Abstract
Description
도 2는 유전자형에 따라 분류된 apoE E4를 함유한 샘플 73개 (도 1)의 ELISA 흡광 결과를 도시한 것이다.
도 3은 APOE 유전자형에 따라 분류된, 분석 샘플 230개의 평균 흡광 결과를 도시한 것이다. 에러 막대는 평균의 표준 오차이다.
도 4는 APOE e4 비-보유체 (E3/E3), 및 이형접합체 (E3/E4) 및 동형접합체성 (E4/E4) APOE e4 보유체에서 여러가지 혈장 희석 수준에서의 apoE E4 / 총 apoE 흡광도 (4E4/pan-apoE) 비율을 도시한 것이다.
도 5는 ELISA 플레이트 포맷에서 폴리스티렌 표면에 대한 혈장 샘플 유래 아포지질단백질 (1:200)의 결합성을 도시한 것이다.
도 6은 ELISA 플레이트에 대한 ApoE의 결합에 있어 염 (NaCl 0-2.4 M)이 함유된 여러 완충제 또는 디터전트 (폴리소르베이트 20 -Tween 20- 또는 Triton X-100, 0-0.5%)의 효과를 도시한 것이다. 진회색 및 연회색 막대는 각각 BSA- 또는 Superblock 차단 용액으로 차단된 웰에서 샘플 분석 결과를 표시한다.

Claims (17)
- 샘플에서 apoAI, apoAIV, apoCI, apoCII, apoCIII, apoE 및 apoJ 로 이루어진 군으로부터 선택되는 아포지질단백질을 검출 및/또는 정량하기 위한 시험관내 방법으로서,
(i) 샘플을 폴리스티렌 표면과 접촉시켜, 아포지질단백질과 폴리스티렌 표면 사이의 직접적인 정전기적 및/또는 소수성 상호작용을 통해 아포지질단백질을 폴리스티렌 표면에 결합시키는 단계;
(ii) 단계 (i)에서 형성된 상기 아포지질단백질이 결합된 표면에, 상기 아포지질단백질에 특이적인 항체를 접촉시켜, 상기 항체 및 상기 아포지질단백질 간에 복합체를 형성하는 단계; 및
(iii) 단계 (ii)에서 형성된 상기 복합체를 검출하는 단계를 포함하며,
상기 폴리스티렌 표면이 상기 샘플과의 접촉 전에 알부민, 카제인, 젤라틴 또는 디터전트로 이루어진 목록에서 선택되는 화합물로 차단되는, 방법. - 샘플에서 아포지질단백질의 전체 함량에 대해, apoAI, apoAIV, apoCI, apoCII, apoCIII, apoE 및 apoJ 로 이루어진 군으로부터 선택되는 아포지질단백질의 이소형의 상대적인 함량을 측정하기 위한 시험관내 방법으로서,
(i) 샘플을 폴리스티렌 표면과 접촉시켜, 아포지질단백질과 폴리스티렌 표면 사이의 직접적인 정전기적 및/또는 소수성 상호작용을 통해 아포지질단백질을 폴리스티렌 표면에 결합시키는 단계;
(ii) 단계 (i)에서 형성된 상기 아포지질단백질이 결합된 표면에, 상기 샘플에 존재하는 상기 아포지질단백질의 이소형에 특이적인 제1 항체 및 상기 아포지질단백질의 모든 이소형에 결합할 수 있는 제2 항체를 접촉시켜, 상기 제1 항체 및 상기 아포지질단백질의 이소형을 포함하는 제1 복합체 및 상기 제2 항체와 상기 아포지질단백질의 모든 이소형을 포함하는 제2 복합체를 형성하는 단계;
(iii) 단계 (ii)에서 형성된 상기 제1 복합체 및 제2 복합체를 검출하는 단계; 및
(iv) 단계 (iii)에서 수득한 상기 제1 복합체 및 제2 복합체의 수준을 기초로 아포지질단백질 전체 함량에 대한 상기 아포지질단백질의 이소형의 상대적인 함량을 측정하는 단계를 포함하며,
상기 폴리스티렌 표면이 상기 샘플과의 접촉 전에 알부민, 카제인, 젤라틴 또는 디터전트로 이루어진 목록에서 선택되는 화합물로 차단되는, 방법. - 제1항 또는 제2항에 있어서,
상기 아포지질 단백질의 이소형이 apoE E4인, 방법. - 제1항 또는 제2항에 있어서,
상기 알부민이 소 혈청 알부민 (BSA)인, 방법. - 제1항 또는 제2항에 있어서,
상기 디터전트가 비-이온성 디터전트인, 방법. - 제1항 또는 제2항에 있어서,
상기 아포지질 단백질이 apoE E4 이고, 알부민이 소 혈청 알부민 (BSA)인, 방법. - 제2항에 있어서,
단계 (i)에서 사용되는 상기 표면이 복수의 엔티티 (entity)에 의해 형성되며,
단계 (ii)에서 상기 제1 항체 및 제2 항체를 접촉시키는 단계는, 상기 형성하는 복수의 엔티티의 일부를 각각 함유한, 별개의 용기에서 수행되며,
단계 (iii)에서 검출 단계는 각각의 용기에서 개별적으로 수행되는, 방법. - 제1항 또는 제2항에 있어서,
상기 폴리스티렌 표면이 상기 단계 (i) 이후에 세척 용액으로 처리되는, 방법. - 개체에서 신경퇴행성 질환의 발병 가능성을 확인하는 방법으로서,
상기 개체 유래 샘플에서 apoE E4 이소형의 수준을 제1항 또는 제2항에 따른 방법에 의해 결정하는 단계를 포함하며,
상기 apoE E4의 수준이 기준값 보다 높을 경우, 이는 상기 개체가 신경퇴행성 질환을 앓을 가능성이 높다는 것을 의미하는 것인, 방법. - 제9항에 있어서,
상기 신경퇴행성 질환이 알츠하이머 질환 (AD), 대뇌 아밀로이드 혈관병증 (cerebral amyloid angiopathy, CAA), 다운 증후군성 치매, 혈관성 치매, 파킨슨 질환, 루이소체 치매 및 크로이츠펠트-야콥 질환으로 이루어진 군으로부터 선택되는, 방법. - 키트로서,
i) 알부민으로 차단된 폴리스티렌 표면; 및
ii) 아포지질단백질 또는 이의 이소형에 특이적인 항체를 포함하는,
제1항 또는 제2항의 방법에 따른 아포지질단백질 apoE의 검출 및/또는 정량화를 위한 시험관내 사용을 위한 키트. - 제11항에 있어서,
상기 키트는 상기 아포지질단백질의 모든 이소형에 결합할 수 있는 제2 항체를 더 포함하는, 키트. - 제11항에 있어서,
