Abstract

Parmi les nouveaux activateurs du plasminogène décrits, l'un est une urokinase qui est modifiée pour la rendre susceptible de former deux urokinases en chaîne et d'augmenter la spécificité de l'urokinase vis-à-vis de la fibrine. Parmis ces nouveaux activateurs du plasminogène, on trouve également des protéines fibrinolytiques constituées d'au moins un domaine capable d'une interaction avec la fibrine et le domaine de protéase de l'urokinase. Ce domaine capable d'une interaction avec la fibrine peut présenter une séquence amino-acide sensiblement identique à celles des domaines kringle-2 de t-PA, kringle-1 et/ou -4 du plasminogène et digital de t-PA. Le domaine de protéase peut présenter une séquence amino-acide sensiblement identique à celle d'une urokinase de 30 Kd. Sont également décrits et revendiqués des séquences d'ADN codant les nouveaux activateurs du plasminogène, des vecteurs d'expression ainsi que des systèmes d'expression qui leur sont destinés.
