Abstract

본 발명은 순도 및 수율이 개선된 데시타빈의 제조방법에 관한 것으로, 더욱 구체적으로는 1,3,5-트리아세틸-2-디옥시라이보스와 실릴기가 도입된 5-아자시토신을 티엠에스오트리플레이트 루이스 산의 존재하에 메틸렌클로라이드 또는 EDC 용매에서 반응시키는 단계(단계 1). 상기 단계 1에서 얻은 반응 생성물을 재결정화 용매로서 에틸아세테이트를 사용하여 결정화된 중간 생성물을 얻는 단계(단계 2); 및 상기 단계 2에서 얻은 결정화된 중간 생성물로부터 아세틸기가 탈보호된 최종 생성물인 데시타빈을 얻는 단계(단계 3)를 포함하여 이루어지는 수율 및 순도가 개선된 데시타빈의 제조방법에 관한 것이다. 본 발명에 의하면, 종래에 언급되지 않은 출발물질과 그의 합성방법을 사용하여 이성질체의 형성을 최소화시킬 수 있고, 또한 전구체를 보호된 아민의 형태가 아닌, 자유 아민(free amine) 형태로 결정화된 새로운 전구체를 경유하여 최종 제품 순도를 99.5% 이상으로 높일 수 있으므로 고순도, 고수율의 데시타빈을 제조하는데 유용하게 사용될 수 있다.
