Abstract
Die Erfindung betrifft transgene nicht-menschliche Säugetiere, die zwei Transgene in ihrem Genom tragen, ein Verfahren zu deren Herstellung, deren Verwendung und Zellgewebe, Zellen sowie Zellbestandteile daraus. Anwendungsgebiete der Erfindung sind die pharmazeutische Industrie und die Lebenswissenschaften: die Biologie, die Biochemie, die Biotechnologie, die Medizin, bevorzugt die Prophylaxe und die Therapie von Krebs, insbesondere Lungenkrebs. Der Erfindung liegt das Ziel zugrunde, ein verbessertes Tiermodell zu schaffen, das unterschiedliche Arten von Tumoren bildet, die mit menschlichen Tumoren, insbesondere Lungentumoren, und deren Metastasen vergleichbar sind. Aufgabe der Erfindung ist es daher, ein nicht menschliches Säugetier mit solchen verbesserten Eigenschaften und/oder dessen Zellgewebe, Zellen und Zellbestandteile, ein Verfahren zu deren Herstellung und deren Verwendung, sowie Mittel zu ihrer Herstellung und deren Verwendung, bereitzustellen. Diese Aufgabe wird mit einem doppelt transgenen nicht menschlichen Säugetier gelöst, dessen Genom a) eine kodierende DNA-Sequenz (1 ), die für ein Protoonkogen kodiert und die operativ mit wenigstens einer regulatorischen Sequenz verbunden ist, und b) eine kodierende DNA-Sequenz (2), die für ein Onkogen kodiert und die operativ mit wenigstens einer regulatorischen Sequenz verbunden ist, umfasst. Die Erfindung betrifft ferner entsprechend doppelt transgene Tumorzellklone für die Verwendung zur in vitro Untersuchung der Pathogenese von Lungentumoren und von gegen Lungenkarzinome gerichteten Substanzen bzw. Therapieverfahren.
