Abstract

Ein gentechnologisches Verfahren, welches Insertionen von 4 bis 11 Codons in klonierte Gene an beliebiger Stelle erlaubt, wird beschrieben. In Genen, welche für Proteine codieren, wird der Leseraster durch eine Insertion nicht verändert. Vielmehr modifizieren die Insertionen die Proteinstruktur, wodurch Proteine eine veränderte biologische Aktivität erhalten können. Der Transpositionsvorgang des Verfahrens kann in vivo, beispielsweise in Escherichia coli oder in vitro stattfinden. Die Methode kann auch zur Insertion von Epitopen dienen.
