Claims (10)
- 단백질 및 히알루론산이 포함된 천연 히알루론산 혼합물로부터 고분자 히알루론산을 정제하는 방법에 있어서,상기 히알루론산 혼합물 내의 단백질을 가수분해한 후, 가수분해된 단백질과 히알루론산을 분리하기 위하여 고속 원심 분리하는 제1단계; 및분리된 히알루론산에서 저분자 히알루론산과 고분자 히알루론산을 분리하기 위하여 고속 원심 분리하는 제2단계;를 포함하여 이루어지는 것을 특징으로 하는 고분자 히알루론산의 정제 방법.
- 제1항에 있어서,상기 가수분해된 단백질과 히알루론산을 분리하기 위하여 고속 원심 분리하는 제1단계에서는 매질로 수분 염류 용액(1)을 사용하며,사용되는 수분 염류 용액(1)의 밀도(ρ1)는, 고분자 히알루론산의 밀도(ρL) 및 저분자 히알루론산의 밀도(ρS)보다는 크고, 가수분해된 단백질의 밀도(ρP)보다는 작은 것을 특징으로 하는고분자 히알루론산의 정제 방법.
- 제1항에 있어서,상기 저분자 히알루론산과 고분자 히알루론산을 분리하기 위하여 고속 원심 분리하는 제2단계에서는 매질로 수분 염류 용액(2)을 사용하며,사용되는 수분 염류 용액(2)의 밀도(ρ2)는, 고분자 히알루론산의 밀도(ρL)보다는 크고, 저분자 히알루론산의 밀도(ρS)보다는 작은 것을 특징으로 하는고분자 히알루론산의 정제 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서,상기 저분자 히알루론산과 고분자 히알루론산을 분리하기 위하여 고속 원심 분리하는 제2단계에 있어서,고분자 히알루론산과 저분자 히알루론산의 분리를 촉진하기 위하여, 고속 원심 분리시 초음파를 발생시키는 것을 특징으로 하는고분자 히알루론산의 정제 방법.
- 제4항에 있어서,상기 초음파의 파장은 저분자 히알루론산의 평균 선형 길이보다는 크고, 고분자 히알루론산의 평균 선형 길이보다는 작은 것을 특징으로 하는고분자 히알루론산의 정제 방법.
- 제1항에 있어서,제1단계에서의 단백질의 가수분해를 위하여 파파인 용액을 사용한 효소 가수분해를 행하는 것을 특징으로 하는고분자 히알루론산의 정제 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서,제1단계 및 제2단계에서 행하여지는 고속 원심 분리의 가속도가,a = ΩR(식중, a는 가속도, Ω는 고속 회전 각속도 및 R은 고속 원심 분리기의 반경을 나타냄)이고,ΩR 》g(식중, g는 9.8m/sec2의 자유 낙하 가속도를 나타냄)의 조건을 만족하는 것을 특징으로 하는고분자 히알루론산의 정제 방법.
- 제1항에 있어서,상기 저분자 히알루론산과 고분자 히알루론산을 분리하기 위하여 고속 원심 분리하는 제2단계 이후,고분자 히알루론산을 증대하기 위하여, 잔류하는 저분자 히알루론산에 대하여 고속 원심 분리를 부가적으로 행하는 제3단계를 포함하는 것을 특징으로 하는고분자 히알루론산의 정제 방법.
- 제8항에 있어서,잔류하는 저분자 히알루론산에 대하여 고속 원심 분리를 부가적으로 행하는 제3단계에 있어서,고분자 히알루론산과 저분자 히알루론산의 분리를 촉진하고 고분자 히알루론산을 증대하기 위하여, 고속 원심 분리시 초음파를 발생시키는 것을 특징으로 하는고분자 히알루론산의 정제 방법.
- 제9항에 있어서,상기 초음파의 파장은 저분자 히알루론산의 평균 선형 길이보다는 크고, 고분자 히알루론산의 평균 선형 길이보다는 작은 것을 특징으로 하는고분자 히알루론산의 정제 방법.
