Abstract
Description
mGlu受容体は8つのサブタイプ1から8(mGluR1から8)が存在し、相同性と共役する情報伝達系及び薬理学的特徴に基づいてグループI(mGluR1、mGluR5)、グループII(mGluR2、mGluR3)及びグループIII(mGluR4、mGluR6、mGluR7、mGluR8)に分類される。グループII mGlu受容体(mGluR2、mGluR3)は主にプレシナプスに存在し、グルタミン酸の放出を抑制的に調節している。したがって、mGlu2/3受容体ネガティブアロステリックモジュレーター(NAM)は抗うつ薬(非特許文献4)、認知機能増強剤(非特許文献5)になる可能性が報告されている。

本発明の化合物は、例えば、下記製造法1~12に示す方法によって製造することができる。これらの製造方法は、有機合成化学に習熟している者の知識に基づき、適宜改良され得る。原料として用いられる化合物は、必要に応じてその塩、又は官能基が保護されたものを用いてもよい。

合成法-2:参考例12の化合物(177mg)、参考例6の化合物(92mg)、炭酸カリウム(196mg)、テトラキス(トリフェニルホスフィン)パラジウム(0)(27mg)、1,4-ジオキサン(1.9mL)および水(0.47mL)の混合物を90℃で3時間半攪拌した。20℃まで冷却し、飽和食塩水を加えて酢酸エチルで抽出した。有機層を無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣を酢酸エチル(約1.5mL)中で室温下撹拌し、固体をろ取した。固体を酢酸エチル(約0.5mL)で洗い、表題化合物(102mg)を得た。
合成法-3:参考例4の化合物(116g)、参考例6の化合物(67.0g)、酢酸パラジウム(6.20g)、X-phos(26.3g)、炭酸カリウム(76g)の1,2-ジメトキシエタン(700mL)/水(350mL)混合物を加熱還流下3時間撹拌した。反応混合物を室温まで放冷後、酢酸エチル(500mL)と2mol/L塩酸(1000mL)を加え、室温で30分間撹拌した。混合物をセライトろ過し、残渣を酢酸エチル(500mL)で洗浄後、ろ液を分液した。有機層を2mol/L塩酸(500mL)で2回洗浄し、合わせた水層に10mol/L水酸化ナトリウム水溶液を加え、pH9とした。水層をクロロホルム(500mL)で3回抽出した後に、飽和食塩水で洗浄後、無水硫酸ナトリウムで乾燥、ろ過し、減圧濃縮した。残渣(125.3g)にエタノール(1253mL)を加え、加熱還流下、30分間撹拌した。混合物に水(626mL)を加え、さらに加熱還流下撹拌した。室温まで放冷後、固体をろ取し、酢酸エチルで洗浄した。得られた固体(44.4g)に酢酸エチル(666mL)を加え、加熱還流下、30分間撹拌した。室温まで放冷後、固体をろ取し、酢酸エチルで洗浄し、減圧下乾燥することにより表題の化合物(39.77g)を得た。
合成法-4:参考例4の化合物(28.5g)、参考例19の化合物(33.5g)、酢酸パラジウム(1.519g)、S-phos(5.56g)、炭酸カリウム(18.70g)の1,2-ジメトキシエタン(100mL)/水(50mL)混合物を加熱還流下3時間撹拌した。反応混合物を室温まで放冷後、クロロホルム(180mL)/メタノール(20mL)で希釈しセライトろ過し、クロロホルム(450mL)/メタノール(50mL)で洗浄した。得られたろ液に炭酸水素ナトリウム水溶液を加え分液した。有機層を飽和食塩水で洗浄後、無水硫酸ナトリウムで乾燥、ろ過し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/メタノール)により精製することで固体を得た。得られた固体(21.23g)を酢酸エチル(210ml)で洗浄し、固体をろ取した。ろ取した固体(13.65g)のクロロホルム(117mL)/メタノール(13mL)溶液に、活性炭(商品名「強力白鷺」)(6.5g)を加え、室温にて1時間撹拌した。その後、溶液にArgoregin MP-TMT(6.5g)を加え、室温にてさらに1時間撹拌した。得られた溶液をセライトろ過し、クロロホルム(720mL)/メタノール(80mL)で洗浄した後、ろ液を減圧濃縮することにより表題の化合物(12.03g)を得た。

以下に、本発明の化合物についての薬理試験方法及びその結果を示すが、本発明はこれらの試験例に限定されるものではない。
(1)ヒトmGlu2受容体安定発現細胞
