Abstract

La présente invention est relative à un procédé de préparation d'une biopuce activée pour la fixation covalente de sondes oligonucléotidiques sur un support solide par l'intermédiaire d'un composé espaceur de type NHS-PEG-VS, ainsi que les biopuces susceptibles d'être obtenues par un tel procédé. L'invention comprend également des méthodes de détection d'acides nucléiques dans un échantillon ou des méthodes de criblage de composés capables de se fixer spécifiquement sur des sondes oligonucléotidiques dans lesquelles sont mises en oeuvre les biopuces selon l'invention. La présente invention a aussi pour objet des kits de détection, d'analyse quantitative ou qualitative d'acides nucléiques dans un échantillon, comprenant de telles biopuces ainsi que l'utilisation de ces dernières comme matrice d'affinité pour la purification d'acide nucléique, pour le séquençage d'acide nucléique, pour l'analyse qualitative ou quantitative de l'expression de gènes ou encore pour l'étude et la détection de polymorphisme génétique.
