Claims (26)
1. Лекарственная форма замедленного высвобождения, в состав которой входят:
a) инертное ядро,
b) первый слой в виде сахарной матрицы замедленного высвобождения, обволакивающий собой инертное ядро, где в состав слоя сахарной матрицы входят будесонид, водонерастворимый полимер и сахар, причем отношение сахара к водонерастворимому полимеру более 3:1.
c) пленочное покрытие отсроченного высвобождения, обволакивающее собой слой сахарной матрицы замедленного высвобождения.
2. Лекарственная форма по п. 1, отличающаяся тем, что в качестве водонерастворимого полимера использована этилцеллюлоза.
3. Лекарственная форма по п. 1, отличающаяся тем, что слой сахарной матрицы содержит сахар в количестве от примерно 50 до 80% от общей массы слоя сахарной матрицы.
4. Лекарственная форма по п. 1, отличающаяся тем, что водонерастворимый полимер присутствует в количестве от примерно 5 до 15% от общей массы слоя сахарной матрицы.
5. Лекарственная форма по п. 1, отличающаяся тем, что пленочное покрытие отсроченного высвобождения содержит сополимеры акриловой кислоты и сложных эфиров метакриловой кислоты.
6. Лекарственная форма по п. 1, отличающаяся тем, что количество будесонида составляет от 6 мг до 9 мг.
7. Лекарственная форма по п. 1, отличающаяся тем, что будесонид присутствует в количестве от 15% до 25% от общей массы слоя сахарной матрицы.
8. Лекарственная форма по п. 1, отличающаяся тем, что будесонид присутствует в количестве от 1,5% до 3,5% от общей массы лекарственной формы замедленного высвобождения.
9. Лекарственная форма замедленного высвобождения в виде капсулы, саше или пакета, содержащая по 9 мг будесонида на капсулу, саше или пакет, которая, при испытаниях на растворимость в Аппарате 2 USP при скорости вращения 75 об/мин в 1000 мл 0.1N HCl в течение 2 часов, с последующей заменой на буфер с рН 5,5 показывает следующий профиль растворимости:
a) менее 5% от общего количества будесонида высвобождается через 2 часа
b) 5% - 15% от общего количества будесонида высвобождается через 3 часа
c) 15% - 20% от общего количества будесонида высвобождается через 4 часа
d) 30% - 40% от общего количества будесонида высвобождается через 6 часов
e) 40% - 50% от общего количества будесонида высвобождается через 8 часов
f) не менее 50% от общего количества будесонида высвобождается через 10 часов, и,
при испытаниях на растворимость в Аппарате 2 USP при скорости вращения 75 об/мин в 1000 мл 0,1N HCl в течение 2 часов, с последующей заменой на буфер с рН 7,5, показывает следующий профиль растворимости:
a) менее 5% от общего количества будесонида высвобождается через 2 часа
b) 30% - 50% от общего количества будесонида высвобождается через 3 часа
c) 40% - 60% от общего количества будесонида высвобождается через 4 часа
d) 65% - 85% от общего количества будесонида высвобождается через 6 часов
e) не менее 80% от общего количества будесонида высвобождается через 8 часов.
10. Применение лекарственной формы по п. 1 или 9 для лечения болезни Крона.
11. Применение лекарственной формы по п. 1 или 9 для поддержания клинической ремиссии болезни Крона средней и легкой степени тяжести, затрагивающей подвздошную кишку и/или восходящую ободочную кишку.
