Description

更加具體而言,本發明係有關多晶型形式(多晶型形式I和多晶型形式II),以及製備此等多晶型的方法,含有此等多晶型的組合物,和此等多晶型的用途。

因此,自4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸這種化合物由Pfizer在2006年公開(WO2006/090224)以來,人們一直致力於尋找或製備4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸的晶體或晶體形式。例如,諸如乙酸乙酯之類的酯類,諸如甲醇、乙醇和異丙醇之類的醇類,諸如乙腈之類的腈類,諸如二乙醚、MTBE(甲基叔丁基醚)之類的醚類,諸如丙酮和甲基乙基酮之類的酮類,諸如二氯甲烷和氯仿之類的鹵代烷烴被用作再結晶溶劑,但是上述此等溶劑均導致失敗。
儘管作出這麼大的努力,但是還未識別4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸這種化合物的藥學上合適的晶體形式。
如前所述,當以本領域技術人員常用的方式使用乙酸乙酯作為再結晶溶劑時,僅僅得到不成功的結果。在徹底和仔細研究後,本發明的發明人設法找出了使用乙酸乙酯製備晶體形式的非常特殊的且獨一無二的條件,該條件可提供上述化合物的非常期望的晶體形式(多晶型形式I)。
如本發明的操作實施例中所公開的,將所述化合物的白色固體懸浮於乙酸乙酯中在40℃下持續1天並且在室溫下持續5天,從而得到多晶型形式I。本領域技術人員從未想到過這樣的結晶條件。
另一晶體(多晶型形式II)也已從在特殊條件下獲得的多晶型形式I中獲得。多晶型形式I在110℃或更高溫度條件下轉化為多晶型形式II,但是當將多晶型形式II冷卻至室溫時,得到的多晶型形式II在諸如氮氣流之類的測量條件下轉變為多晶型形式I。本發明的發明人還發現在室溫下獲得多晶型形式II的條件。
此外,一旦獲得晶體形式的晶種,那麼相同晶體形式可在小規模合成中容易地獲得。在大規模合成中,溫度控制對於製備藥學上合適的晶體形式而言是必要的。
因此,本發明的目的在於提供4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸的藥學上合適的晶體形式,該晶體形式可容易地,經濟地並且可再生地製備成用於具有諸如與穩定性和生物利用度相關的特徵之類的始終如一的性能特徵的藥物劑型中。本發明的目的還在於提供製備此等多晶型形式的方法,含有此等多晶型形式的組合物以及此等多晶型形式的用途。

(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶型形式II之方法,係包括將4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸置於110℃或更高溫度之條件下之步驟。
目前已出乎意料地發現,本發明的目的已被本發明實現,本發明的目的在於提供4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸的晶體多晶型形式(稱為多晶型形式I和多晶型形式II)以及製備每個晶體的多晶型形式的富有創造性的方法。發現多晶型形式I和多晶型形式II比現有技術WO2006/090224中公開的白色固體更加穩定。此外,根據吸濕性,多晶型形式I和多晶型形式II這兩者具有超過現有技術WO2006/090224中公開的白色固體的明顯優勢。而且,發現本發明的多晶型形式可用於大規模合成。多晶型形式I和多晶型形式II這兩者為無水晶體形式,並且具有用於固體劑型研發的可接受的固態性質。

下面的實施例僅供參考。
PXRD分析由使用Cu-K α放射線的Rigaku RINT-TTR X射線粉末繞射儀進行。樣品還可經由使用不同溫度樣品架的附件在高溫度/低溫度條件下測量。儀器裝配有細焦X-射線管。管電壓和電流分別設置為50kV和300mA。發散和散射狹縫設置為0.25°,並且接收狹縫設置為0.15mm。繞射的放射線經由NaI閃爍探測器檢測。以4°/min(步長0.02°)的速度從32 θ °至40 2 θ °使用θ-2 θ連續掃描。分析矽標準品以檢查機器準線。收集資料並使用Rigaku X-射線系統進行分析。經由將樣品置於鋁樣品架中來製備用於分析的樣品,在資料獲取過程中所述鋁樣品架以60rpm的速度水平旋轉。

