Abstract

L'invention concerne une nouvelle catégorie de réactions isothermes pour amplifier l'ADN. Ces réactions homogènes synthétisent rapidement des oligonucléotides courts (par ex. 8-16 bases), spécifiés par la séquence d'une matrice d'amplification. Des versions des réactions peuvent se poursuivre soit en mode d'amplification linéaire, soit en mode d'amplification exponentielle. Ces deux formes de réaction font appel à des conditions simples et stables. Le taux d'amplification dépend entièrement des paramètres moléculaires régissant les interactions des molécules dans la réaction. La version exponentielle du procédé est une réaction moléculaire en chaîne utilisant les produits oligonucléotides de chaque réaction linéaire, afin de créer un producteur de davantage du même oligonucléotide. Il s'agit d'une réaction en chaîne à haut niveau de sensibilité qui peut être déclenchée par des séquences d'ADN spécifiques et peuvent effectuer des amplifications supérieures à 106 plis. Plusieurs réactions similaires de ladite catégorie sont décrites dans la présente invention. La robustesse, la vitesse et la sensibilité de la réaction exponentielle la rendent utile dans la détection rapide de la présence de petites quantités d'une séquence d'ADN spécifique dans un échantillon, et une série d'autres applications comprenant plusieurs utilisations courantes de la réaction en chaîne de la polymérase (PCR).
