Abstract

 重原子、セレン、反応性官能基、蛍光基、クロスリンカー等、有用な官能基を有する非天然アミノ酸として好適なリジン誘導体、特にNε-ベンジルオキシカルボニル-リジン(Z-Lys)誘導体を所望のタンパク質に部位特異的に導入する方法を提供する。  配列番号2に示したピロリジル-tRNA合成酵素のアミノ酸配列においてピロリジン結合部位を構成する306位のチロシン、309位のロイシン、及び348位のシステインから選択される少なくとも1つのアミノ酸残基が置換された変異体酵素であって、当該アミノ酸置換が、  306位のチロシンからグリシン又はアラニンへ、  309位のロイシンからグリシン又はアラニンへ、及び  348位のシステインからバリン、セリン又はアラニンへ、  の置換である変異体ピロリジル-tRNA合成酵素。
