Abstract
Det er beskrevet en fremgangsmåte til å fremstille zolmitriptanforbindelser, særlig zolmitriptan eller et farmasøytisk akseptabelt salt derav som inkluderer a) fremstilling av diazoniumsalt av anilinhydroklorid (II); etterfulgt av reduksjon og surgjøring for å gi hydrazin (III), b) in situ reaksjon av hydrazinhydroklorid (III) med (-keto-(valerolakton for å gi hydrazon (IV); c) Fischer indolsyntese av hydrazon (IV) for å gi pyranoindolon med formel (V); d) transesterifisering av pyranoindolon (V) for å tilveiebringe forbindelsen (VI), hvori R er en rett eller forgrenet C I -C4-alkyl; e) konversjon av hydroksylgruppen til forbindelsen (VI) til dimetylamino for å gi indolkarboksylatet (VII) hvori R betyr en rett eller forgrenet C 1-C4-alkyl; f) saponifisering av 2karboalkoksygruppen til forbindelsen (VII) for åtilveiebringe indolkarboksylsyre (Vlll); g) dekarboksylering av indolkarboksylsyren (VIII) for å tilveiebringe zolmitriptan og eventuelt åtilveiebringe et farmasøytisk akseptabelt salt derav.
