本発明は、乾燥葉の単位質量当たりのチラミン含量が0.092%未満であるチラミン低含有ステビア植物体(以下、「本発明の植物体」又は「本発明のステビア植物体」と総称する場合がある)を提供する。
ステビアは南米パラグアイを原産地とする菊科多年生植物で、学名をステビア・レバウディアナ・ベルトニー(Stevia Rebaudiana Bertoni)という。
ここで乾燥葉とは、本発明のステビア植物体の新鮮葉を乾燥させることにより含水量を3~4重量%にまで減らしたものをいう。
一態様において、本発明のステビア植物体は、配列番号1の201位に相当する位置の塩基がAであるアレルについてホモ接合性であるという遺伝的特徴(以下、「本発明の遺伝的特徴A」と称する場合がある)を有する。
別の態様において、本発明のステビア植物体は、以下の(B-1)~(B-4)の少なくとも1つの遺伝的特徴(以下、「本発明の遺伝的特徴B」と称する場合がある)を有する。
(B-1)配列番号2の40位に相当する位置の塩基がTであるアレルについてホモ接合性である(以下、「本発明の遺伝的特徴B-1」と称する場合がある)。
(B-2)配列番号3の44位に相当する位置の塩基がTであるアレルについてホモ接合性である(以下、「本発明の遺伝的特徴B-2」と称する場合がある)。
(B-3)配列番号4の41位に相当する位置の塩基がCであるアレルについてホモ接合性である(以下、「本発明の遺伝的特徴B-3」と称する場合がある)。
(B-4)配列番号5の55~72位に相当する部分が欠失しているアレルについてホモ接合性である(以下、「本発明の遺伝的特徴B-4」と称する場合がある)。
別の態様において、本発明のステビア植物体は、配列番号6の49位に相当する位置の塩基がAであるアレルについてヘテロ接合性であるという遺伝的特徴(以下、「本発明の遺伝的特徴C」と称する場合がある)を有する。
ここでは簡潔のため本発明の遺伝的特徴Aを例に説明したが、本発明の遺伝的特徴X、B(遺伝的特徴B-1~B-4を含む)及びCについても同様である。
特定の態様において、「配列番号1に相当する塩基配列からなる部分」には、例えば、配列番号7の塩基配列を含むフォワードプライマーと、配列番号8の塩基配列を含むリバースプライマーとを用いたPCRにより増幅され得るステビア植物のゲノムの部分が含まれる。
特定の態様において、「配列番号2に相当する塩基配列からなる部分」には、例えば、配列番号9の塩基配列を含むフォワードプライマーと、配列番号10の塩基配列を含むリバースプライマーとを用いたPCRにより増幅され得るステビア植物のゲノムの部分が含まれる。
特定の態様において、「配列番号3に相当する塩基配列からなる部分」には、例えば、配列番号11の塩基配列を含むフォワードプライマーと、配列番号12の塩基配列を含むリバースプライマーとを用いたPCRにより増幅され得るステビア植物のゲノムの部分が含まれる。
特定の態様において、「配列番号4に相当する塩基配列からなる部分」には、例えば、配列番号13の塩基配列を含むフォワードプライマーと、配列番号14の塩基配列を含むリバースプライマーとを用いたPCRにより増幅され得るステビア植物のゲノムの部分が含まれる。
特定の態様において、「配列番号5に相当する塩基配列からなる部分」には、例えば、配列番号15の塩基配列を含むフォワードプライマーと、配列番号16の塩基配列を含むリバースプライマーとを用いたPCRにより増幅され得るステビア植物のゲノムの部分が含まれる。

また、(X)配列番号150の201位に相当する位置におけるTからCへの変異、(A)配列番号1の201位に相当する位置におけるCからAへの変異、(B-1)配列番号2の40位に相当する位置におけるAからTへの変異、(B-2)配列番号3の44位に相当する位置におけるCからTへの変異、(B-3)配列番号4の41位に相当する位置におけるGからCへの変異、(B-4)配列番号5の55~72位に相当する部分の欠失、及び(C)配列番号6の49位に相当する位置におけるCからAへの変異からなる群から選択される変異を、「本発明の多型」又は「本発明の変異」と総称することがある。
