Abstract
L'invention vise à proposer une nouvelle enzyme et son gène qui peuvent être utilisés de façon efficace dans la production à grande échelle de fusicocca-2,10(14)-diène qui est utile comme matière pour produire la cotylénine, les 12-désoxyfusicoccines et leurs dérivés ayant un grand potentiel comme agent anti-cancer. On vise également à proposer un procédé de fabrication de fusicocca-2,10(14)-diène à l'aide de l'enzyme décrite ci-dessus ou d'un vecteur à utiliser dans la production de l'enzyme et d'un transformant. Une fusicoccane synthase chimérique qui est caractérisée comme ayant un domaine de terpène cyclase comprenant la séquence d'acides aminés représentée SEQ ID NO:1 ou une séquence fonctionnelle équivalente à celle-ci dans la région N-terminale et un domaine de prényl transférase comprenant la séquence d'acides aminés représentée par SEQ ID NO:2 ou une séquence équivalent fonctionnel à celle-ci dans la région C-terminale. Par la culture d'un transformant qui porte un gène codant pour l'enzyme décrite ci-dessus, l'enzyme et le fusicocca-2,10(14)-diène sont obtenus.
