Abstract
Analizador automatizado (50) para su utilización en la realización de un ensayo basado en ácidos nucleicos, comprendiendo el analizador (50) en el interior de una carcasa (60): una estructura móvil de imán (810) para someter una muestra a un campo magnético, en el que la estructura móvil de imán (810) soporta una pluralidad de imanes (814); un tubo aspirador (860) para aspirar un componente fluido de la muestra mientras la muestra es sometida al campo magnético de la estructura móvil de imán (810); una incubadora de amplificación (604) que comprende una carcasa (610) que comprende una abertura de acceso de recipiente (614) que está dimensionada para desplazar un recipiente de reacción (162) en o fuera de la incubadora de amplificación (604), en el que el recipiente de reacción (162) se cree contiene un ácido nucleico diana separado del componente fluido de la muestra; y un dispositivo de detección (950) para detectar una señal luminosa asociada con un producto de amplificación formado en la incubadora de amplificación (604), indicando el producto de amplificación la presencia de un ácido nucleico diana en la muestra.
Claims (14)
1. Analizador automatizado (50) para su
utilización en la realización de un ensayo basado en ácidos
nucleicos, comprendiendo el analizador (50) en el interior de una
carcasa (60):
una estructura móvil de imán (810) para someter
una muestra a un campo magnético, en el que la estructura móvil de
imán (810) soporta una pluralidad de imanes (814);
un tubo aspirador (860) para aspirar un
componente fluido de la muestra mientras la muestra es sometida al
campo magnético de la estructura móvil de imán (810);
una incubadora de amplificación (604) que
comprende una carcasa (610) que comprende una abertura de acceso de
recipiente (614) que está dimensionada para desplazar un recipiente
de reacción (162) en o fuera de la incubadora de amplificación
(604), en el que el recipiente de reacción (162) se cree contiene un
ácido nucleico diana separado del componente fluido de la muestra;
y
un dispositivo de detección (950) para detectar
una señal luminosa asociada con un producto de amplificación
formado en la incubadora de amplificación (604), indicando el
producto de amplificación la presencia de un ácido nucleico diana
en la muestra.
2. Analizador automatizado (50) según la
reivindicación 1, en el que la estructura móvil de imán (810)
resulta móvil entre las primera y segunda posiciones, en el que los
imanes (814) no presentan un efecto sustancial sobre las partículas
sensibles magnéticamente presentes en la muestra cuando la
estructura móvil de imán (810) está en la primera posición, y en el
que los imanes (814) presentan un efecto sustancial sobre las
partículas sensibles magnéticamente presentes en la muestra cuando
la estructura móvil de imán (810) está en la segunda posición.
3. Analizador automatizado (50) según la
reivindicación 1 ó 2, en el que el dispositivo de detección (950)
comprende un tubo fotomultiplicador (956) para la detección de la
señal luminosa.
4. Analizador automatizado (50) según la
reivindicación 1 ó 2, en el que el dispositivo de detección (950)
comprende un sensor óptico para detectar la señal luminosa.
5. Analizador automatizado (50) según la
reivindicación 4, en el que el sensor óptico es un diodo.
6. Analizador automatizado (50) según cualquiera
de las reivindicaciones 1 a 5, que comprende además una estación de
rampa de temperatura (700) dispuesta entre la estructura móvil de
imán (810) y la incubadora de amplificación (604), en el que la
estación de rampa de temperatura (700) comprende un calentador o
refrigerador para elevar o disminuir la temperatura de un fluido
que se cree contiene el ácido nucleico diana.
7. Analizador automatizado (50) según cualquiera
de las reivindicaciones 1 a 6, que comprende además un mecanismo de
transporte (502) para desplazar el recipiente de reacción (160) en y
fuera de la incubadora de amplificación (604).
8. Analizador automatizado (50) según la
reivindicación 7, en el que el mecanismo de transporte (502)
presenta un gancho de manipulación retráctil y extensible (506)
para acoplar y desplazar el recipiente de reacción (160) en y fuera
de la incubadora de amplificación (604).
9. Analizador automatizado (50) según la
reivindicación 7 u 8, en el que el mecanismo de transporte (502) es
giratorio.
10. Analizador automatizado (50) según
cualquiera de las reivindicaciones 1 a 9, que comprende además los
reactivos almacenados para realizar una reacción de amplificación
basada en ácidos nucleicos.
11. Procedimiento para determinar la presencia
de un ácido nucleico diana en una muestra, comprendiendo el
procedimiento realizar dentro de una carcasa las etapas
automatizadas que consisten en:
a) combinar una muestra con un material de
soporte sólido en condiciones que permitan la inmovilización del
ácido nucleico diana sobre el material de soporte sólido;
b) aislar el material de soporte sólido de los
componentes no inmovilizados de la muestra y aspirar los componentes
no inmovilizados de la muestra;
d) lavar el material de soporte sólido una o más
veces con un tampón de lavado;
e) amplificar una secuencia diana contenida en
el ácido nucleico diana; y

f) detectar un producto de amplificación formado
en la etapa (e) como una indicación de la presencia del ácido
nucleico diana en la muestra.
12. Procedimiento según la reivindicación 11, en
el que el material de soporte sólido es una partícula sensible
magnéticamente, y en el que la etapa (b) comprende someter el
material de soporte sólido a un campo magnético.
13. Procedimiento según la reivindicación 11 ó
12, en el que el ácido nucleico diana está inmovilizado sobre el
material de soporte sólido con una sonda de captura.
14. Procedimiento según cualquiera de las
reivindicaciones 11 a 13, en el que la etapa de detección comprende
hibridar una sonda marcada de manera detectable al producto de
amplificación.