候補植物体が遺伝的特徴Xを有する場合、例えば、候補植物体のゲノムDNAに対し、配列番号156に示す塩基配列を有するフォワードプライマー及び配列番号157に示す塩基配列を有するリバースプライマーを用いてPCR増幅を行い、得られたPCR産物(約238bp長、例えば、配列番号158又は159)に対し制限酵素Hpy188Iによる処理を行うと、約51bp長(例えば、配列番号160)、約107bp長(例えば、配列番号161)及び約80bp長のバンド(例えば、配列番号162)が生じる。一方、約51bp長(例えば、配列番号160)及び約187bp長(例えば、配列番号163)の制限酵素処理産物を生じる場合、その候補植物体は遺伝的特徴Xを有しない。
上記bp長に関し、「約」とは、±5bpを意味する。制限酵素処理は、使用する各制限酵素の販売元が推奨する条件に従って行うことができる。
(1)ステビア植物体の新鮮葉をトリクロロ酢酸水溶液(例えば、20%トリクロロ酢酸水溶液)でホモジナイズし、所定時間放置する。
(2)上清にオクタンスルホン酸ナトリウム(例えば、0.1mol/L)を添加、混和し、固相抽出デバイス(例えば、Sep-Pak(R) Vac 6cc 1g C18 Cartridges、Watersを使用)に供する。
(3)固相抽出デバイスを洗浄後、メタノール水溶液(例えば、80%メタノール水溶液)でアミン類を溶出する。
(4)溶出液を必要に応じて濃縮し(例えば10倍濃縮)、内部標準溶液(例えば、1,8-ジアミノオクタンを20μg/mLの濃度で0.5mol/L塩酸に溶解したもの)、無水炭酸ナトリウム、ダンシルクロライド・アセトン溶液を加えてよく混和し、遮光して45℃の水浴中で所定時間(例えば1時間)加温する。
(5)プロリン溶液(例えば、10%プロリン溶液)を加え、振とうし所定時間(例えば10分間)放置する。
(6)トルエンを加えて所定時間(例えば1分間)振とうし、上層のトルエン層を採取する。必要に応じて残液に対し同様の操作を行う。
(7)トルエン層を減圧濃縮・乾固し(例えば40℃以下で)、残留物にアセトニトリルを加えてよく混和する。
(8)得られた溶液をHPLCで分析する。
さらに、このようにして得られた抽出液に対し、酢酸エチルその他の有機溶媒:水の勾配、高速液体クロマトグラフィー(High Performance Liquid Chromatography:HPLC)、ガスクロマトグラフィー、飛行時間型質量分析(Time-of-Flight mass spectrometry:TOF-MS)、超高性能液体クロマトグラフィー(Ultra (High) Performance Liquid chromatography:UPLC)等の公知の方法を用いることによりRebDを精製することができる。
本発明は、別の実施態様において、本発明のステビア植物体と第2のステビア植物体とを交雑させる工程を含む、乾燥葉の単位質量当たりのチラミン含量が0.092%未満であるチラミン低含有ステビア植物体を作出する方法(以下、「本発明の作出方法」と称する場合がある)を提供する。
当該方法により作出される「乾燥葉の単位質量当たりのチラミン含量が0.092%未満であるチラミン低含有ステビア植物体」は、本発明の植物体と同じ表現型と遺伝的性質を有する。
本発明の作出方法によって得られる植物体におけるチラミン及びRebD含量の範囲は、本発明の植物体について上記したとおりである。
一般に、植物細胞は、培養の間に変異を伴うことがあるため、より安定した形質維持のために植物個体に戻すことが好ましい。
本発明の植物体を宿主として事後的に遺伝子組み換え(例えばゲノム編集等により)を行って得られた植物体(例えば、本発明の植物体を宿主として遺伝子組み換えを行って、さらに別の形質を付加した植物体)も、本発明の範囲から除外されるものではない。
