Abstract

La présence d'un analyte contaminant dans un mélange de protéines peut obscurcir la détection d'une protéine recherchée. En outre, la détection des anticorps à l'élution est obscurcie par le passage continu de protéines contaminantes lorsque l'on charge des colonnes d'affinité avec du fluide de culture cellulaire. La présente invention concerne un dosage chromatographique en ligne qui permet une mesure en temps réel d'un analyte contaminant en chromatographie préparative, durant la purification ou l'élution d'un anticorps pendant le chargement et le recueil d'une protéine recherchée. Les courbes d'élution sont présentées pour permettre à l'utilisateur de mettre fin à un chargement de fluide de culture cellulaire et/ou de stopper un recueil d'éluat, pour maximiser la purification et éliminer le gaspillage de fluide de culture cellulaire.
