Abstract
リボソームにおける生体内翻訳系を用いて、ポリペプチド主鎖に少なくとも1つのエステル結合を含む非天然タンパク質を合成する。(a)α-ヒドロキシ酸を活性化しうるアミノアシルtRNA合成酵素と、(b)前記アミノアシルtRNA合成酵素の存在下において前記α-ヒドロキシ酸と結合可能なサプレッサーtRNAと、(c)所望の位置にナンセンス変異又はフレームシフト変異を受けた所望のタンパク質をコードする遺伝子と、を前記α-ヒドロキシ酸の存在下に細胞内又は細胞抽出液内で発現させる。
