化合物1について、先に述べた方法により合成した。以下に示す処方に従って、本発明のシワ改善剤を含む化粧料1(ローション)を調製した。
上記化粧料1について、光老化モデルを用いた試験におけるシワ改善効果を評価した。
試験開始時6週齢のへアレスマウス1群5匹、対照群(エタノ-ル溶媒投与群)と検体投与群の2群で計10匹用い、ヘアレスマウス背部にUVBを、1日1回、週3回の頻度にて10週間連続で照射することにより光老化を誘発させた。UVB照射量は、最初の1週間を50 mJ/cm2、2週目以降を100 mJ/cm2とした。
検体投与群には、上記化粧料1をへアレスマウス背部に投与した。10週間のUVB照射終了後、翌日からヘアレスマウス背部に化粧料1の投与を開始し、1日1回100μLを8週間投与した。対照群には、表1の化粧料1(ロ-ション1)に代えてエタノ-ルを投与し、あとの処置は検体投与群と同様に行った。投与終了1日後にレプリカを採取し、30度の斜光照明下で投影された2cm四方のレプリカ像を用い、測定者によるシワスコア基準(表2)に従ったスコア化作業を行い、シワ改善効果を評価した。スコア化作業は、3名の測定者が個別に行い、目視によりスコア1~6まで、0.5刻みで評価した結果に付いて平均値を算出した。
化合物2および3について、先に述べた方法により合成した。化合物1を化合物2および3に変更した以外は実施例1と同様の方法で、化粧料2および化粧料3を調製した。化粧料1の場合と同様にシワ改善作用を評価した結果を図2及び図3に示す。これらの図より、本発明の化合物2及び化合物3は、優れたシワ改善効果を有することがわかる。
本発明の化合物1~8を、先に述べた方法により合成し、プロコラーゲン(procollagen)産生促進作用を評価した。
ケラチノサイト増殖用培地(Humedia-KG2、倉敷紡績株式会社製)を用い、4.5×104細胞のヒト由来正常ケラチノサイト培養細胞を24ウェルプレ-トに播種し、37℃・二酸化炭素濃度5%中にて4日間培養した。同時に、10%FBSを添加したDMEM培地(SIGMA製)を用い、2.5×104細胞のヒト由来正常皮膚ファイブロブラスト培養細胞を24ウェルプレ-トに播種し、37℃・二酸化炭素濃度5%中にて培養した。
次に、2% FBSを添加したDMEM培地(SIGMA製)に、最終濃度10μMになるように化合物1~8をそれぞれ添加した培地を作製した。また、比較対照(Control)には、化合物1~8に換わり最終濃度0.01×10-3(v/v%)になるようにジメチルスルホキシド(シグマ・アルドリッチ社製)及び50%エタノール(シグマ・アルドリッチ社製)を添加した培地を作製した。
培養したケラチノサイトをPBS(和光純薬工業株式会社製)にて洗浄した後、前記のそれぞれの化合物を含む培地、ならびにジメチルスルホキシド及び50%エタノールを含む培地に置換し、37℃・二酸化炭素濃度5%中にて24時間培養した。24時間後、この培養上清を回収した。
培養したファイブロブラストをPBSにて洗浄した後、回収した培養上清に置換し、37℃・二酸化炭素濃度5%中にて48時間培養した。48時間後、ファイブロブラストをPBSで洗浄した後、DMEM(SIGMA製)に置換し、37℃・二酸化炭素濃度5%中にて2時間培養後、培養上清を回収した。この培養上清中のプロコラーゲン量をELISA法を用いて測定した。
ジメチルスルホキシド及び50%エタノール添加培地で培養したファイブロブラストを比較対照(Control)とし、比較対照に対する化合物1~8それぞれのプロコラーゲン産生量の比率を図4~7に表した。
Claims (10)
- 下記一般式(1)で表される化合物、その立体異性体、又はそれらの薬理学的に許容される塩を含有する、シワ改善剤。
[式中、R1は、水素原子、又は炭素数1~8の直鎖若しくは分岐のアルキル基を表し、R2は、-SH、-SO3H、-S-S-X1、-S-X2、-SO-X3、-SO2-X4、-SO2-NY1-X5、又は-SO2-NY2-Y3を表し、前記X1~X5は、独立して、水素原子若しくは炭素原子が複素原子で置換されていても良い、炭素数1~8の脂肪族炭化水素基又は炭素数5~12の芳香族部であり、前記Y1~Y3は、独立して、水素原子、又は炭素数1~8の直鎖若しくは分岐のアルキル基を表し、R3は、水素原子、又は炭素数1~8の直鎖若しくは分岐のアルキル鎖を有するアシル基を表し、R4は無置換若しくは置換基を有していても良い、炭素数5~12の芳香族基又は多環縮合芳香族基を表し、mは、0~3の整数を表し、nは、1又は2の整数を表す。] - 前記一般式(1)で表される化合物が、下記一般式(5)で表される化合物である、請求項1に記載のシワ改善剤。
