Abstract

Die Erfindung betrifft Verfahren zur Analyse von Makromolekülen mit Hilfe eines Microarrays, das aus mehreren Spots in bekannter Anordnung besteht, die eine Vielzahl von ersten zumindest teilweise unterschiedlichen Makromolekülen umfassen, wobei sich das Microarray auf einer Festkörperoberfläche, vorzugsweise eines Chips, befindet, bei dem zumindest ein Flüssigkeitstropfen mit Makromolekülen auf die Festkörperoberfläche gebracht wird, wobei die Flüssigkeitsmenge so klein ist, dass im Wesentlichen nur ein oder eine Gruppe von wenigen Spots des Microarrays mit dem Flüssigkeitstropfen in Berührung kommt, und nach Ablauf einer vorbestimmten Zeit der Flüssigkeitstropfen mit Hilfe des Impulsübertrages einer Oberflächenschallwelle zu einem nächsten Spot oder einer weiteren Gruppe weniger Spots bewegt wird. Die mit dem Flüssigkeitstropfen in Berührung gekommenen Spots werden daraufhin untersucht, ob eine Reaktion mit dem Material des Flüssigkeitstropfens stattgefunden hat. Die Erfindung betrifft weiterhin ein Verfahren zur Herstellung einer Analysevorrichtung für Makromoleküle, bei dem zumindest ein Flüssigkeitstropfen mit zumindest einer Art von Makromolekülen auf eine Festkörperoberfläche, vorzugsweise eines Chips, aufgebracht wird, wobei die Makromoleküle eine aktive Gruppe aufweisen, die bei Änderung eines entsprechenden Parameters aktiviert wird, um eine Bindung an die Oberfläche oder bereits gebundenen Molekülen zu ermöglichen. Der Tropfen wird mit Hilfe des Impulsübertrages einer oder mehrerer Oberflächenschallwellen an einen gewünschten Ort auf der Oberfläche bewegt. Der äussere Parameter wird homogen und nicht lokal verändert, so dass eine Bindung der Makromoleküle ermöglicht wird, so dass der Spot eines Microarrays gebildet wird, in dem Makromoleküle aus dem Flüssigkeitstropfen an der Oberfläche gebunden sind. Schliesslich betrifft die Erfindung eine Analysevorrichtung zur Durchführung der erfindungsgemässen Analyseverfahren.