本発明の植物体及び本発明の植物体と同じ表現型と遺伝的性質を有する植物体は、当該植物体の組織から本発明の遺伝的特徴を検出することによりスクリーニングすることができる。ここで、「スクリーニング」とは、本発明の植物体とそれ以外の植物体とを識別し、本発明の植物体を選択することを意味する。
したがって、本発明は、別の側面において、被験植物のゲノムから本発明の遺伝的特徴X、A、B及びCの少なくとも1つの存在及び/又は不在を検出する工程を含む、チラミン低含有ステビア植物体をスクリーニングする方法(以下、「本発明のスクリーニング方法」と称する場合がある)を提供する。
本発明のスクリーニング方法は、上記の少なくとも1つの遺伝的特徴の存在が検出された植物体を被験植物の中から選択する工程をさらに含んでもよい。

シークエンス法とは、変異を含む領域をPCRにて増幅させ、Dye Terminatorなどを用いてDNA配列をシークエンスすることで、変異の有無を解析する方法である(Sambrook, Fritsch and Maniatis, ”Molecular Cloning: A Laboratory Manual” 2nd Edition (1989), Cold Spring Harbor Laboratory Press)。
DNAマイクロアレイは、ヌクレオチドプローブの一端が支持体上にアレイ状に固定されたものであり、DNAチップ、Geneチップ、マイクロチップ、ビーズアレイ等を包含する。本発明の多型と相補的な配列を含むプローブを用いることで、網羅的に本発明の多型の有無を検出することができる。DNAチップなどのDNAマイクロアレイアッセイとしてはGeneChipアッセイが挙げられる(Affymetrix社;米国特許第6,045,996号、同第5,925,525号、及び同第5,858,659号参照)。GeneChip技術は、チップに貼り付けたオリゴヌクレオチドプローブの小型化高密度マイクロアレイを利用するものである。
TILLING(Targeting Induced Local Lesions IN Genomes)法とは、変異導入した突然変異体集団のゲノム中の変異ミスマッチをPCR増幅とCEL Iヌクレアーゼ処理によってスクリーニングする方法である。

(ii)本発明の遺伝的特徴が検出された被験ステビア植物組織のチラミンの含有量を測定する工程、
(iii)本発明の遺伝的特徴が検出された被験ステビア植物体のうち、チラミンの含有量が低い個体を選択する工程、
(iv)選択したチラミンの含有量が低い個体を他のステビア植物体と交配する工程、
(v)交配により得られた子植物体のゲノムから、本発明の遺伝的特徴を検出する工程、
(vi)本発明の遺伝的特徴が検出された子植物組織のチラミンの含有量を測定する工程、
(vii)本発明の遺伝的特徴が検出された子植物体のうち、チラミンの含有量が低い個体を選択する工程。
本発明のスクリーニング方法において、被験ステビア植物体は、突然変異誘発処理を行ったステビア植物及びその子孫植物を含んでもよい。突然変異誘発処理については、本発明の植物体の項に記載したとおりであり、変異誘発剤による処理や、放射線又は光線の照射による処理等を含む。
当該キットにおいて、(B-1)、(B-1’)及び(B-1’’)からなる群より選択される選択されるいずれか一つ以上のプライマーセットを用いる場合、前記キットに含まれる制限酵素はKpnIである。
当該キットにおいて、(B-2)、(B-2’)及び(B-2’’)からなる群より選択される選択されるいずれか一つ以上のプライマーセットを用いる場合、前記キットに含まれる制限酵素はXbaIである。
当該キットにおいて、(B-3)、(B-3’)及び(B-3’’)からなる群より選択される選択されるいずれか一つ以上のプライマーセットを用いる場合、前記キットに含まれる制限酵素はAflIIである。