DSC分析使用Seiko DSC 6200R或Mettler Toledo

酸多晶型形式I的製備

將4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]-甲基}-四氫-2H-吡喃-4-羧酸(1.326 kg,2.807 mol,白色固體)的乙酸乙酯的漿狀物(18.564L)在70℃下溶解。在35分鐘內將溶液冷卻至64℃,並將200mg多晶型形式I的晶種(0.423mmol)加至混合物中。在5小時內將混合物冷卻至40℃並在該溫度下攪拌14.5小時。在6小時內該漿狀物逐漸冷卻至19℃並且在該溫度下攪拌混合物持續46小時。通過過濾收集形成的沉澱,並且用2.0L乙酸乙酯洗滌濾餅。在50℃,減壓條件下乾燥濾餅,從而得到1.140kg(86%)期望的4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸的晶體形式。

多晶型形式I在約110℃下開始轉變為多晶型形式
II。

在經由動態蒸汽吸附(DVS)分析的吸濕性研究中,多晶型形式I在90%相對濕度(RH),25℃條件下吸收少於0.2wt%。另一方面,現有技術WO2006/090224中公開的白
色固體在85%RH和90%RH條件下分別吸收1.2wt%和5.5wt%的水。下表1表示多晶型形式I和現有技術WO2006/090224中公開的白色固體的增重%。
固態穩定性研究使用Nagano Science Constant溫度/濕度控制腔室LH-20-11M,LH-21-11M,LTL-200D3CJ-14或LTX-01進行。將樣品置於腔室中並且暴露在25℃/60% RH,40℃/75% RH條件下和/或用氙燈照射條件下。在暴露或照射之後得到的樣品的晶體形式,熱行為,純度和/或重量的變化分別經由XRPD,TG/DTA或DSC,H專利文獻C,微量天平分析來評估。
發現多晶型形式是穩定的。

Claims (12)
- 一種4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶型形式I,其特徵為由Cu-K α放射線照射所獲得的粉末X-射線繞射圖譜(PXRD)包括2-θ °5.9,9.3,9.8,11.9,13.7,14.3,15.0,17.8,18.2-19.3,19.7,22.6,23.4-24.5和24.9 +/- 0.2位置的主峰。
- 如申請專利範圍第1項所述之4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶型形式I,其中,進一步於差示掃描量熱分析(DSC)中,在169℃展現吸熱現象。
- 如申請專利範圍第1或2項所述之4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶型形式I,其中,紅外線(IR)光譜(KBr)顯示出2948,1723,1615,1535,1506,1437,1383,1366,1287,1262,1245,1180,1164,1120,1095,1059,1032,992,974,935,918,869,858,828,784,746,732,654和556 +/- 2cm-1位置的吸收譜帶。
- 一種4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶型形式II,其特徵為由Cu-K α放射線照射所獲得的粉末X-射線繞射圖譜(PXRD)包括2-θ °5.8, 9.7,10.5,11.8,12.4,13.5,14.2,14.6-14.9,15.4,17.8,18.2,19.9-20.5,21.2,21.8,23.6,24.1和24.6 +/- 0.2位置的主峰。

- 如申請專利範圍第4或5項所述之4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶型形式II,其中,紅外線(IR)光譜(KBr)顯示出2950,1724,1614,1534,1507,1438,1383,1366,1287,1262,1245,1180,1164,1121,1095,1059,1031,992,974,935,918,869,857,828,784,746,732,654和555 +/- 2cm-1位置的吸收譜帶。
- 一種藥物組合物,係包含申請專利範圍第1至6項中任一項所述之4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶型形式以及一種或一種以上藥學上可接受的賦形劑。
- 一種申請專利範圍第1至6項中任一項所述之4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶 型形式之用途,係用於作為藥物。
- 一種申請專利範圍第1至6項中任一項所述之4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶型形式或申請專利範圍第7項所述之藥物組合物之用途,係用在製備用於治癒性治療、緩解性治療或預防性治療由5-HT4受體活性媒介的疾病病症的藥物。
- 一種製備申請專利範圍第1至3項中任一項所述之4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶型形式I之方法,係包括將4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸的白色固體懸浮於乙酸乙酯中在40℃下1天並且在室溫下5天之步驟。
- 一種製備多晶型形式I之方法,係包括在包含乙酸乙酯的有機溶劑中,於4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸添加申請專利範圍第1至3項中任一項所述之多晶型形式I之晶種,並結晶化。
- 一種製備申請專利範圍第1至3項中任一項所述之4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2-苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶型形式I之方法,係包括將申請專利範圍第4至6項中任一項所述之4-{[4-({[4-(2,2,2-三氟乙氧基)-1,2- 苯並異唑-3-基]氧基}甲基)哌啶-1-基]甲基}-四氫-2H-吡喃-4-羧酸多晶型形式II置於氮氣下之冷卻步驟。
