Claims (26)
- 체액을 각 혈구 성분으로 분리하는 방법으로서,
(a) 체액을 혈구 분리재와 접촉시킴으로써 백혈구와 혈소판을 혈구 분리재로 포착하여,
적혈구 풍부 분획을 얻는 공정과,
(b) 분리 용액을 사용하여 혈구 분리재로부터 백혈구 풍부 분획을 분리하는 공정
을 포함하는 것을 특징으로 하는 분리 방법. - 제1항에 있어서,
혈구 분리재가 체액의 입구와 출구를 구비한 용기에 충전되어, 체액을 당해 입구로부터 통액(通液)함으로써 혈구 분리재와 접촉시키는 것을 특징으로 하는 분리 방법. - 제1항 또는 제2항에 있어서,
(a) 공정에 있어서, 체액을 혈구 분리재와 접촉시킨 후에, 혈구 분리재에 잔존하는 적혈구를 세정액으로 세정하는 분리 방법. - 제2항 또는 제3항에 있어서,
(b) 공정에 있어서, 체액의 출구측으로부터 분리 용액을 주입하여 백혈구를 분리 회수하는 것을 특징으로 하는 분리 방법. - 제1항 내지 제4항 중 어느 한 항에 있어서,
분리 용액이 생리 식염수, 완충액, 덱스트란, 배지, 또는 수액을 함유하는 것을 특징으로 하는 분리 방법. - 제1항 내지 제5항 중 어느 한 항에 있어서,
체액이 혈액, 골수, 제대혈, 월경혈, 또는 조직 추출물인 분리 방법. - 제1항 내지 제6항 중 어느 한 항에 있어서,
(a) 공정의 체액을 혈구 분리재와 접촉시키기 전에, 미리 용액을 혈구 분리재와 접촉시키는 것을 특징으로 하는 분리 방법. - 제7항에 있어서,
미리 혈구 분리재와 접촉시키는 용액이 생리 식염수, 또는 완충액을 함유하는 것을 특징으로 하는 분리 방법. - 제1항 내지 제8항 중 어느 한 항에 있어서,
혈액 분리재가 부직포로 이루어지는 분리 방법. - 제9항에 있어서,
부직포가 폴리에스테르 섬유, 폴리프로필렌 섬유, 나일론 섬유, 또는 아크릴 섬유로 구성되는 분리 방법. - 제10항에 있어서,
폴리에스테르 섬유가 폴리에틸렌테레프탈레이트 섬유, 또는 폴리부틸렌테레프탈레이트 섬유인 분리 방법. - 제10항에 있어서,
부직포가 나일론 섬유, 폴리프로필렌 섬유, 또는 폴리부틸렌테레프탈레이트 섬유로 구성되는 분리 방법. - 제9항에 있어서,
부직포가 분할 섬유로 구성되는 분리 방법. - 제9항 내지 제13항 중 어느 한 항에 있어서,
부직포의 밀도 K가 2.0×104 이상 1.9×105 이하, 섬유경이 1㎛ 이상 15㎛ 이하이며, 백혈구 풍부 분획이 단핵구 풍부 분획인 분리 방법. - 제1항 내지 제14항 중 어느 한 항에 있어서,
분리 용액으로서, 점도가 1mPa?s 이상 5mPa?s 미만의 용액을 사용하는 분리 방법. - 제1항 내지 제15항 중 어느 한 항에 있어서,
백혈구 풍부 분획 중 또는 단핵구 풍부 분획 중의 혈소판 회수율이 백혈구 회수율 이하인 것을 특징으로 하는 분리 방법. - 제1항 내지 제15항 중 어느 한 항에 있어서,
백혈구 풍부 분획 중 또는 단핵구 풍부 분획 중의 혈소판 회수율이 단핵구 회수율 이하인 것을 특징으로 하는 분리 방법. - 제1항 내지 제15항 중 어느 한 항에 있어서,
백혈구 풍부 분획 중 또는 단핵구 풍부 분획 중의, 단핵구 회수율의 과립구 회수율에 대한 비가 1.0보다 큰 것을 특징으로 하는 분리 방법. - 제1항 내지 제15항 중 어느 한 항에 있어서,
백혈구 풍부 분획 또는 단핵구 풍부 분획이, 조혈 간세포(幹細胞), 간엽계 간세포, 또는 CD34 양성 세포를 포함하는 것을 특징으로 하는 분리 방법. - 제1항 내지 제13항 중 어느 한 항에 기재된 방법에 의해 분리된 적혈구 풍부 분획, 백혈구 풍부 분획, 또는 혈소판 풍부 분획.
- 밀도 K가 2.0×104 이상 1.9×105 이하, 섬유경이 1㎛ 이상 15㎛ 이하인, 체액으로부터 단핵구 풍부 분획을 분리 가능한 부직포로 이루어지는 혈구 분리재.
- 제21항에 있어서,
부직포가 나일론 섬유, 폴리프로필렌 섬유, 또는 폴리부틸렌테레프탈레이트 섬유로 구성되는 혈구 분리재. - 제21항 또는 제22항에 기재된 혈구 분리재가 체액의 입구와 출구를 구비한 용기에 적층 상태로 충전되어 있는 것을 특징으로 하는 혈구 분리 디바이스.
- 제23항에 있어서,
상기 용기의 입구측의 상류에 설치된 유로 개폐 수단,
유로 개폐 수단에 연결된 백혈구 풍부 분획 회수 수단, 및
상기 용기의 출구측의 하류에 설치된 분리 용액의 도입 수단
을 더 갖는 혈구 분리 디바이스. - 혈구 분리재 또는 부직포에 포착된 백혈구를 회수하기 위한 용액의 점도가 1mPa?s 이상 5mPa?s 미만인 분리 용액.
- 제25항에 있어서,
덱스트란을 함유하지 않는 것을 특징으로 하는 분리 용액.