具体的には、本発明のチラミン低含有ステビア植物体、本発明のスクリーニング方法により選別されたチラミン低含有ステビア植物体又は本発明の方法により製造されたチラミン低含有ステビア植物体からステビオール配糖体を含む抽出物を得る工程、及び、得られた抽出物からステビオール配糖体を精製する工程を含む、ステビオール配糖体精製品の製造方法が提供される。
また、ステビオール配糖体を含む抽出物を酢酸エチルその他の有機溶媒:水の勾配、高速液体クロマトグラフィー(High Performance Liquid Chromatography:HPLC)、ガスクロマトグラフィー、飛行時間型質量分析(Time-of-Flight mass spectrometry:TOF-MS)、超高性能液体クロマトグラフィー(Ultra (High) Performance Liquid chromatography:UPLC)等の公知の方法を用いることにより個々のステビオール配糖体を精製することができる。
本発明の抽出物は、野生型ステビア種から得られた抽出物と比べチラミンを95%以下、90%以下、85%以下、80%以下、75%以下、70%以下、65%以下、60%以下、55%以下、50%以下、45%以下、40%以下、35%以下、30%以下、25%以下、20%以下、15%以下、10%以下、5%以下、4%以下、3%以下、2%以下、1%以下、0.5%以下、0.3%以下、0.1%以下、0.05%以下、0.03%以下、0.001%以下などの含量で含んでいてもよい。ここで、本発明の抽出物と、野生型ステビア種から得られた抽出物は、同じ方法で得られたものであってよい。
本発明は、別の側面において、本発明のステビア植物体に係る塩基配列を提供する。本発明の遺伝的特徴Xを有するステビア植物体に係る塩基配列の特定の態様は、配列番号164~167から選択される塩基配列を含む、又はそれからなる。本発明の遺伝的特徴Xと遺伝的特徴Aとを有するステビア植物体に係る塩基配列の特定の態様は、配列番号164~167から選択される塩基配列と、配列番号19~21及び69から選択される塩基配列とを含む。本発明の遺伝的特徴Xと遺伝的特徴B(すなわち、本発明の遺伝的特徴B-1~B-4の少なくとも1つ)とを有するステビア植物体に係る塩基配列の特定の態様は、配列番号164~167から選択される塩基配列と、配列番号22~33及び70~73から選択される塩基配列とを含む。本発明の遺伝的特徴Xと遺伝的特徴Cとを有するステビア植物体に係る塩基配列の特定の態様は、配列番号164~167から選択される塩基配列と、配列番号34~36及び74から選択される塩基配列を含む。本発明の遺伝的特徴Xと遺伝的特徴Aと遺伝的特徴Bとを有するステビア植物体に係る塩基配列の特定の態様は、配列番号164~167から選択される塩基配列と、配列番号19~21及び69から選択される塩基配列と、配列番号22~33及び70~73から選択される塩基配列とを含む。本発明の遺伝的特徴Xと遺伝的特徴Aと遺伝的特徴Cとを有するステビア植物体に係る塩基配列の特定の態様は、配列番号164~167から選択される塩基配列と、配列番号19~21及び69から選択される塩基配列と、配列番号34~36及び74から選択される塩基配列とを含む。本発明の遺伝的特徴Xと遺伝的特徴Bと遺伝的特徴Cとを有するステビア植物体に係る塩基配列の特定の態様は、配列番号164~167から選択される塩基配列と、配列番号22~33及び70~73から選択される塩基配列と、配列番号34~36及び74から選択される塩基配列とを含む。本発明の遺伝的特徴XおよびA~Cをすべて有するステビア植物体に係る塩基配列の特定の態様は、配列番号164~167から選択される塩基配列と、配列番号19~21及び69から選択される塩基配列と、配列番号22~33及び70~73から選択される塩基配列と、配列番号34~36及び74から選択される塩基配列とを含む。
(1)チラミン含量の測定
各個体よりサンプリングした新鮮葉5gをホモジナイザーカップに採り、これに20%トリクロロ酢酸10mL及び水50mLを加えて15000rpmで10分間ホモジナイズした。これを水で200mLに定容し、混和して30分間放置後、必要に応じて遠心・濾過し試験溶液とした。試験溶液5mLを分取し、0.1mol/Lオクタンスルホン酸ナトリウム5mLを加えてよく混和後、C18、1000mgミニカラム(Sep-Pak(R) Vac 6cc 1g C18 Cartridges、Waters)に負荷した。次いで水20mLでミニカラムを洗浄した後、メタノール:水(8:2)10mLでアミン類を溶出し、溶出液を全量採取し40℃以下で減圧濃縮し、約1mLの濃縮液とした。この濃縮液に内部標準溶液(1,8-ジアミノオクタンを20μg/mLの濃度で0.5mol/L塩酸に溶解したもの)0.5mL、無水炭酸ナトリウム0.2g、1%ダンシルクロライド・アセトン溶液2mLを加えてよく混和し、遮光して45℃の水浴中で1時間加温した。加温後、10%プロリン溶液0.5mLを加え、振とうし10分間放置した。その後トルエン5mLを加えて1分間激しく振とうし、上層のトルエン層を採取した。再度トルエンを加えこの操作を2回行い、トルエン層を合わせて40℃以下で減圧濃縮・乾固した。残留物にアセトニトリル1mLを加えてよく混和したものをHPLC分析用試料とした。
(1)と同じ個体より適量の新鮮葉をサンプリングし、0.25gの新鮮葉をフリーズドライにより乾燥し、破砕物乾物0.05gを純水中に投入した。超音波処理20分にて抽出し、遠心・濾過した後に0.33mLの抽出液を得た。この抽出液をLCMS-8050(島津)にてLC/MS/MS分析を行い、RebDの濃度を定量した。
上記分析で得られたチラミン含量とRebD含量との相関性の有無を調べたところ、両者の間には、相関係数-0.615の負の相関が認められた。図1は、チラミン含量とRebD含量との相関分析の結果を示す。また、WelchのT検定で有意差の有無を判定すると、チラミン含量0.037%未満の個体のRebD含量と、それ以外の個体のRebD含量との間のp値は0.0003、チラミン含量0.058%未満の個体のRebD含量と、それ以外の個体のRebD含量との間のp値は0.0454、チラミン含量0.092%未満の個体のRebD含量と、それ以外の個体のRebD含量との間のp値は0.0239と、それぞれ有意に異なっていた。
実施例1で試験した個体2-1~2-3はいずれも、高TSG含有型の遺伝的特徴を有する雄株(P1)と、高RebM含有型の遺伝的特徴を有する雌株(P2)とを交配させて得られたものである。
高RebM含有型の遺伝的特徴は、以下の少なくとも1つの特徴を有する。
B-1:配列番号2の40位に相当する位置の塩基がTであるアレルについてホモ接合性。
B-2:配列番号3の44位に相当する位置の塩基がTであるアレルについてホモ接合性。
B-3:配列番号4の41位に相当する位置の塩基がCであるアレルについてホモ接合性。
B-4:配列番号5の55~72位に相当する部分が欠失しているアレルについてホモ接合性。
これらの遺伝的特徴が高RebM含有特性に係ることは、本出願人による未公開の先願(2018年10月12日に出願したPCT/JP2018/038064)に示されている。
高TSG含有型の遺伝的特徴は、以下の特徴を有する。
C:配列番号6の49位に相当する位置の塩基がAであるアレルについてヘテロ接合性。
上記の遺伝的特徴が高TSG含有特性に係ることは、本出願人による未公開の先願(2018年7月31日に出願した特願2018-144512)に示されている。
遺伝的特徴B-1の検出には、以下のプライマーを用いてPCRを行い、PCR産物に制限酵素(KpnI)を添加し、37℃で酵素反応を行い、制限酵素による処理を行った。制限酵素処理後、マイクロチップ型電気泳動装置LabChip GX Touch HT(PerkinElmer)により電気泳動を行い、電気泳動後のバンドパターンによってマーカーの識別を行った。
プライマーの配列は以下のとおり。
Fwプライマー:5’-TAATCATCCAAACCCTAATCTCGCCAAACAACCGGGTAC-3’(配列番号45)
Rvプライマー:5’-GAGGAAGACATTGGCAACTC-3’(配列番号46)
得られたPCR産物(約297bp長)に対しKpnI制限酵素処理を行い、約260bpの制限酵素処理産物(例えば、配列番号49)を生じなかったものを遺伝的特徴B-1陽性とした。
フォワードプライマー:5’-TTATTTAATGATCCAATGGAGGGGGTGATTCAGGTAATAAAAGGCACT-3’(配列番号64)
リバースプライマー:5’-TGAGGGTTCTCAATTGATTTCCGATTGG-3’(配列番号65)
得られた約367bpPCR産物(例えば、配列番号66又は67)に対しSpeI制限酵素処理を行い、約321bpの制限酵素処理産物(例えば、配列番号68)を生じたものを遺伝的特徴C陽性とした。
さらに、低チラミン含有個体を識別するためのマーカーを検出するため、個体2-1~2-3の新鮮葉からゲノムDNAを抽出し、NGS(HiSeq2500、Illumina)によるシーケンシングを行った。
Claims (32)
- 乾燥葉の単位質量当たりのチラミン含量が0.092%未満である、低チラミン含有ステビア植物体。
- 乾燥葉の単位質量当たりのレバウジオシドD含量が1%以上である、請求項1に記載の植物体。
- 配列番号150の201位に相当する位置の塩基がCであるアレルについてホモ接合性である、請求項1又は2に記載の植物体。
- 配列番号1の201位に相当する位置の塩基がAであるアレルについてホモ接合性である、請求項1~3のいずれか一項に記載の植物体。
- 以下の遺伝的特徴の少なくとも1つをさらに有する、請求項1~4のいずれか一項に記載の植物体。
(1)配列番号2の40位に相当する位置の塩基がTであるアレルについてホモ接合性である。
(2)配列番号3の44位に相当する位置の塩基がTであるアレルについてホモ接合性である。
(3)配列番号4の41位に相当する位置の塩基がCであるアレルについてホモ接合性である。
(4)配列番号5の55~72位に相当する部分が欠失しているアレルについてホモ接合性である。 - 配列番号6の49位に相当する位置の塩基がAであるアレルについてヘテロ接合性である、請求項1~5のいずれか一項に記載の植物体。
- 非遺伝子組換植物体である、請求項1~6のいずれか一項に記載の植物体。
- 突然変異誘発処理を行ったステビア植物体及びその子孫植物体を含む、請求項1~7のいずれか一項に記載の植物体。
- 請求項1~8のいずれか一項に記載の植物体の種子、組織、組織培養物又は細胞。
- 胚、分裂組織細胞、花粉、葉、根、根端、花弁、プロトプラスト、葉の切片及びカルスから選択される、請求項9に記載の組織、組織培養物又は細胞。
- 請求項1~8のいずれか一項に記載の植物体と第2のステビア植物体とを交雑させる工程を含む、乾燥葉の単位質量当たりのチラミン含量が0.092%未満である低チラミン含有ステビア植物体を作出する方法。
- 第2の植物体が請求項1~8のいずれか一項に記載の植物体である、請求項11に記載の方法。
- チラミン含量の低い、請求項1~8のいずれか一項に記載の植物体、請求項9又は10に記載の種子、組織、組織培養物又は細胞の抽出物。
- 請求項1~8のいずれか一項に記載の植物体、請求項9又は10に記載の種子、組織、組織培養物又は細胞から抽出物を得る工程を含む、低チラミン含有ステビア抽出物の製造方法。
- 請求項13に記載のステビア抽出物からステビオール配糖体を精製する工程を含む、低チラミン含有ステビオール配糖体精製品の製造方法。
- ステビオール配糖体が、レバウジオシドA、レバウジオシドB、レバウジオシドC、レバウジオシドD、レバウジオシドE、レバウジオシドF、レバウジオシドM、レバウジオシドN、レバウジオシドO、ステビオシド、ステビオールビオシド、ルブソシド、ズルコシドA又はこれらの組合せを含む、請求項15に記載の方法。