ヒトmGlu2受容体安定発現細胞を作製し、培養に供した。具体的には、pcDNA4/TO(K1020-01, Lifetechnologies, Carlsbad,CA,USA)にヒトmGlu2受容体遺伝子を挿入し、TR発現ヒト腎臓由来HEK細胞(cat#CCL-82.2, ATCC, USA)に導入した。その後Geneticin(cat#10131-027, Lifetechnologies, Carlsbad, CA, USA)による選別を行い、ヒトmGlu2受容体安定発現細胞を得た。
培地には10% Dialysed-FBS(cat#26400-044, Lifetechnologies, Carlsbad, CA, USA), 50μg/mL Blasticidin S(cat#ANT-BL-1, Lifetechnologies, Carlsbad, CA, USA)、2mg/mL G418(cat#16513-84, nacalai tesque, Kyoto, Japan)を含むHigh glucose-DMEM培地(cat#11995-065, Lifetechnologies, Carlsbad, CA, USA)を用い、細胞培養用フラスコ(cat#3133-150, AGC Thechno Glass, Shizuoka, Japan)にて培養を行った。培養中、3~4日毎にTrypLE Express(cat#12604-013, Lifetechnologies, Carlsbad, CA, USA)処理にて細胞を回収し、継代培養を行った。
継代から3~4日後、約80%コンフルエントな状態でTrypLE Express処理にて細胞を回収し、0.1% BSA(cat#12604-013, Lifetechnologies, Carlsbad, CA, USA), 0.1μg/mL Tetracycline(cat#33031-64, nacalai tesque, Kyoto, Japan)含有のHanks(cat#14065-056, Lifetechnologies, Carlsbad, CA, USA)/20mmol/L HEPES(cat#15630-080, Lifetechnologies, Carlsbad, CA, USA)Buffer(pH7.4)培地下で、Gα16, apoaequorinを一過的に導入し、その後に384wellプレート(cat#781090, Greiner bio-one, Frickenhausen, Germany)に1,500cells/30μL/wellとなるように播種した。
播種翌日、Coelentetrazine h(cat#S2011, Promega, Madison, WI, USA)を終濃度1μmol/Lとなるように添加し(10μl/well)、遠心後、室温で4時間以上静置した。
(2)試験化合物の調製
試験化合物はDMSOを用いて評価濃度の1000倍濃度になるように溶解した。このDMSO溶液を、培地(Hanks、20mmol/L HEPES、0.1% BSA)にて評価濃度の6倍濃度になるように希釈した。GlutamateはHanks/20mmol/L HEPES/0.1% BSA培地にてEC80濃度の6倍濃度になるように希釈した。
(3)mGlu2受容体ネガティブアロステリックモジュレーター活性評価
ヒトmGlu2受容体安定発現細胞を作製し、培養に供した。mGlu2受容体刺激による発光シグナルの検出にはFDSS7000(浜松ホトニクス)を用いた。細胞及び発光基質を添加したプレートに、上記調製の化合物溶液を添加した(10μl/well)。添加120秒後EC80のGlutamate含有溶液を添加し(10μl/well)、添加後300秒間発光シグナル(中心波長:465nm)を測定してRLU(Integration)を算出した。化合物のmGlu2受容体のネガティブアロステリックモジュレーター活性は、(100-100×(各化合物の各濃度のRLU/DMSO群のRLU))にて算出した。
本発明の化合物を上述の生物学試験で評価したところ、mGlu2受容体ネガティブアロステリックモジュレーター活性を示す化合物を見出した。各化合物のmGlu2受容体ネガティブアロステリックモジュレーター活性(IC50値(nmol/L))を下表に示す。
ヒトmGlu2受容体安定発現細胞を作製し、培養に供する。mGlu2受容体刺激による発光シグナルの検出にはFDSS7000(浜松ホトニクス)を用いる。細胞及び発光基質を添加したプレートに、上記(2)で調製した試験化合物溶液を添加する(10μl/well)。添加2分後、15分後、30分後、60分後及び120分後にそれぞれEC80のGlutamate含有溶液を添加し(10μl/well)、添加後300秒間発光シグナル(中心波長:465nm)を測定してRLU(Integration)を算出する。化合物各添加時間のmGlu2受容体のネガティブアロステリックモジュレーター活性は、(100-100×(各化合物の各濃度のRLU/DMSO群のRLU))にて算出する。
