When the dosage form of the present invention is a suspension, liquid carrier diluents such as water, ethanol or propylene glycol, suspending agents such as ethoxylated isostearyl alcohol, polyoxyethylene sorbitol ester and polyoxyethylene sorbitan ester, microcrystalline Cellulose, aluminum metahydroxy, bentonite, agar or tracant and the like can be used.

When the formulation of the present invention is a surfactant-containing cleansing, the carrier component is an aliphatic alcohol sulfate, an aliphatic alcohol ether sulfate, a sulfosuccinic acid monoester, an isethionate, an imidazolinium derivative, a methyltaurate, a sarcosinate, a fatty acid amide. Ether sulfates, alkylamidobetaines, aliphatic alcohols, fatty acid glycerides, fatty acid diethanolamides, vegetable oils, linolin derivatives or ethoxylated glycerol fatty acid esters and the like can be used.

Joiner extract of the present invention exhibits an antimicrobial activity against the acne bacteria P. acnes can be usefully used as an external skin pharmaceutical composition and cosmetic composition for the improvement and treatment of acne.

Hereinafter, the present invention will be described in detail by the following Examples and Experimental Examples.

However, the following Examples and Experimental Examples are only illustrative of the present invention, the contents of the present invention is not limited by the following Examples, Reference Examples and Experimental Examples.

Example 1 Preparation of Joiner Extract

The seedling extract was mixed with 1 L of methanol (Methanol) in 200 g of the dried amount of the seedling (purchased by a pharmacist), and then extracted by stirring three times at room temperature for 72 hours. The extract was filtered with filter paper (Whatman, No. 2), concentrated under reduced pressure with a rotary vacuum evaporator (SB-1000, Japan) at 60 ± 2 ° C., and lyophilized to obtain 5.11 g of a seedling extract. The obtained joiner extract was dissolved in DMSO (dimethyl sulfoxide), filtered through a 0.2 μM filter (Millipore Corporation, Bedford, MA) and used as a sample of the following experimental example.

Reference Example  One. Experimental strain  And culture

P. acnes KCCM 41747 received from Korea Microbial Conservation Center was incubated in BHI culture (Difco, USA) and then mixed with glycerol (glycerol) in a ratio of 3: 7 and stored at -70 ℃. Strains stored in the freezer were activated 3 days before the start of the experiment and inoculated in a BHI agar plate (incubated in an anaerobic incubator supplied with 5% H ₂ and 95% N ₂ were used in the experiment.

Experimental Example 1. Measurement of the growth inhibitory effect of acne causative bacteria

1-1. Minimum Inhibition Concentration (MIC) Measurement by Broth Dilution Test

In order to measure the minimum inhibitory concentration (MIC) that inhibits the growth of P. acnes , which is the acne bacterium, by using the method described in the literature, experiments were performed as follows (Lee Sung-rim et al., Korea Environmental Health) Journal, 32; 192-197, 2006).

P. acnes bacteria were precultured for 24 hours by the method of Reference Example 1. The cultured bacteria were measured for absorbance at 600 nm using a spectrophotometer (TECAN, Crailsheim, Germany), prepared at a concentration of 1 × 10 ⁷ CFU / ml and 200uL was dispensed in 96-well plate (96well plate). Joiner extract was diluted to 0.5 ug / mL by diluting stepwise 2-fold dilution with the highest concentration of 500 ug / mL. Diluted joiner extracts were added to 96 well plates inoculated with test strains and cultured under anaerobic culture conditions (5% H ₂ + 95% N ₂ , 37 ° C.). After 48 hours of incubation, the absorbance was measured at 600 nm with a spectrophotometer, and the lowest concentration at which growth of acne was suppressed was determined as the minimum inhibitory concentration, which is shown in Table 1 below.

As shown in Table 1, the minimum inhibitory concentration for acne bacteria of the joiner extract was 62.5ug / mL.

1-2. Measurement of clear zone by disk diffusion test

In order to determine the clear zone of P. acnes acne bacteria by the joiner extract, the experiment was conducted using the disk method (Chomnawang et al., Journal of Ethnopharmacology, 101; 330-333, 2005).

P. acnes bacteria were pre-incubated for 24 hours using a BHI agar plate and prepared at a concentration of 1 × 10 ⁸ CFU / ml and maintained in 5 mL of top agar maintained at 45 ° C. 1 mL of each was added and mixed well, and then smeared onto a BHI agar plate. Joiner extracts were prepared at concentrations of 0.25, 0.5 and 1 mg / 25 uL, loaded 25uL each on 8mm diameter paper disks and 25uL DMSO on the control disk. The top agar was placed on a dry BHI agar plate, and the disks with the seedling extracts were raised and incubated for 48 hours under anaerobic conditions. Growth inhibition produced after 48 hours was measured using a micrometer (Mitutoyo, Kawasaki, Japan).

As a result, as shown in Figure 1, DMSO-only control showed no growth inhibitory effect on acne bacteria, whereas the seedling extract showed growth inhibition effect in a concentration-dependent manner. Therefore, it was confirmed that the extract of the joiner had an antimicrobial effect against P. acnes (propionibacterium acnes) , which is the causative agent of acne (see Fig. 1).

Thus, the preparation examples using the joiner extract below, but this is intended to explain in detail only, not intended to limit the present invention.

Claims (6)

A skin external pharmaceutical composition for improving and treating acne, comprising as an active ingredient a joiner extract. The method of claim 1, wherein the extract is one selected from the group consisting of water, ethanol, methanol, propanol, butanol, acetone, ethyl acetate, hexane, butylene glycol, propylene glycol, hydrobutylene glycol, hydrous propylene glycol, hydrous glycerin Skin external pharmaceutical composition which is an extract soluble in the above solvents.
