Abstract
Description

前記基盤上に固着された洩糸性高分子材料からなる突起状の基剤部と、
前記突起状の基剤部の先端に配設された目的物質部を有することを特徴とする。
メス型に目的物質などを注入する工程と、
前記メス型にさらに基剤を注入する工程と、
多孔性を有する基盤を前記メス型に押し付け、乾燥・硬化する工程と、
前記基盤から前記メス型を剥がす工程を有する。
単発打錠器の臼に錠剤用酢酸セルロースを入れ、約10kNの打錠圧で直径1.5cmの錠剤基盤を作った。メス型は、1平方センチメートルのブロック毎に深さ約500ミクロン、開口部直径約300ミクロンの百~数百個の逆円錐状細孔を有する樹脂製のものを用意した。また、モデル抗原である卵白アルブミンと、ヒアルロン酸と、エバンスブルーに脱気した精製水を加えて粘調性濃厚液を作製した。そして、この粘調性濃厚液をメス型の穴の上に塗布した。
単発打錠器に結晶セルロースを入れ、約10kNの打錠圧で直径1.5cmの錠剤基盤を作った。実施例1と同様にしてメス型の穴にモデル抗原である卵白アルブミン、コンドロイチン硫酸Cナトリウムおよびエバンスブルーを精製水に溶かして調製した粘調性濃厚液を塗布した。メス型上の残留物を除去した後、卓上遠心分離器を用いてメス型ごと回転させ、遠心力を利用して加重下、充填した。
単発打錠器にキチンを入れ、約12kNの打錠圧で直径1.5cmの錠剤基盤を作った。実施例2と同様にしてメス型の穴にモデル抗原である卵白アルブミンを含有する粘調性濃厚液を塗布した。メス型上の残留物を除去した後、卓上遠心分離器を用いてメス型ごと回転させ、遠心力を利用して加重下、充填した。
ポリエチレン製の多孔性の板を裁断することにより縦1.5cm、横1.5cmの正方形のフィルターを作成した。インスリンナトリウム(自家製)、エバンスブルーおよびコンドロイチン硫酸ナトリウムに脱気した精製水を加えてよく溶解及び混和し、粘調性濃厚液とした。
ポリメチルメタアクリレート製の多孔性の板を裁断することにより縦1.5cm、横1.5cmの正方形のフィルターを作成した。ヒト成長ホルモン、リサミングリーンおよびコンドロイチン硫酸ナトリウムに脱気した精製水を加えてよく溶解及び混和し、粘調性のある濃厚液を作成した。実施例1と同様にしてメス型の穴に成長ホルモンを含有する粘調性濃厚液を塗布した。メス型上の残留物をスキジーにて除去した後、卓上遠心分離器を用いてメス型ごと回転させ、遠心力を利用して加重下、充填した。
ポリ塩化ビニル製の多孔性の板を裁断することにより縦1.5cm、横1.5cmの正方形のフィルターを作成した。デスモプレシンをpH6.5のリン酸緩衝液に溶解した。さらに、エバンスブルー、コンドロイチン硫酸ナトリウムを加えて溶かして粘調性の濃厚液を作成した。実施例1と同様にしてメス型の穴にデスモプレシンを含有する粘調性濃厚液を塗布した。メス型上の残留物をスキジーにて除去した後、卓上遠心分離器を用いてメス型ごと回転させ、遠心力を利用して加重下、充填した。
塩素化ポリエチレン・スチレン樹脂製の多孔性の板を裁断することにより縦1.5cm、横1.5cmの正方形のフィルターを作成した。エバンスブルー、高分子デキストランにエリスロポエチン注射液(商品名エスポー、24,000IU/mL、協和キリン社)および脱気した精製水を加えて溶かして粘調性の濃厚液を作成した。実施例1と同様にしてメス型の穴にエリスロポエチンを含有する粘調性濃厚液を塗布した。メス型上の残留物をスキジーにて除去した後、卓上遠心分離器を用いてメス型ごと回転させ、遠心力を利用して加重下、充填した。
実施例1と同様に、1平方センチメートルのブロック毎に深さ約500ミクロン、開口部直径約300ミクロンの百~数百個の逆円錐状細孔を有するメス型の穴の上に、モデル抗原である卵白アルブミン、ヒアルロン酸、エバンスブルーに脱気した精製水を加えて作成した粘調性濃厚液を塗布した。メス型上の残留物を除去した後、卓上遠心分離器を用いてメス型ごと回転させ、遠心力を利用して加重下、充填した。
比較例1と同様に実験を行った。ただし、アクリル板の代わりに厚さ0.75mmのポリプロピレン製のシートを用いて、メス型からマイクロニードル・アレイを取り出そうとしたが、比較例1と同様にメス型よりマイクロニードル・アレイを取り出すことができなかった。
単発打錠器の臼にエチルセルロースあるいは酢酸セルロースを入れ、約15kNの打錠圧で直径1.5cmの錠剤基盤を作った。実施例2と同様にメス型の穴の上に、モデル抗原である卵白アルブミン、コンドロイチン硫酸ナトリウム、エバンスブルーを脱気精製水に溶かして調製した粘調性濃厚液を塗布した。
そこで基盤の側面にもコンドロイチン硫酸ナトリウム糊を塗った。酢酸セルロース製あるいはエチルセルロース製の錠剤基盤の側面に糊を塗ったサンプルは、過度に乾燥が進んでも錠剤基盤の周辺部がめくれ返ることがなかった。
1平方センチメートルのブロック毎に深さ約500ミクロン、開口部直径約300ミクロンの百~数百個の逆円錐状細孔を有する樹脂製基盤の穴の上に、モデル抗原である卵白アルブミン、コンドロイチン硫酸ナトリウムおよびエバンスブルーに脱気精製水を加えて作成した粘調性濃厚液を塗布した。