ラット強制水泳試験において、既存の抗うつ薬(三環系抗うつ薬やセロトニン再取込阻害剤など)は、無動時間を短縮させることが知られている。そこで本試験系を用い、無動時間を指標に本発明の化合物の抗うつ作用を検討する。
7週齢雄性Wistarラットを用いて、ラット強制水泳試験を行う。すなわち水道水(水温25±1℃)5.8Lを満たした水槽に動物を入れた後、15分間泳がせる(水泳訓練)。水泳訓練終了後、速やかに動物に付着している水滴を拭い取りホームケージに戻す。水泳訓練の翌日、水泳試験は動物を水泳訓練と同様の操作で5分間水泳させることによって実施する。各個体の水泳行動は水槽の側面からビデオカメラで撮影する。水泳試験終了後、速やかに動物に付着している水滴を拭い取りホームケージに戻した。被験化合物、陽性対照であるimipramineは、0.5%メチルセルロース溶液に懸濁し経口投与する。媒体及び被験化合物の投与は、水泳訓練終了15分後及び水泳試験の2時間前に行う。Imipramineの投与は、水泳訓練終了15分後及び水泳試験の1時間前に行う。動物が水槽中で前肢及び胴体を動かさずに浮遊している状態を無動と定義し、5分間の水泳試験における無動を示した時間の累積値を、その個体の無動時間として計測する。統計学的処理にはスチューデントのt検定及びDunnet型多重比較を用いる。
皮質脳波におけるγ周波数帯パワー変化は、皮質活動の指標と考えられており、mGluR2のアンタゴニストやNAM、あるいはNMDA受容体の拮抗薬(Ketamineなど)では、γ周波数帯パワーを増強させることが知られている。本発明の化合物においてもγ周波数帯増強作用を示すかどうかについて検討する。
脳波測定電極留置手術を施した雄性Wistarラットを用いて、暗期に試験を行う。化合物は0.5%メチルセルロース溶液に懸濁し、経口投与する。脳波測定及び周波数解析はProgress in Neuro-Psychopharmacology & Biological Psychiatry 63 (2015) 6-13に記載の方法に従う。脳波は0.5から80Hzの間で周波数解析を行い、30から80Hzの周波数帯をγ周波数帯として、パワー値を算出する。投与前値を100%として投与後2時間までのパワー変化を1時間毎に集計する。統計学的処理は繰り返し測定を考慮した二元配置分散分析後、Dunnet型多重比較を行う。
チトクロームP450(以下CYP)は薬物代謝に関わる最も重要な酵素群で、薬物動態学的な相互作用の多くはこれらCYP活性の阻害に基づく。CYPには複数の分子種が存在し、特にCYP3A4はCYPによる酸化反応では医薬品の代謝に関与する割合が最も大きく、また、肝臓に存在するCYPのうちの大部分を占める。
CYP阻害様式には「可逆的阻害」と「不可逆的阻害(mechanism-based inhibition:MBI)」の大きく2種類があり、特にMBIに基づくCYP阻害は薬物相互作用のみならず重篤な副作用(肝毒性等)を引き起こす可能性が知られている(Curr Opin Drug Discov Devel. 2010 January, 13(1), 66-77,Therapeutics and Clinical Risk Management, 2005,1(1), 3-13)。
本実施例化合物を用いて、CYP3A4のMBI及び酵素不活性化クリアランスを評価した。
酵素源としてヒト肝ミクロソーム、基質はミダゾラム又はテストステロンを用い、CYP3A4に対する試験化合物の阻害効果ならびに阻害様式を評価した。37℃、30分の代謝反応後の試験化合物添加群(4濃度)の基質由来代謝生成物と非添加群の基質由来代謝生成物をLC-MS/MSにて測定し、その比から阻害率を計算した。また、濃度プロットからIC50値を算出した。試験化合物にMBI作用が認められる場合、NADPH(補因子)存在下でのプレインキュベーション後に基質を添加し反応を開始することにより、IC50が低下することが知られていることから(Xenobiotica,2009,39(2),99-112)、プレインキュベーションによるIC50値の変動が2倍以上の場合MBI作用ありと判定した。
MBI作用が認められた場合は、kinact(最大酵素不活性化速度定数)及びKI(最大酵素不活性化速度の50%の速度をもたらす阻害薬の濃度)を非線形最小二乗法により算出した。さらに、Drug Metabolism and Disposition,2011,39(7),1247-1254に記載の方法に準じ、酵素不活性化クリアランスを算出した(CLint = kinact/KI(ml/min/mmol)×CYP contents(pmol/mg protein))。
