- the present invention relates to devices and methods for picking up spots of gel from sheets of gel.
- biochemical analytical methods produce results in the form of sheets of gel upon which the analysed substances are grouped in spots. Further analyse of the substances can be made by cutting out and picking up the cylindrical plugs of gel containing the substances of interest and transferring the plugs to other devices such as microtitre plates or test tubes for further analysis.
- the plugs are extracted from the sheet of gel by lowering a cylindrical hollow needle through liquid covering the gel and the gel until it reaches the supporting base plate, applying a suction to the upper end of the needle in order to lift the plug off the base plate and into the needle where it is caught by a pieced bulkhead near the tip of the needle.
- the plug can then be transported by the picker head to a position above a well in a micro-titre plate or a test tube and then ejected out of the needle by pressurised liquid applied to the upper part of the needle.
- a device suitable for performing this method is known from JP 3240482.
- a problem with this method is that some of the liquid covering the gel can often collect around the base of the needle where it is mounted in the picking head.
- the ejecting liquid will follow the plug into the well. This liquid is ejected with a high velocity and may bounce back up the sides or bottom of the well, drawing the plug with it. If this ej ected liquid contacts the liquid already surrounding the base of the needle then capillary action will tend to draw the ejected liquid into the liquid around the base. This will also draw the plug with it, leading to the plug becoming attached to the outside of the needle instead of being in the well.
- the purpose of the present invention is to overcome the problems of the prior art spot pickings devices. This is achieved by a device having the characterising features of claim 1.
- the present invention will be described in more detail below by means of non-limiting examples of preferred embodiments and with reference to the following drawings.

- the upper end of body 5 is adapted by means of complementing fastening means (which are well known in the prior art and therefore not shown) to be attached to a picker arm (not shown).
- the lower end of body 5 has an opening 6 to which a hollow needle 7 is attached. Needle 7 is open at each end.
- the upper end of body 5 can be connected to a supply of liquid at a pressure lower than atmospheric pressure in order to form a suction at the lower end of needle 7 and it can also be connected to a supply of pressurised liquid in order to form an overpressure at the lower end of needle 7.
- Needle 7 contains a bulkhead 9 with a centre hole 11. When a gel plug 13 is sucked up from the plate 3, the bulkhead 9 prevents it from being sucked all the way up into the body 5.
- Picker head 1 is shown in figure 1 in a position where it has been lowered through the fluid 4 and the sheet of gel 2 and is in contact with the plate 3.
- the needle contains a plug 13 of gel.
- the gel can be lifted of the plate 3 by connecting a supply of liquid at a pressure lower than atmospheric pressure to the upper end of body 5. This sucks the gel plug 13 off the plate 3 and draws it and some of the fluid 4 up into the needle 7 until the gel plug comes into contact with the bulkhead 9.

- the needle In order to allow the needle to be made small but strong enough to withstand the forces applied when it is pressed though the gel 2 and against the underlying plate 3, it is preferable that it is made of a high-strength material such as a titanium alloy (e.g. titanium grade 5) or a ceramic material (e.g. aluminium oxide) or the like.
- the needle 47 is provided with a hydrophobic coating 49 (not drawn to scale) which covers parts of the exterior parts of the body 45 and needle 47 which during use are not intended to be in contact with a fluid but which accidentally may collect fluid.
- substantially the whole of the exterior surface of the needle 47 is provided with this hydrophobic coating 49 as well as the exterior of the bottom end of the body 45.
- the hydrophobic layer 49 can preferably be made of a thin layer of Teflon TM (PTFE) which is hydrophobic and has the advantage that it can be applied in very thin layers, thereby keeping the diameter of the needle 47 small.
