Abstract
Description
以下に、本発明の実施の形態について説明する。ただし、本発明が以下の実施の形態に限定される訳ではない。また、説明を明確にするため、以下の記載及び図面は、適宜、簡略化されている。
複数のマイクロ空間を有する細胞培養容器で保存した場合と平板で保存した場合の実施例
<実施例01>
図3、4に示す凹凸パターン形状であって、a=200μm、b=20μm、c=50μm、のパターンをフォトリソグラフィにより作製し、Ni電解メッキを行い、対応する凹凸形状を有する金型を得た。その金型を用い、ホットエンボス成形によりポリスチレン上に凹凸パターン形状の転写を行い、前記寸法の樹脂基材を作製した。その樹脂基材表面へ真空蒸着により二酸化ケイ素膜を100nm形成させたフィルムを作製し、ポリスチレン製の培養底面のない24穴プレートに、レーザ溶着法でフィルムを貼り付けた後、γ線滅菌を行い、24ウェルの複数のマイクロ空間を有する培養容器を作製し保存試験に用いた。
<比較例01>
市販(ベクトン・ディッキンソン製、ファルコン(登録商標))のγ線滅菌済み平面状24ウェル培養プレートを保存試験に用いた。
ヒト肝癌由来細胞株HepG2(財団法人ヒューマンサイエンス研究資料バンク 資源番号JCRB1054)を、培養フラスコ(CORNIGN社製)を用い、37℃、5%CO2インキュベータ内で、所定の細胞数まで増殖させた。培地は、10%ウシ胎児血清を含むDMEM(GIBCO社製)培地を用いた。増殖させた細胞を、0.25%トリプシン溶液を用いて培養底面から剥離し、遠心分離法にて細胞を回収した。回収した細胞を、10%FBSを含むDMEM培養液で、細胞濃度が4×105個/mLになるように調整し、各ウェルに500μLずつ添加した。その後、細胞を37℃、5%CO2インキュベータ内で3日間培養した。
実施例01、比較例01に示した培養容器を用いて、37℃、5%CO2インキュベータ内で3日間培養した後、培地を吸引し、各ウェルに新たに500μLの10%FBSを含むDMEM培地を入れ、培養プレートをプラスチック製のフィルムで密閉した後、37℃のインキュベータに入れ、24時間保存した。
24時間保存した後、培地を吸引し、各ウェルに10%FBSを含むDMEM培地を入れ37℃、5%CO2インキュベータ内で2日間培養した。培養後の上澄みを回収し、ヒトアルブミン分析ELISAキット(Bethyl Laboratories社製)を用いて、2日間のヒトアルブミン量を測定した。
表1にアルブミン分泌量を測定した結果を示した。実施例01は、比較例01の3倍の分泌量となった。
保存温度についての実施例
図3、4に示す凹凸パターン形状であって、a=200μm、b=20μm、c=50μm、のパターンをフォトリソグラフィにより作製し、Ni電解メッキを行い、対応する凹凸形状を有する金型を得た。その金型を用い、ホットエンボス成形によりポリスチレン上に凹凸パターン形状の転写を行い、前記寸法の樹脂基材を作製した。その樹脂基材表面へ真空蒸着により二酸化ケイ素膜を100nm形成させたフィルムを作製し、ポリスチレン製の培養底面のない24穴プレートに、レーザ溶着法でフィルムを貼り付けた後、γ線滅菌を行い、24ウェルの複数のマイクロ空間を有する培養容器(培養プレート)を作製し保存試験に用いた。
ヒト肝癌由来細胞株HepG2(財団法人ヒューマンサイエンス研究資料バンク 資源番号JCRB1054)を、培養フラスコ(CORNIGN社製)を用い、37℃、5%CO2インキュベータ内で、所定の細胞数まで増殖させた。培地は、10%ウシ胎児血清を含むDMEM(GIBCO社製)培地を用いた。増殖させた細胞を、0.25%トリプシン溶液を用いて培養底面から剥離し、遠心分離法にて細胞を回収した。回収した細胞を、10%FBSを含むDMEM培養液で、細胞濃度が4×105個/mLになるように調整し、培養容器の各ウェルに500μLずつ添加した。その後、細胞を、37℃、5%CO2インキュベータ内で3日間培養した。
