Abstract

L'invention concerne des procédés d'isolement d'une molécule nucléique cible d'intérêt à partir d'un échantillon. Les procédés comprennent la mise en contact d'un échantillon comprenant une molécule nucléique cible d'intérêt avec au moins un acide nucléique à simple brin ciblant une sonde comprenant une séquence d'acide nucléique d'environ 30 nucléotides et la longueur de l'acide nucléique cible qui s'hybride à la molécule d'acide nucléique cible d'intérêt dans des conditions d'hybridation hautement stringentes. La sonde comprend une fraction de capture liée de façon covalente à une ou plusieurs bases nucléotidiques dans la sonde. La sonde de ciblage peut être capturée avec un agent de liaison spécifique qui se lie spécifiquement à la fraction de capture, isolant ainsi la molécule nucléique cible d'intérêt. Dans certains modes de réalisation, l'échantillon est en outre mis en contact avec un agent de réticulation protéine-acide nucléique, un agent de réticulation acide nucléique-acide nucléique, un agent de réticulation protéine-protéine ou n'importe quelle combinaison de ceux-ci, avant la mise en contact de l'échantillon avec une sonde de ciblage. Par conséquent, les complexes formés avec l'acide nucléique cible peuvent également être isolés.