- 請求項15又は16に記載の方法により製造された、低チラミン含有ステビオール配糖体精製品。
- 請求項13に記載の組織、組織培養物又は細胞の抽出物、あるいは、請求項17に記載の精製品を含む、低チラミン含有飲食品、甘味組成物、香料又は医薬品。
- 請求項13に記載の組織、組織培養物又は細胞の抽出物、あるいは、請求項17に記載の精製品を提供する工程、及び
前記抽出物又は精製品を、飲食品、甘味組成物、香料又は医薬品の原料に添加する工程
を含む、飲食品、甘味組成物、香料又は医薬品の製造方法。 - 被験ステビア植物体のゲノムから、配列番号150の201位に相当する位置の塩基がCであるアレルについてホモ接合性であるという遺伝的特徴の存在及び/又は不在を検出する工程を含む、低チラミン含有ステビア植物体をスクリーニングする方法。
- 被験ステビア植物体のゲノムから、配列番号1の201位に相当する位置の塩基がAであるアレルについてホモ接合性であるという遺伝的特徴の存在及び/又は不在を検出する工程をさらに含む、請求項20に記載の方法。
- 被験ステビア植物体のゲノムから以下の(1)~(4)の遺伝的特徴の存在及び/又は不在を検出する工程をさらに含む、請求項20又は21に記載の方法。
(1)配列番号2の40位に相当する位置の塩基がTであるアレルについてホモ接合性である。
(2)配列番号3の44位に相当する位置の塩基がTであるアレルについてホモ接合性である。
(3)配列番号4の41位に相当する位置の塩基がCであるアレルについてホモ接合性である。
(4)配列番号5の55~72位に相当する部分が欠失しているアレルについてホモ接合性である。 - 被験ステビア植物体のゲノムから、配列番号6の49位に相当する位置の塩基がAであるアレルについてヘテロ接合性であるという遺伝的特徴の存在及び/又は不在を検出する工程をさらに含む、請求項20~22のいずれか一項に記載の方法。
- 遺伝的特徴を検出する工程が、CAPS法、dCAPS法又はTaqMan PCR法を用いて行われる、請求項20~23のいずれか一項に記載の方法。
- 被験ステビア植物組織のチラミンの含有量を測定する工程をさらに含む、請求項20~24のいずれか一項に記載の方法。
- 配列番号150の201位に相当する位置の塩基がCであるアレルについてホモ接合性であるという遺伝的特徴の存在及び/又は不在を検出するための試薬を含む、低チラミン含有ステビア植物体のスクリーニングキット。
- 配列番号1の201位に相当する位置の塩基がAであるアレルについてホモ接合性であるという遺伝的特徴の存在及び/又は不在を検出するための試薬をさらに含む、請求項26に記載のキット。
- 以下の(1)~(4)の遺伝的特徴の存在及び/又は不在を検出するための試薬をさらに含む、請求項26又は27に記載のキット。(1)配列番号2の40位に相当する位置の塩基がTであるアレルについてホモ接合性である。
(2)配列番号3の44位に相当する位置の塩基がTであるアレルについてホモ接合性である。
(3)配列番号4の41位に相当する位置の塩基がCであるアレルについてホモ接合性である。
(4)配列番号5の55~72位に相当する部分が欠失しているアレルについてホモ接合性である。 - 配列番号6の49位に相当する位置の塩基がAであるアレルについてヘテロ接合性であるという遺伝的特徴の存在及び/又は不在を検出するための試薬をさらに含む、請求項26~28のいずれか一項に記載のキット。
- 試薬が、CAPS法、dCAPS法又はTaqMan PCR法に使用するプライマー及び/又はプローブを含む、請求項26~29のいずれか一項に記載のキット。
- 配列番号150の201位に相当する位置にTからCへの変異を導入する工程を含む、低チラミン含有ステビア植物体を作出する方法。
- 変異の導入が、突然変異誘発処理によって行われる、請求項31に記載の方法。