各実施例化合物のCYP3A4のMBI及び酵素不活性化クリアランスを下表に示す。

R3及びR4が、各々独立して、水素原子、フッ素、塩素、ジフルオロメチル、トリフルオロメチル又はジフルオロメトキシであり;
R5及びR6が、各々独立して、水素原子又は-NH2であり;
Xが、窒素原子又は-CH-である、

(7S)-3-(イミダゾ[1,2-a]ピラジン-6-イル)-7-メチル-5-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロピラゾロ[1,5-a]ピラジン-4(5H)-オン;
(7S)-3-(イミダゾ[1,2-a]ピリジン-6-イル)-7-メチル-5-[3-クロロ-4-フルオロフェニル]-6,7-ジヒドロピラゾロ[1,5-a]ピラジン-4(5H)-オン;
(7S)-5-[4-(ジフルオロメチル)フェニル]-3-(イミダゾ[1,2-a]ピリジン-6-イル)-7-メチル-6,7-ジヒドロピラゾロ[1,5-a]ピラジン-4(5H)-オン;
(7S)-5-(4-クロロ-3-フルオロフェニル)-3-(イミダゾ[1,2-a]ピリジン-6-イル)-7-メチル-6,7-ジヒドロピラゾロ[1,5-a]ピラジン-4(5H)-オン;

(7S)-7-メチル-3-(3-メチルイミダゾ[1,2-a]ピリジン-6-イル)-5-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロピラゾロ[1,5-a]ピラジン-4(5H)-オン;
(7S)-3-(3-エチルイミダゾ[1,2-a]ピリジン-6-イル)-7-メチル-5-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロピラゾロ[1,5-a]ピラジン-4(5H)-オン;及び
(7S)-7-メチル-3-(3-プロピルイミダゾ[1,2-a]ピリジン-6-イル)-5-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロピラゾロ[1,5-a]ピラジン-4(5H)-オン。 - 請求項1~20のいずれか一項に記載の化合物、又はその製薬学的に許容される塩を含有する医薬組成物。
- 請求項1~20のいずれか一項に記載の化合物、又はその製薬学的に許容される塩を有効成分として含有する、グループII mGlu受容体が関与する疾患の治療剤及び/又は予防剤。
- グループII mGlu受容体が代謝型グルタミン酸受容体サブタイプ2(mGluR2)である、請求項22に記載の治療剤及び/又は予防剤。
- グループII mGlu受容体が関与する疾患が、精神疾患又は神経変性疾患である、請求項22又は23に記載の治療剤及び/又は予防剤。
- 精神疾患又は神経変性疾患が、うつ病性障害、抑うつ障害、双極性障害及び関連障害、不安障害、心的外傷後ストレス障害、強迫性障害、急性ストレス障害、統合失調症、自閉症スペクトラム障害、アルツハイマー病、認知機能障害、認知症、薬物依存、肥満、痙攣、振戦、疼痛又は睡眠障害である、請求項24に記載の治療剤及び/又は予防剤。
- グループII mGlu受容体が関与する疾患の治療剤及び/又は予防剤を製造するための、請求項1~20のいずれか一項に記載の化合物又はその製薬学的に許容される塩の使用。
- グループII mGlu受容体が関与する疾患の治療及び/又は予防に使用するための、請求項1~20のいずれか一項に記載の化合物、又はその製薬学的に許容される塩。
- 治療が必要な患者に、治療上の有効量の請求項1~20のいずれか一項に記載の化合物又はその製薬学的に許容される塩を投与することを含む、グループII mGlu受容体が関与する疾患を治療及び/又は予防するための方法。
- 請求項1~20のいずれか一項に記載の化合物又はその製薬学的に許容される塩を含有する医薬と、うつ病性障害、抑うつ障害、双極性障害及び関連障害、不安障害、心的外傷後ストレス障害、強迫性障害、急性ストレス障害、統合失調症、自閉症スペクトラム障害、アルツハイマー病、認知機能障害、認知症、薬物依存、肥満、痙攣、振戦、疼痛又は睡眠障害の治療剤から選択される1種類以上の薬剤とを組み合わせてなる医薬。
- 1種以上の抗精神病薬と併用して、精神疾患又は神経変性疾患を治療するための、請求項1~20のいずれか一項に記載の化合物又はその製薬学的に許容される塩を含有する医薬。
- 式(1a)で表される化合物またはその製薬学的に許容される塩:
