Abstract

본 발명은 유산균을 함유한 씨드(seed)에 수계코팅기제를 사용하여 저온에서 1 차 코팅을 수행한 다음, 필요에 따라 방출제어형 코팅기제를 사용한 2 차 코팅을 수행함을 특징으로하여 제조된 미세 장용성 코팅과립에 관한 것이다.

Description

유산균의 안정화를 위한 미세 장용성 코팅과립
본 발명은 유산균의 안정화를 극대화시키기 위한 미세 장용성 코팅과립에 관한 것이다. 본 명세서에서 유산균이란 인간의 장내에 기생하여 장의 연동운동이 활발하게 유지되도록 도와주는 유익한 균을 의미한다.
유산균을 섭취하면 음식물의 이상발효가 저해되고 장기능이 활성화되어 변비와 설사 등의 장기능 이상이 개선되며, 섭생활동이 원활하게 유지된다. 또한, 유산균을 가축의 사료에 첨가하면 같은 사료의 반복투여로 인한 가축의 장내 이상발효에 의한 가스의 축적, 변비, 설사 등을 예방함으로써 고기의 육질을 개선할 수 있으므로 낙농업의 발전에도 크게 기여할 수 있다.
이와 같이 효용과 가치면에서 매우 유익한 균임에도 불구하고, 유산균은 산에 상당히 불안정하여 실제 사용에 많은 제약이 따르고 있다. 통상적으로 유산균은 pH 4 이하에서 매우 불안정하여 위산과 같이 낮은 산도(pH 2 정도)에서는 투입된 유산균이 거의 모두 사멸하기 때문이다. 이에 따라, 섭취된 유산균중 생존하여 장에 도달하는 것은 거의 무시될 정도의 소량(약 백만분의 일)에 불과하며, 그 결과 유산균이 장내에서 제대로 효과를 발휘하도록 하기 위해서는 많은 시간과 비용을 필요로 하게 된다.
이러한 점을 극복하고자 식품과 제약업계에서는 유산균을 10배 이상 과량으로 투입함으로써 유산균의 장내 도달을 높이는 방법을 사용하고 있다. 그러나, 이는 지극히 소모적인 미봉책으로서 근본적인 해결책을 제시하지 못하는 단편적인 방법에 불과하다. 또한, 최근에는 유산균을 지방, 유화제 및 보호제와 함께 혼합하여 미세캡슐로 제조함으로써 위산에 견디게 하여 장내 도달율을 높인 식품류가 선보이고 있으나(참조: 대한민국 특허공개 제 97-25405 호), 실험결과 이 캡슐화된 유산균은 그 환경이 위액이건 장액이건 상관없이 30분이내에 코팅이 붕해되는 것으로 확인되었다. 또한, 젤라틴을 코팅기제로 사용하는, 시판되는 코팅유산균은 인공위액과 인공장액에 대한 특이성의 차이없이 어떤 환경에서든 10시간이상 붕해되지 않음을 실험결과로 알 수 있었는데(비교예 1 참조), 이는 코팅유산균이 위, 장의 환경, 즉 액성에 따라 민감하게 반응하지 않는 일반적인 코팅에 불과하기 때문인 것으로 생각된다. 이밖에 일반 의약품 영역에서 각종 고분자를 이용한 유기용매 코팅이 여러 가지 소개되고 있다(참조: PCT/JP94/001675, 일본국 특허출원 평 3-235667, 4-364123, 4-41434, 5-186335, 5-186336). 그러나, 이러한 코팅기술을 그대로 적용하여서는 본 발명에서 목적하는 바와 같이 유산균을 위산으로부터 보호하여 그 효능을 효과적으로 이용하는데 충분치 못한 것으로 생각되었다. 특히, 코팅제제의 용매로서 유기용매를 사용하거나 70℃ 이상의 고온에서 코팅하게되면 유산균의 실제 생존율이 기대에 훨씬 미치지 못하였다.
한편, 균 자체의 내산성을 높인 변이균주의 개발도 시도되고 있으나, 이는 많은 비용과 시간을 필요로 하면서도 코팅방법에 비하여 그 효과가 적다는 문제점을 안고 있다.
이에 본 발명자들은 유산균의 안정성 및 경제적 측면에서 이용가치가 있는 장용성 코팅에 대하여 연구를 진행한 결과, 유산균을 특정의 수계코팅기제로 1 차 코팅하고, 필요에 따라 일반적인 방출제어형 코팅기제를 사용하여 2 차 코팅을 수행하면 코팅과립의 제조과정중에 유산균의 사멸이 격감할 뿐아니라, 위산으로부터 유산균을 안정하게 보호하여 실제로 유산균이 그 기능을 발휘하여야할 장소인 장에까지 무사히 운반될 수 있는 코팅과립을 제조할 수 있음을 발견하고 본 발명을 완성하게 되었다. 즉, 본 발명에 따른 코팅과립은 활성 유산균을 고비율로 함유하며, 산도에 민감하게 반응하므로 과립중에 함유된 유산균은 인간의 위내환경에 견디고 장에서 신속히 붕해될 수 있다.
따라서, 본 발명은 유산균의 안정화를 극대화시켜 장내에서 그의 활성을 나타내도록 특별히 고안된 미세 장용성 코팅과립에 관한 것이다.
본 발명에 따른 유산균 코팅과립에 대하여 보다 구체적으로 설명하면 다음과 같다.
본 발명에 따른 유산균 코팅과립은 유산균을 함유한 씨드(seed)를 수계코팅기제로 저온에서 1 차 코팅한 다음, 필요에 따라 방출제어형 코팅기제를 사용한 2 차 코팅을 수행하여 제조한다. 본 발명에서는 이와 같이 수계코팅기제를 사용하여 저온에서 1차로 수계코팅을 수행함으로써 제조과정중에 유산균이 사멸화되는 것을 최소화할 수 있었다.
본 발명에서 유산균으로는 스트렙토코커스(Streptococcus)속, 락토코커스(Lactococcus)속, 루코노스톡(Leuconostoc)속, 페디오코커스(Pediococcus)속, 엔테로코커스(Enterococcus)속, 락토바실러스(Lactobacillus)속, 비피도박테리움(Bifi- dobacterium) 속의 균주중에서 인체에 유익한 균을 1 종 이상 선택하여 사용할 수 있다.
1 차 코팅시 사용가능한 수계코팅기제로는 미역 등 해조류 추출물의 주성분인 알긴산나트륨, 알긴산, 폴리메틸메타크릴레이트[Eudragit L30D, Eudragit LS30D, Kollicoat MP(BASF 사 제조) 등], 소맥단백, 대두단백, 메틸셀룰로오스(MC), 하이드록시프로필셀룰로오스(HPC), 하이드록시프로필메틸셀룰로오스[HPMC; 파마코트(pharma coat), 아쿠아코트(aqua coat) 등], 검류, 예를들어 구아르검(Guar gum), 로커스트빈검(Locust bean gum), 잔탄검(Xanthan gum), 겔란검(Gellan gum), 아라비아검(Arabic gum) 등을 언급할 수 있다. 이들 수계코팅기제는 수용성이거나 수분산성이므로 물을 용매로 사용하여 간단히 코팅을 수행할 수 있다는 장점을 지니고 있으며, 이는 인체에 유해할 뿐아니라 유산균의 안정성에 매우 치명적인 유기용매를 사용하지 않아도 되고 이에 따라 유기용매의 잔존여부에 대한 문제가 근본적으로 해결된다는 점에서 매우 중요하다. 더구나, 기존에 유산균의 코팅기제로 사용되어온 대부분의 고분자물질들이 이를 용해시키기 위하여 반드시 유기용매를 사용해야만 했던 것과는 명확히 구별되는 장점이다. 본 발명에서 바람직하게는 알긴산나트륨을 1 차 수계코팅기제로 사용하는데, 특히 알긴산 나트륨은 수용성일뿐아니라 물에 용해되었을 때 중성을 나타내므로 유산균의 안정화에 더욱더 유리하다.
씨드는 유산균자체일 수도 있고, 유산균을 올리고당류, 당알콜류, 글루콘산칼슘, 젖산칼슘 및 글루콘산 중에서 선택된 1 종 이상의 물질과 혼합한 것일 수 있다. 이들 부가물질들은 타균주를 억제하고 유산균만을 활성화시키거나, 유산균의 증식을 향상시킬 목적으로 첨가한다.
1 차 코팅화에서 수계코팅기제는 씨드를 기준으로하여 1 내지 80중량% 범위의 양으로 사용하는 것이 좋다.
이상 설명한 바에 따라 1 차 코팅된 유산균 코팅과립은 그 자체로 사용되어도 충분히 효과적일 수 있지만, 이를 통상의 방출제어형 코팅기제로 2 차 코팅하여 사용하면 더욱더 효과적으로 사용될 수 있다. 따라서, 이와 같이 1 차 및 2 차 코팅이 완료된 미세 장용성 코팅과립도 본 발명의 범위에 포함된다.
2 차 코팅기제로는 방출제어형 코팅기제를 사용하는데, 구체적으로는 일반의약품용 장용성 코팅기제; 카보폴, 아라비아검과 같이 팽윤을 목적으로 하는 코팅기제; 및 그밖의 방출제어형 코팅기제를 사용할 수 있다. 좀더 구체적으로는 옥수수단백추출물(USP/NF 수재) 및 그의 인위적 가공물질, 예를들어 Zein-DP 또는 프롤라민(prolamin), 알긴산나트륨, 알긴산, 폴리메틸메타크릴레이트, 예를들어 Eudragit L30D, Eudragit LS30D, Kollicoat MP(BASF사 제조) 등, 쉘락, 하이드록시프로필메틸셀룰로오스프탈레이트(HPMCP), 하이드록시프로필메틸셀룰로오스(HPMC), 하이드록시프로필메틸셀룰로오스아세테이트숙시네이트(HPMCAS), 카복시메틸셀룰로오스(CMC), 하이드록시프로필셀룰로오스(HPC), 셀룰로오스아세테이트프탈레이트(CAP), 에틸셀룰로오스(EC), 메틸셀룰로오스(MC), 대두단백, 소맥단백(대두단백 및 소맥단백은 식품첨가물공정에 수재), 키틴, 키틴산, 한천, 카라기난, 펙틴, 카보폴, 검류, 예를들어 구아르검, 로커스트빈검, 잔탄검, 겔란검, 아라비아검 등을 언급할 수 있다. 이중에서도 옥수수단백추출물, 하이드록시프로필메틸셀룰로오스프탈레이트(HPMCP) 및 쉘락 중에서 선택된 1 종 이상의 물질을 2 차 코팅기제로서 바람직하게 사용한다.
2 차 코팅을 수행하는 경우에는 상기 언급된 코팅기제중 선택된 1 종 이상의 물질을 1 차 코팅된 과립을 기준으로하여 1 내지 95중량% 범위로 사용한다. 특히, 일반의약품용 장용성 코팅기제는 1 내지 40중량% 범위로 사용하고 팽윤 등을 목적으로 하는 그밖의 코팅기제는 30 내지 95중량% 범위로 사용하는 것이 바람직하다. 어떤 코팅기제를 어떤 양으로 사용하는가 하는 것은 코팅기제의 종류 및 그 사용목적에 따라 당업자에 의하여 적절한 범위로 조절될 수 있다.
1 차 코팅시에 유산균 보호측면에서 물만을 용매로 사용하는 것과는 달리 2 차 코팅시에는 물, 알콜, 아세톤, 아세토니트릴, 메틸렌클로라이드, 에테르, 헥산, 클로로포름, 1,4-디옥산, 테트라하이드로푸란, 디메틸설폭사이드, 에틸아세테이트 및 메틸아세테이트 중에서 선택된 1 종 이상의 다양한 용매를 사용할 수 있다. 또한, 코팅기제가 용매중에 잘 용해되지 않는 경우에는 경우에 따라 아세트산, 염산, 인산, 각종 완충액, 구연산, 주석산, 사과산 등의 pH 조절제를 사용하여 pH를 원하는 범위로 조절함으로써 용해도를 증가시킬 수 있으며, 이는 당업자에 의하여 용이하게 수행될 수 있다.
1 차 및 2 차 코팅을 수행하는 경우, 각 코팅기제로는 서로 다른 것을 사용한다. 또한, 1 차 또는 2 차 코팅시 필요에 따라 폴리에틸렌글리콜류, 미바세트(myvacet), 프로필렌글리콜, 글리세린, 구연산트리에틸, 트리아세틴, 세틸알콜, 스테아릴알콜 중에서 선택된 1 종 이상의 가소제를 사용할 수 있다. 가소제는 코팅에 사용된 코팅기제를 기준으로하여 1 내지 50중량% 사용하는 것이 바람직하다.
본 발명에 따른 코팅과립을 제조함에 있어서, 본 발명자들은 유산균의 안정화를 극대화시키기 위하여, (a) 원료 유산균을 함유하는 씨드를 부유시키면서 코팅액을 분무하여 코팅하거나 (b) 코팅액중에 씨드를 분산시켜 코팅액과 함께 챔버내에 분산시키는 방법을 사용하였다. 따라서, 이러한 제조방법을 적용함으로써 보다 바람직하게 본 발명을 수행할 수 있다.
상기 코팅을 수행하기 위하여 유동층조립기, CF-그래뉼레이터 또는 이와 유사한 장치를 사용할 수 있으며, 본 발명에서는 유동층조립기(SFC-MINI, Freund co., Japan)를 바람직하게 사용하였다. 이 장치를 이용하여 제조할 경우, 유입공기의 온도는 35 내지 70℃의 범위로 유지시키고, 유입공기의 풍량은 3 내지 10㎥/분, 배기풍량은 5 내지 10㎥/분, rotor 회전수는 150 내지 800rpm, 교반기의 회전수는 200 내기 900rpm, lump breaker의 회전수는 1500rpm 이상, 분무액의 속도는 7 내지 25㎖/분, 분사공기의 풍량은 10 내지 50㎥/분으로 유지시키는 것이 바람직하다. 또한, 단계별 장치내 과립의 온도는 모두 35℃ 이상으로 되어야 흡습과 과립끼리의 응집을 막을 수 있고, 바람직하게는 모든 공정을 통하여 35 내지 55℃의 범위를 유지시킨다. 왜냐하면, 55℃를 초과하는 경우에는 유산균이 공정중 사멸될 수 있기 때문이다. 그러나, 초기 코팅공정을 신속히 수행함으로서 유산균의 사멸을 방지할 수 있으면 70℃까지의 온도에서도 코팅화를 수행하는 것이 가능하다.
하기 실시예를 통하여 본 발명을 보다 구체적으로 설명한다. 그러나, 이들 실시예는 본 발명에 대한 이해를 돕기위한 것일 뿐, 어떤 의미로든 본 발명의 범위가 이들 실시예로 한정되는 것은 아니다.
실시예 1
(A) 1차 코팅
씨드 : 락토바실러스 애시도필루스(Lactobacillus acidophilus):
락토바실러스 비피더스(Lactobacillus bifidus):
스트렙토코커스 패칼리스(Streptococcus faecalis)
= 1:1:1(w/w/w) 혼합물 250g
코팅액 : 알긴산나트륨 3g
물 300㎖
(B) 2차 코팅
씨드 : (A)의 코팅과립 253g
코팅액 : Zein-DP(옥수수단백추출물 가공물질) 50g
80% 에탄올 500㎖
글리세린 5㎖
i) 1차 코팅과립의 제조
유산균을 유동층조립기에서 부유시키면서 상기 코팅액을 분사시켜 코팅을 수행하였다. 이때, 유동층조립기의 작동조건은 하기 표 1에 나타낸 바와 같이 조정하였으며, 특히 유산균분말, 코팅된 분말의 장치내 온도가 35 내지 55℃를 벗어나지 않도록 유의하였다.
ii) 2차 코팅과립의 제조
i)의 방법에 따라 1차 코팅된 과립을 유동층조립기에서 부유시키면서 상기 Zein-DP 와 80% 에탄올로 구성된 코팅액을 분사하여 코팅하였다. 코팅액에는 또한 가소제의 용도로서 글리세린을 첨가하여 사용하였다. 유동층조립기의 작동조건은 하기 표 1에 나타낸 바와 같이 조정하였으며, 특히 유산균분말, 코팅된 분말의 장치내 온도가 35 내지 55℃를 벗어나지 않도록 유의하였다.

실험예 1
본 발명에 따른 실시예 1 내지 8의 코팅과립이 인공위액과 인공장액[USP(미국약전)의 기준에 준하여 제조한 것을 사용함]에서 어떤 변화를 일으키는지 조사하기 위하여 다음과 같이 시험관내 실험을 수행하였으며, 그 결과를 시판중인 빙그레사의 닥터캡슐 및 셀바이오텍사의 비피더스균원말-1(108배)과 비교하였다.
먼저 각 코팅유산균 10g을 인공위액 100㎖중에서 1시간동안 50rpm으로 교반한다음, 잔류물을 인공장액 100㎖로 옮겼다. 인공장액에서는 5시간동안 가볍게 교반한 후 배양하여(배지: Elliker broth; 배양조건: 혐기성, 37℃, 72시간) 유산균의 붕해정도를 조사하였으며, 이때 붕해정도는 육안으로 관찰하여 해면상으로 보이는 시점을 기준으로 하였다. 한편, 하기 표 2에서 붕해율은 코팅과립이 100% 붕해되는 시점을 나타내며, 생존율은 위액에서 1 시간 및 장액에서 5 시간 교반한 후 배양하여 수득된 균수를 장액에서만 5 시간 교반한 후 배양하여 수득된 유산균 균수로 나누고 100을 곱한 결과치이다. 표 2의 결과는 모두 3 회 반복수행하여 얻은 결과치의 평균값이다.

상기 표 2의 결과로부터 알 수 있듯이, 본 발명에 따라 코팅된 유산균 과립은 기존의 시판제품에 비해 인공위액에서 월등히 높은 생존율을 나타냄과 동시에 인공장액에서는 신속히 붕해되는 것을 알 수 있었다. 따라서, 본 발명에 따라 제조된 유산균과립은 유산균의 생체내 활성을 가장 바람직하게 통제할 수 있는 최적의 형태라고 생각된다.

Claims (14)

  1. 유산균을 함유한 씨드(seed)를 수계코팅기제로 1 차 코팅하여 제조된 것임을 특징으로하는 장용성 코팅과립.
  2. 제 1 항에 있어서, 유산균이 스트렙토코커스(Streptococcus)속, 락토코커스(Lactococcus)속, 루코노스톡(Leuconostoc)속, 페디오코커스(Pediococcus)속, 엔테로코커스(Enterococcus)속, 락토바실러스(Lactobacillus)속, 비피도박테리움(Bifi- dobacterium) 속의 균주중에서 선택된 1 종 이상인 코팅과립.
  3. 제 1 항에 있어서, 수계코팅기제가 알긴산나트륨, 알긴산, 폴리메틸메타크릴레이트, 소맥단백, 대두단백, 메틸셀룰로오스(MC), 하이드록시프로필셀룰로오스(HPC), 하이드록시프로필메틸셀룰로오스(HPMC), 구아르검(Guar gum), 로커스트빈검(Locust bean gum), 잔탄검(Xanthan gum), 겔란검(Gellan gum) 및 아라비아검(Arabic gum) 중에서 선택된 1 종 이상인 코팅과립.
  4. 제 3 항에 있어서, 수계코팅기제가 알긴산나트륨인 코팅과립.
  5. 제 3 항 또는 4 항에 있어서, 수계코팅기제가 씨드를 기준으로하여 1 내지 80중량% 양으로 사용된 코팅과립.
  6. 제 1 항에 있어서, 씨드가 유산균 이외에 올리고당류, 당알콜류, 글루콘산칼슘, 젖산칼슘 및 글루콘산 중에서 선택된 1 종 이상의 물질을 함유하는 코팅과립.
  7. 제 1 항에 있어서, 1 차 수계코팅을 수행한 다음, 방출제어형 코팅기제를 사용하여 2 차 코팅을 수행하여 제조된 것임을 특징으로하는 코팅과립.
  8. 제 7 항에 있어서, 방출제어형 코팅기제가 옥수수단백추출물 및 그의 인위적 가공물질, 알긴산나트륨, 알긴산, 폴리메틸메타크릴레이트, 쉘락, 하이드록시프로필메틸셀룰로오스프탈레이트(HPMCP), 하이드록시프로필메틸셀룰로오스(HPMC), 하이드록시프로필메틸셀룰로오스아세테이트숙시네이트(HPMCAS), 카복시메틸셀룰로오스(CMC), 하이드록시프로필셀룰로오스(HPC), 셀룰로오스아세테이트프탈레이트(CAP), 에틸셀룰로오스(EC), 메틸셀룰로오스(MC), 대두단백, 소맥단백, 키틴, 키틴산, 한천, 카라기난, 펙틴, 카보폴, 구아르검, 로커스트빈검, 잔탄검, 겔란검 및 아라비아검 중에서 선택된 1 종 이상인 코팅과립.
  9. 제 8 항에 있어서, 방출제어형 코팅기제가 옥수수단백추출물, 하이드록시프로필메틸셀룰로오스프탈레이트(HPMCP), 및 쉘락 중에서 선택된 1 종 이상인 코팅과립.
  10. 제 8 항 또는 9 항에 있어서, 방출제어형 코팅기제가 1 차 코팅된 과립을 기준으로하여 1 내지 95중량% 양으로 사용된 코팅과립.
  11. 제 7 항에 있어서, 물, 알콜, 아세톤, 아세토니트릴, 메틸렌클로라이드, 에테르, 헥산, 클로로포름, 1,4-디옥산, 테트라하이드로푸란, 디메틸설폭사이드, 에틸아세테이트 및 메틸아세테이트 중에서 선택된 1 종 이상의 용매를 사용하여 제조된 코팅과립.
  12. 제 1 항 또는 7 항에 있어서, 폴리에틸렌글리콜류, 미바세트(myvacet), 프로필렌글리콜, 글리세린, 구연산트리에틸, 트리아세틴, 세틸알콜, 스테아릴알콜 중에서 선택된 1 종 이상의 가소제가 코팅기제에 혼합된 코팅과립.
  13. 제 12 항에 있어서, 가소제가 코팅기제를 기준으로하여 1 내지 50중량% 사용된 코팅과립.
  14. 제 1 항 또는 7 항에 있어서, 35 내지 70℃ 온도범위에서 코팅된 코팅과립.