3日間培養した後、培地を吸引し、各ウェルに500μLの10%FBSを含むDMEM培地を入れた。培養容器をプラスチック製のフィルムで密閉した後、以下の実施例の保存方法で保存した。保存中の庫内温度は温度センサーを用いて測定し、目的温度を保っていることを確認した。
<実施例02>
18℃の定温保温剤が入った保存容器(日立物流社製)を用い、24時間保存した。このときの庫内の温度は、18℃±0.5℃であった。
<実施例03>
6℃の定温保温剤が入った保存容器(日立物流社製)を用い、24時間保存した。このときの庫内の温度は、6℃±0.5℃であった。
<実施例04>
発泡スチロール製の容器の中に氷を入れ、その上に培養容器を置き24時間保存した。このときの培養容器底面の温度は0℃±0.5℃であった。
<実施例05>
37℃のインキュベータに入れ、24時間保存した。
24時間保存後、培地を吸引し、各ウェルに10%FBSを含むDMEM培地を入れ、37℃、5%CO2インキュベータ内で24時間培養した。その後、倒立顕微鏡を用いて観察を行い、任意の視野について、30~200μmの細胞塊が形成された数をカウントし、以下の式で細胞塊形成率を算出した。また、保存前の細胞塊形成率についても同様の方法で算出した。
ただし、(式1)において、
「1視野内マイクロ空間の個数」は、1視野(任意の範囲の視野)に存在するマイクロ空間(マイクロ容器)の個数をいう。
「細胞塊が形成された1視野内マイクロ空間個数」は、1視野内マイクロ空間のうち、30~200μmの細胞塊が形成されているマイクロ空間の個数をいう。
表2に細胞塊形成率を示した。保存前の細胞塊形成率は、80~100%であった。実施例02(18℃)では、保存前の形態がほぼ完全に保たれていた。実施例03(6℃)は、細胞塊形成率が低下するものの、60%~80%の形態が維持されていた。実施例04(0℃)は30%と形成率が低くほとんど形態が維持されなかった。実施例05(37℃)は25%と形成率が一番低く、ほとんど形態が維持されなかった。
この結果から、保存温度は、10℃以上20℃以下が好ましく、18℃が特に好ましいことがわかった。
細胞密度についての実施例
実施例06、07は、実施例02-05と同じ培養容器を使用した。
ヒト肝癌由来細胞株HepG2(財団法人ヒューマンサイエンス研究資料バンク 資源番号JCRB1054)を、培養フラスコ(CORNIGN社製)を用い、37℃、5%CO2インキュベータ内で、所定の細胞数まで増殖させた。培地は、10%ウシ胎児血清を含むDMEM(GIBCO社製)培地を用いた。増殖させた細胞を、0.25%トリプシン溶液を用いて培養底面から剥離し、遠心分離法にて細胞を回収した。回収した細胞を、10%FBSを含むDMEM培養液で、実施例に示した細胞濃度に調整し、培養容器の各ウェルに500μLずつ添加した。その後、37℃、5%CO2インキュベータ内で3日間培養した。
細胞を播種する際、細胞濃度を4×105個/mLに調整し、以下に示す保存条件で保存した。
<実施例07>
細胞を播種する際、細胞濃度を40×105個/mLに調整し、以下に示す保存条件で保存した。
3日間培養した後、培地を吸引し、各ウェルに500μLの10%FBSを含むDMEM培地を入れ、培養プレートをプラスチック製のフィルムで密閉した後、18℃の定温保温剤が入った容器(日立物流社製)に入れ、24時間保存した。
(アルブミン分析)
24時間保存した後、培地を吸引し、各ウェルに10%FBSを含むDMEM培地を入れ37℃、5%CO2インキュベータ内で2日間培養した。培養後の上澄みを回収し、ヒトアルブミン分析ELISAキット(Bethyl Laboratories社製)を用いて、2日間のヒトアルブミン分泌量(ng/mL/2days)を測定した。次に、0.25%のトリプシン溶液で細胞を剥離し、トリパンブルーで生細胞の値を算出、105個あたりの値を、以下に記載の表3のアルブミン分泌量とした(pg/105/2days)。
(形態観察)
24時間保存した後、培地を吸引し、各ウェルに10%FBSを含むDMEM培地を入れ37℃、5%CO2インキュベータ内で、24時間、37℃、5%CO2インキュベータ内で培養した後、倒立顕微鏡を用いて形態を観察した。
図8A、8Bは、保存容器から取り出した後、24時間、37℃、5%CO2インキュベータ内で培養した時の写真である。
実施例06(図8A)では球状の細胞塊が形成されているのに対し、実施例07(図8B)ではマイクロ空間内に細胞が密に詰まった状態で塊となり、死んだ細胞が区画外で凝集していた。表3にアルブミン分泌量を示した。アルブミン分泌量については、実施例06は実施例07の約2.8倍という高い値を示した。

Claims (15)
- 複数のマイクロ空間を有する細胞培養容器を用いて生細胞を保存する細胞保存方法であって、
生細胞を前記複数のマイクロ空間の表面に接着させて培養し、
前記培養後、前記複数のマイクロ空間を覆うように、培地を前記細胞培養容器へ注入し、
前記細胞培養容器を、培地が漏れ出さないように密封して前記生細胞を保存する細胞保存方法。 - 前記細胞培養容器は、4℃以上37℃未満の温度に保たれて前記生細胞が保存されることを特徴とする請求項1記載の細胞保存方法。
- 前記培養は、前記表面に接着され、かつ、各マイクロ空間の空間内に形成される三次元構造体の細胞集団が隔離される細胞数になるように培養することを特徴とする請求項1または2記載の細胞保存方法。
- 前記複数のマイクロ空間は、底部面積が0.01~0.1mm2であり、深さが25~150μmであることを特徴とする請求項1乃至3のいずれか一項に記載の細胞保存方法。
- 前記生細胞は、肝細胞、膵ベータ細胞、心筋細胞、神経細胞、皮膚上皮細胞、軟骨細胞、骨細胞、組織幹細胞、ES細胞、及びiPS細胞のうちのいずれかであることを特徴とする請求項1乃至4のいずれか一項に記載の細胞保存方法。
- 前記生細胞は、組織幹細胞、ES細胞、及びiPS細胞のうちのいずれかから分化させた、肝細胞、膵ベータ細胞、心筋細胞、神経細胞、皮膚上皮細胞、軟骨細胞、及び骨細胞のうちのいずれかであることを特徴とする請求項1乃至4のいずれか一項に記載の細胞保存方法。
- 前記培養後、非接着細胞を除去してから前記培地を前記細胞培養容器へ注入することを特徴とする請求項1乃至6のいずれか一項に記載の細胞保存方法。
- 前記生細胞の培養は、生細胞を接着させて培養した後、さらに培養して、増殖、伸展、または凝集させることを特徴とする請求項1乃至7のいずれか一項に記載の細胞保存方法。
- 細胞播種密度1×102~1×106細胞/cm2で、生細胞が前記複数のマイクロ空間に播種されたことを請求項1乃至8のいずれか一項に記載の細胞保存方法。
- 前記複数のマイクロ空間に、生細胞が集積した細胞塊が形成されていることを特徴とする請求項1乃至9のいずれか一項に記載の細胞保存方法。
- 前記細胞塊の直径は、30~200μmであることを特徴とする請求項1乃至10のいずれか一項に記載の細胞保存方法。
- 前記培地は、血清、増殖因子、及び血中成分の少なくとも一つを含むことを特徴とする請求項1乃至11のいずれか一項に記載の細胞保存方法。
- 酸素または二酸化炭素を透過させる膜を用いて前記細胞培養容器を密封することを特徴とする請求項1乃至12のいずれか一項に記載の細胞保存方法。
- 複数のマイクロ空間を有する細胞培養容器へ生細胞を保存して輸送する細胞輸送方法であって、
生細胞を前記複数のマイクロ空間の表面に接着させて培養し、
前記培養後、前記複数のマイクロ空間を覆うように、培地を前記細胞培養容器へ注入し、
前記細胞培養容器を、培地が漏れ出さないように密封し、
密封した前記細胞培養容器を、車両、船舶、または航空機のいずれかの輸送手段を用いて輸送する細胞輸送方法。 - 前記細胞培養容器は、4℃以上37℃未満の温度に保たれることを特徴とする請求項14記載の細胞輸送方法。
