  • a perfusion flow controller switches the flow of solution through the microchannel from delivering cells to delivering drug solutions and the experiment is initiated.
  • Patch-clamp modules may be cascaded such that in the event more than one cell is detected by the FACS, multiple recordings may be made. Excess cells are simply recycled back to the cell incubator.
  • the FACS front end is not required although it would have the beneficial effect of eliminating debris from the system. In the case where cells come from a mixed background a FACS front end allows selection of even minor components of the overall cell suspension.
  • the system can be fully automated and because it also recycles cells and solutions, can run for extended periods of time without intervention.
  • a device for supplying conventional glass patch pipettes will be incorporated or alternatively a system for rejuvenating and hence re-using quartz glass pipettes will be used.
  • Data obtained can be automatically downloaded to a server for off-line analysis etc. without interrupting data acquisition.

Abstract

La présente invention concerne un procédé permettant de réaliser à partir d'une seule cellule un enregistrement selon la technique patch-clamp. On commence par établir un micro-canal admettant un écoulement axial du liquide. Dans ce micro-canal, on ménage au moins un orifice permettant un accès radial depuis l'extérieur du micro-canal (air) vers l'intérieur du micro-canal (liquide), le liquide formant à l'intérieur du micro-canal au niveau de l'orifice un ménisque constituant une interface air-liquide. Avec une pipette pour procédé patch-clamp dont la pointe, capable de passer dans l'orifice d'accès, permet de former entre la pointe et la cellule une fermeture électrique à haute résistance, c'est-à-dire giga-ohmique, on laisse passer axialement dans la longueur du micro-canal le liquide portant la cellule, ce qui amène la cellule jusqu'à l'orifice d'accès. Après avoir déplacé la pointe de pipette radialement par rapport au micro-canal jusqu'à toucher l'interface air-liquide dans l'orifice d'accès, on applique au patch-clamp une force d'aspiration de façon à attirer sur la pointe la cellule pour former une fermeture. Il ne reste plus qu'à réaliser l'enregistrement en patch-clamp. L'invention concerne également un appareil pour mettre en oeuvre ce procédé.
