivindicación 1: Una molécula de ARN recombinante que comprende, del extremo 5’ al extremo 3’: (a) un elemento de replicación del ARN en 5’ reconocido por una ARN polimerasa dependiente de ARN (RdRP) de Martellivirales; (b) una molécula de ARN de carga; y (c) un elemento de replicación del ARN en 3’ reconocido por la RdRP; en donde el elemento de replicación del ARN en 5’, la molécula de ARN de carga y el elemento de replicación del ARN en 3’ están unidos operativamente, en donde la molécula de ARN de carga es heteróloga al elemento de replicación del ARN en 5’ y al elemento de replicación del ARN en 3’; y opcionalmente en donde: (i) el elemento de replicación del ARN en 5’ y el elemento de replicación del ARN en 3’ se obtienen del mismo genoma de Martellivirales o de genomas de Martellivirales que tienen al menos el 85%, 90%, 95%, 98% o 99% de identidad de secuencia entre sí; (ii) el elemento de replicación del ARN en 5’, el elemento de replicación del ARN en 3’ y la RdRP se obtienen del mismo genoma de Martellivirales o de genomas de Martellivirales que tienen al menos el 85%, 90%, 95%, 98% o 99% de identidad de secuencia entre sí; o (iii) el elemento de replicación del ARN en 5’, el elemento de replicación del ARN en 3’ y/o la región codificante de la RdRP se obtienen de dos genomas de Martellivirales diferentes, y en donde los miembros de cada conjunto respectivo de elementos de replicación del ARN en 5’, elementos de replicación del ARN en 3’ y/o las regiones codificantes de la RdRP tienen al menos el 85%, 90%, 95%, 98% o 99% de identidad de secuencia entre sí. Reivindicación 29: Una formulación agrícola que comprende la molécula de ARN recombinante de una cualquiera de las reivindicaciones 1 a 28. Reivindicación 34: Una célula que comprende la molécula de ADN recombinante de una cualquiera de las reivindicaciones 1 a 28, en donde la célula es una célula bacteriana, una célula fúngica, una célula vegetal, una célula de insecto o una célula de un animal invertebrado. Reivindicación 37: Un sistema de expresión que comprende: (a) una molécula de ARN que comprende la molécula de ARN recombinante de una cualquiera de las reivindicaciones 1 a 28; y (b) una célula que contiene la molécula de ARN recombinante y una proteína RdRP que reconoce los elementos de replicación del ARN en 5’ y en 3’ de la molécula de ARN recombinante. Reivindicación 49: Un método para proporcionar un ARN satélite de Martellivirales sintético a una planta, que comprende poner en contacto la planta con la molécula de ARN recombinante de una cualquiera de las reivindicaciones 1 a 28. Reivindicación 52: Un método para establecer un ARN satélite de Martellivirales sintético en una célula vegetal, que comprende: proporcionar a una planta la molécula de ARN recombinante de una cualquiera de las reivindicaciones 1 a 28; en donde la célula vegetal comprende una proteína RdRP que reconoce el elemento de replicación del ARN en 5’ y el elemento de replicación del ARN en 3’, en donde la molécula de ARN comprende opcionalmente un ERE y está encapsulada por una proteína de la cápside, por lo que la proteína RdRP cataliza la síntesis del ARN satélite de Martellivirales sintético a partir de la molécula de ARN recombinante. Reivindicación 57: Un método de obtención de un cambio fenotípico en una planta o célula vegetal que comprende proporcionar a una planta o célula vegetal una molécula de ARN recombinante de una cualquiera de las reivindicaciones 1 a 28, en donde la molécula de ARN de carga comprende ARN que provoca un cambio fenotípico en la planta o célula vegetal en comparación con una planta o célula vegetal que carece del ARN recombinante, en donde la planta o célula vegetal comprende una proteína RdRP que reconoce el elemento de replicación del ARN en 5’ y el elemento de replicación del ARN en 3’ y cataliza la síntesis de un ARN de Martellivirales sintético a partir de la molécula de ARN recombinante, y en donde la molécula de ARN de carga efectúa el cambio fenotípico. Reivindicación 72: Un método de fabricación de una partícula satélite de Martellivirales sintética, que comprende combinar la molécula de ARN recombinante de una cualquiera de las reivindicaciones 1 a 28 con una proteína de la cápside vírica, en donde la molécula de ARN recombinante comprende un elemento de reconocimiento de la encapsidación (ERE), y en donde el ERE proporciona la encapsidación del ARN por la proteína de la cápside vírica. Reivindicación 81: Una planta que comprende la molécula de ARN recombinante de una cualquiera de las reivindicaciones 1 a 28 y una RdRP de Martellivirales, opcionalmente en donde el propágulo vegetal comprende además una RdRP del virus de ARN heterólogo que reconoce una región de replicación en 5’ o en 3’ del HRV y/o el promotor subgenómico en el ARN satélite de Martellivirales sintético. Reivindicación 92: Un sistema de satélites de Martellivirales que se autorreplica cuando se introduce en una planta o célula vegetal, que comprende: (a) un ARN satélite de Martellivirales recombinante de una cualquiera de las reivindicaciones 1 a 28;y (b) un virus exógeno del orden Martellivirales que es capaz de replicarse en la planta o las células vegetales y que codifica la RdRP de Martellivirales que reconoce las secuencias de reconocimiento de la replicasa en 5’ y en 3’ en el ARN satélite de Martellivirales recombinante, opcionalmente en donde el sistema de satélites de Martellivirales comprende además una RdRP de un virus de ARN heterólogo que reconoce una región de replicación en 5’ o en 3’ del HRV y/o el promotor subgenómico en el ARN satélite de Martellivirales sintético.
