Abstract

L'invention concerne une bibliothèque combinatoire de fragments d'anticorps semi-synthétiques fondée sur des gènes d'immunoglobuline aviaires. Ces gènes d'immunoglobuline aviaires sont des gènes de l'immunoglobuline de volailles ou de poulets. Les anticorps sont des fragments d'anticorps monocaténaires et peuvent être représentés sur de grands phages. Au moins une partie de la troisième région déterminante de complémentarité (CDR 3) du domaine variable (V) des chaînes lourdes (H) des fragments d'anticorps est remplacée par des peptides randomisés synthétiquement. Des régions variables lourdes (VH) randomisées synthétiquement sont appariées avec le répertoire naturel de régions VH aviaires et non avec des chaînes légères définies. Cet arrangement est effectué de façon que les régions variables légères (VL) et les régions variables lourdes (VH) contribuent à la diversité combinatoire et que le répertoire VH naturel des régions VH aviaires augmente cette diversité.
