Description

신규 형태의 연질캡슐 제조방법
본 발명은 수용성 의약물질을 함유하는 필림으로 오일성 물질을 감싸는 신규 형태의 연질 캡슐에 관한 것이다.
따라서 본 발명의 목적은 수용성 의약물질에 적용할 수 있는 신규 형태의 연질 캡슐을 제공하는 것이다.
의약물질을 함유하는 오일성 물질을 젤라틴 캡슐로 감싸서 연질캡슐을 제조하는 것은 공지이다.
의약물질이 수용성인 경우에는, 연질캡슐은 수용성 의약물질의 오일 현탁액을 젤라틴 필림으로 감싸서 제조하여 왔다.
그러나, 이 방법에 의하면 제조 공정중에 각 캡슐에 충진되는 현탁액이 일정하게 유지하기가 대단히 어렵기 때문에, 각 캡슐중의 의약물질의 양을 일정하게 함유하게 하기가 어려움으로 캡슐의 질이 저하하는 것은 필수적이었다. 이러한 경향은 의약물질의 함량의 저하에 의하여 점점 더욱 현저해져서, 때로는 의약품의 질에 치명적인 결과를 초래하는 경우도 있었다.
본 발명자들은 이러한 종류의 문제점을 해결하기 위하여 오랜 연구를 행하였으며, 드디어 수용성 의약물질을 필림에 함유하게 하는 생각을 가지게 되었다. 이러한 생각은 의약물질을 연질캡슐의 오일상에 함유하게 하는 통상의 지식을 타파함으로서 얻어질 수 있었다.
본 발명의 연질캡슐은 약전에 정의된 통상의 연질캡슐과는 다른 구조를 가진다. 따라서 본 발명의 연질캡슐은 본 명세서에서 "신규형태의 연질캡슐"이라고 부른다.
본 발명의 신규 형태의 연질캡슐은 수용성 의약물질을 함유하는 필림으로 오일성 물질을 감싸서 제조한다.
필림의 극성분은 제한되어 있지 않으나, 통상으로 필림은 임의로 유기 또는 무기산 또는 알칼리성 물질로 처리한 젤라틴으로 구성된다.
착색제, 가소제, 방부제, 습윤제, 용해보조제, 붕해제, 향미제, 항산화제 등과 같은 필림에 통상으로 사용하는 첨가제를 본 발명의 필림 성분에 임의로 가할 수 있다.
본 발명은 필림 성분에 용해될 수 있는 수용성 의약물질에 적용할 수 있다. 의약물질이 필림 성분에 쉽게 용해하지 않는 경우에는 용해보조제를 임의로 사용할 수 있다.
따라서, 본 발명의 수용성 의약물질은 물에 쉽게 용해되는 것뿐만 아니라, 용해보조제를 사용함으로서 물에 쉽게 용해될 수 있는 것까지도 포함한다. 필림에 함유된 의약물질의 양은 의약물질이 필림 성분에 용해될 수 있는 범위 내에서 의약의 용량에 따라서 선택할 수 있다.
전술한 바와 같이 필림에 의하여 감싸지는 오일성 물질은 식물유(예 : 올리브유, 새플라우어유, 옥수수유, 해바라기유, 면실유, 동백유, 미강유, 대두유, 호마유, 밀배아유, 낙화생유, 코코닛유, 평지유 등), 어유(예 : 오징어유, 대구유 등), 간유(상어간유, 대구간유 등), 바다짐승(예 : 강치유, 청색 고래유 등), 조개유(예 : 전복유, 굴유 등), 이들 동식물유의 혼합물, 오일상 약학적 물질(예 : 간유, 피마자유, 지방산 글리세라이드, 비타민E, 비타민A, 비타민K 등), 폴리에틸렌글리콜 등의 통상의 것이다. 이들 통상의 오일상 물질은 임의로 그 안에 용해될 수 있는 미약 물질을 함유할 수 있다.
본 발명의 신규형태의 연질캡슐은 통상의 방법, 특히 일본공개특허공고 제1067/1978호에 기술된 방법으로 제조할 수 있으며, 이 방법으로 필림의 중량이 일정한 신규 형태의 연질캡슐을 쉽게 제조할 수 있다.
본 발명의 신규 형태의 연질캡슐은 다음과 같은 우수한 효과를 가진다. 즉, 각 캡슐내의 의약물질의 함량은 의약물질이 수용성이지만 쉽게 균일하게 할 수 있다. 더구나, 의약물질이 연질캡슐의 표면부에 함유되어 있기 때문에 의약물질은 투약후 짧은 시간 내에 용해될 수 있다.
상기의 효과 이외에도, 오일상 물질과 필림이 투명한 경우에는 의약물질이 오일상 물질에 현탁되지 않고, 필림 성분에 용해되기 때문에 이러한 연질캡슐이 얻어질 수 있다.
본 발명의 신규 형태의 연질캡슐을 제조하는 방법은 다음의 실시예로서 더욱 상세히 설명한다.

D-솔비톨 1mg
글리세롤 2mg

프로필파라벤 0.001mg
홍색 착색제 적당량
오일상 물질
트리캐프릴산 글리세라이드 23mg
전술한 비율로 필림 성분과 오일상 물질을 사용하는 일본특허공개공고 제1067/1978호 명세서에 기술된 방법으로 직경 4mm의 신규 형태의 연질캡슐을 제조한다.

실시예 1에서 트리캐프릴산 글리세라이드 대신에 호마유를 사용하여 일본공개특허공고 제1067/1978호의 명세서에 기술된 방법으로 신규 형태의 연질캡슐을 제조한다.
[실시예 3]
실시예 1에서 소디움 피코설페이트 대신에 말레인산 클로로페닐아민(2mg)을 사용하여 일본공개특허공고 제1067/1978호의 명세서에 기술된 방법으로 신규 형태의 연질캡슐을 제조한다.
