Claims (23)
- 칼루스 또는 현탁 배양물 유래의 탁수스종 세포를 성장 및 산물 형성 조건 하에서 성장 및 탁산 생산에 대해 영양 배지가 동일하거나 상이한 하나 이상의 영양 배지 중에서 현탁 배양으로 배양하는 단계; 및 상기 세포, 세포 배양 배지, 또는 그 둘다로부터 1종 이상의 탁산을 회수하는 단계를 포함하며, 여기서, 하나 이상의 영양 배지는 은 함유 화합물, 은 착물 또는 은 이온의 형태로 10nM 내지 900μM의 농도로 은을 함유하고, 하나 이상의 영양 배지 중 하나 이상이, 증강제로서 옥신 관련 성장 조절제를 증강제 대 은의 몰비가 0.011 내지 1000이 되도록 하는 양으로 함유하는 것인, 탁수스종의 세포 배양에서 탁산을 고수율로 생산하는 방법.
- 제1항에 있어서, 하나 이상의 영양 배지가 탁산 전구체를 추가로 함유하는 방법.
- 제2항에 있어서, 탁산 전구체가 α-페닐알라닌, β-페닐알라닌 또는 그 혼합물인 방법.
- 제1항에 있어서, 하나 이상의 영양 배지가 글루타민을 추가로 함유하는 방법.
- 제1항에 있어서, 하나 이상의 영양 배지가 글루탐산, 아스파르트산 또는 이들의 혼합물을 추가로 함유하는 방법.
- 제1항에 있어서, 하나 이상의 영양 배지가 탄소원으로서 말토스를 함유하는 방법.
- 제1항에 있어서, 하나 이상의 영양 배지가 탄소원으로서 슈크로스를 함유하는 방법.
- 제1항에 있어서, 하나 이상의 영양 배지가 탄소원으로서 글루코스, 프럭토스 또는 이들의 혼합물을 함유하는 방법.
- 제1항에 있어서, 하나 이상의 영양 배지가 말토스, 슈크로스, 글루코스, 프럭토스 또는 이들의 혼합물을 1차 탄소원으로 함유하는 방법.
- 제1항에 있어서, 영양 배지가 세포 성장과 탁산 생산에 대해 동일한 것인 방법.
- 제1항에 있어서, 세포 성장을 위한 영양 배지의 조성과 탁산 생산을 위한 영양 배지의 조성이 상이한 방법.
- 제11항에 있어서, 탁산 생산중 1회 이상의 영양 배지 교환을 추가로 포함하는 방법.
- 제1항에 있어서, 현탁 배양 단계중 1회 이상의 영양 배지 교환을 추가로 포함하는 방법.
- 제1항에 있어서, 탁산 생산중 배양물로부터 탁산의 제거 단계를 추가로 포함하는 방법.
- 제1항에 있어서, 상기 탁수스종의 세포를 공급 배치(fed batch) 공정에 의해 배양하는 방법.
- 제1항에 있어서, 탁산의 부피 생산성이 탁산 생산 기간에 걸쳐 평균 15㎎/L/일 이상인 산물 형성 기간을 갖는 방법.
- 제1항에 있어서, 상기 1종 이상의 탁산이 탁솔을 포함하고, 상기 세포, 세포 배양 배지, 또는 그 둘다로부터 탁솔을 회수하는 것인 방법.
- 제17항에 있어서, 상기 1종 이상의 탁산이 탁솔을 포함하고, 탁솔 생산 기간 에 걸쳐 평균한 탁솔의 부피 생산성이 10㎎/L/일 이상인 방법.
- 제1항에 있어서, 상기 1종 이상의 탁산이 바카틴 III을 포함하고, 상기 세포, 세포 배양 배지, 또는 그 둘다로부터 바카틴 III을 회수하는 것인 방법.
- 제19항에 있어서, 상기 1종 이상의 탁산이 바카틴 III을 포함하고, 탁산 생산 기간에 걸쳐 평균한 바카틴 III의 부피 생산성이 15㎎/L/일 이상인 방법.
- 제1항에 있어서, 탁수스종이 탁수스 브레비폴리아(T. brevifolia), 탁수스 카나덴시스(T. canadensis), 탁수스 키넨시스(T. chinensis), 탁수스 쿠스피다타(T. cuspidata), 탁수스 바카타(T. baccata), 탁수스 글로보사(T. globosa), 탁수스 플로리다나(T. floridana), 탁수스 왈리치아나(T. wallichiana) 또는 탁수스 메디아(T. media)인 방법.
- 제1항에 있어서, 상기 1종 이상의 탁산이 탁솔을 포함하고, 상기 탁수스종 세포가 증강제를 함유하지 않는 배지에서의 칼루스 배양 또는 현탁 배양시 ELISA로 검출가능한 양의 탁솔을 생산하는 것인 방법.
- 제1항에 있어서, 탁수스종 세포가 티오황산은, 메틸 자스모네이트 및 1-나프탈렌아세트산을 함유하는 배지에서 현탁 배양시 10㎎/L/일의 평균 부피 생산성으로 탁산을 생산하는 것인 방법.
