Abstract
Los transportadores son una familia diana emergente con un potencial enorme, que ofrece oportunidades científicas y economicas. El intercambiador sodio/calcio es un mecanismo importante para eliminar el Ca2+ de diversas células. En el corazon,extruye el Ca2+ que ha ingresado a través de los canales de Ca2+ para iniciar la contraccion mientras que el Na+ ingresa a las células del corazon. Resulta de considerable interés identificar compuestos que modulen la actividad de losintercambiadores sodio/calcio. Se divulga un ensayo basado en fluorescencia para detectar compuestos moduladores del ômodo directoö del NCX. También se refiere a un kit de partes que comprende células que exprimen el NCX y al uso del kit de partespara ensayar la actividad como agonista o antagonista del NCX en un compuesto. Reivindicacion 1: El ensayo para determinar la actividad de la proteína del NCX, comprende: e) proveer células que expresan NCX; f) proveer una sustancia coloreadapara determinar el calcio intracelular; g) poner en contacto las células con un activador de la actividad del NCX; y h) comparar el cambio mediado por calcio en la senal luminiscente de dicha sustancia coloreada con una senal luminiscente producidaen un experimento de control.
