Abstract
A murine cDNA clone encoding a BIN1-Associated U1-specific protein (Bau) are provided. Also provided are methods of using the nucleic acid sequences, polypeptides, and antibodies directed against same in the diagnosis and treatment of cancers, hyperplastic disease states, or degenerative diseases.
Claims
1. A mammalian nucleic acid sequence encoding a BIN 1 -Associated Ul -specific protein (Bau) or a fragment thereof, isolated from cellular materials with which it is naturally associated, selected from the group consisting of:
(a) SEQ ID NO: 1;
(b) a nucleotide sequence comprising 50 nucleotides which hybridizes to SEQ ID NO:l under stringent conditions;
(c) an allelic variation of SEQ ID NO: 1 ; and
(d) a fragment of SEQ ID NO: 1 comprising 15 nucleotides selected from the sequences of nucleotides 1 to 300 and 325 to 926 of SEQ ID NO:l.
2. The mammalian nucleic acid sequence according to claim 1 which encodes murine Bau SEQ ID NO: 2 or a fragment thereof.
3. The mammalian nucleic acid sequence according to claim 1, wherein the fragment of (d) is selected from the group consisting of:
(a) nucleotides 231 to 674 of SEQ ID NO: 1 ;
(b) nucleotides 48 to 527 of SEQ ID NO:l;
(c) nucleotides 312 to 362 of SEQ ID NO: 1;
(d) nucleotides 294 to 356 of SEQ ID NO: 1; and
(e) nucleotides 540 to 554 of SEQ ID NO: 1.
4. A murine cDNA sequence SEQ ID NO: 1 which encodes a Bau polypeptide. 45
5. A mammalian BIN 1 -Associated Ul-specific protein (Bau) polypeptide, said polypeptide selected from the group consisting of:
(a) murine Bau, SEQ ID NO:2;
(b) human Bau;
(c) a fragment of (a) or (b) having biological activity; and
(d) analogues or homologs of (a) or (b) characterized by having at least 85% homology with SEQ ID NO: 2.
6. The Bau polypeptide according to claim 5, wherein the fragment is selected from the group consisting of:
(a) amino acids 62 to 209 of SEQ ID NO:2;
(b) amino acids 1 to 160 of SEQ ID NO:2;
(c) amino acids 89 to 105 of SEQ ID NO:2;
(d) amino acids 83 to 103 of SEQ ID NO:2; and
(e) amino acids 165 to 169 of SEQ ID NO:2.
7. A vector comprising a mammalian nucleic acid sequence encoding a BIN 1 -Associated Ul-specific protein (Bau) polypeptide under the control of suitable regulatory sequences, said polypeptide selected from the group consisting of:
(a) murine Bau SEQ ID NO:2;
(b) human Bau;
(c) a fragment of (a) or (b) having biological activity; and
(d) analogues or homologs of (a) or (b) characterized by having at least 90% homology with SEQ ID NO: 2.
8. A host cell transformed with the vector according to claim 6. 46
9. An oligonucleotide probe comprising a mammalian nucleic acid sequence encoding a BIN 1 -Associated Ul-specific protein (Bau) or a fragment thereof according to claim 1 and a detectable label which is associated with said sequence.
10. An anti-BINl-Associated Ul-specific protein (Bau) antibody.
11. The anti-Bau antibody according to claim 10 directed against a Bau peptide, said peptide selected from the group consisting of:
(a) SEQ ID NO:2; and
(b) a fragment of (a) comprising about 8 amino acids.
12. The antibody according to claim 10, selected from the group consisting of a chimeric antibody, a humanized antibody, a monoclonal antibody and a polyclonal antibody.
13. An anti-idiotype antibody specific for the antibody of claim 10.
14. A diagnostic reagent comprising the antibody according to claim 10 and a detectable label.
15. A pharmaceutical composition comprising the vector according to claim 7 and a pharmaceutically acceptable carrier.
16. A pharmaceutical composition comprising the anti-idiotype antibody according to claim 13 and a pharmaceutically acceptable carrier.
