Abstract

　リボソームにおける生体内翻訳系を用いて、ポリペプチド主鎖に少なくとも１つのエステル結合を含む非天然タンパク質を合成する。（ａ）α－ヒドロキシ酸を活性化しうるアミノアシルｔＲＮＡ合成酵素と、（ｂ）前記アミノアシルｔＲＮＡ合成酵素の存在下において前記α－ヒドロキシ酸と結合可能なサプレッサーｔＲＮＡと、（ｃ）所望の位置にナンセンス変異又はフレームシフト変異を受けた所望のタンパク質をコードする遺伝子と、を前記α－ヒドロキシ酸の存在下に細胞内又は細胞抽出液内で発現させる。
