- the medium was replaced with serum-free medium and cultured for 4 hours, and then the peptides of the present invention (5 ⁇ M and 50 ⁇ M) and Th17 type proinflammatory cytokines used as stimulants, TNF- ⁇ (10 nM) and hIL-17A (200 ng /ml), and further incubated for 1 hour.
- RNA extraction kit Qiagen RNeasy kit
- cDNA was synthesized using a cDNA synthesis kit (Intron, Korea) after RNA quantification.
- Polymerase chain reaction (PCR) was performed using PCR premix (Intron, Korea), synthesized cDNA, TNF- ⁇ , COX2, IL-6, IL-8 and GAPDH specific to each gene in [Table 3]. This was done by mixing the indicated primers.
- PCR products were electrophoresed on a 5% agarose gel and stained with ethidium bromide to determine the mRNA expression levels of TNF- ⁇ , COX2, IL-6, and IL-8, which are genes associated with inflammatory and autoimmune diseases. Confirmed.
