Claims (20)
- (i) 서열 번호 1에서 규정되는 전장(full length) 아미노산 시퀀스에 대하여 적어도 95% 또는 99% 상동성을 갖고, (ii) 서열 번호 1에 비하여 치환 D203C + S248C을 갖고, 여기에서 포지션은 서열 번호 1에서 규정되는 아미노산 시퀀스를 기준으로 번호가 매겨지며, (iii) 폴리에스테르 분해 활성을 갖고, 및 (iv) 서열 번호 1의 에스테라아제에 비하여 증가된 열안정성(thermostability)을 나타내는, 에스테라아제 변종(esterase variant).
- 제1항에 있어서,
E173, L202, N204 및 F208로부터 선택되는 하나 이상의 포지션에서의 치환을 더 포함하고, 여기에서 포지션은 서열 번호 1에서 규정되는 아미노산 시퀀스를 기준으로 번호가 매겨지는, 에스테라아제 변종. - 제2항에 있어서,
E173A/R, L202R, N204D 및 F208W로부터 선택되는 하나 이상의 치환을 포함하는, 에스테라아제 변종. - 제1항 또는 제2항에 있어서,
T61, Y92 및 V177로부터 선택되는 포지션에서의 서열 번호 1에 비하여 하나 또는 그 이상의 아미노산 치환(들)을 포함하고, 여기에서 포지션은 서열 번호 1에서 규정되는 아미노산 시퀀스를 기준으로 번호가 매겨지고, 상기 치환은 T61A/G, Y92A 및 V177A와는 다른, 에스테라아제 변종. - 제4항에 있어서,

D203C+S248C+E173R, D203C+S248C+E173A, D203C+S248C+F208W D203C+S248C+F208I, F208W + D203C + S248C + E173A, F208I + D203C + S248C + E173A 및 D203C + S248C + E173R + N204D + L202R로부터 선택되는 치환을 포함하는 에스테라아제 변종. - 제1항에서 정의되는 에스테라아제를 인코딩하는 핵산.
- 제7항의 핵산을 포함하는 발현 카세트.
- 제7항의 핵산을 포함하는 벡터.
- 제7항의 핵산을 포함하거나, 또는 상기 핵산을 포함하는 발현 카세트를 포함하거나, 또는 상기 핵산을 포함하는 벡터를 포함하는, 숙주 세포.
- (a) 제10항에 따른 숙주 세포를 에스테라아제를 인코딩하는 핵산을 발현하기에 적절한 조건하에서 배양하는 단계; 및
(b) 세포 배양물로부터 상기 에스테라아제를 회수하는 단계,
를 포함하는, 에스테라아제의 제조 방법. - 하기로 구성된 군에서 선택되는 어느 하나를 포함하는, 조성물:
- 제1항에 따른 에스테라아제, 또는
- 상기 에스테라아제를 인코딩하는 핵산, 또는
- 상기 핵산을 포함하는 발현 카세트, 또는
- 상기 핵산을 포함하는 벡터, 또는
- 상기 핵산 또는 상기 발현 카세트 또는 상기 벡터를 포함하는 숙주 세포 또는 이의 추출물. - 제12항에 있어서,
하나 또는 그 이상의 부형제 또는 첨가제를 포함하는, 조성물. - (a) 플라스틱 제품을 제1항에 따른 에스테라아제 또는 제10항에 따른 숙주 세포 또는 제12항에 따른 조성물과 접촉시켜 플라스틱 제품을 분해시키는 단계,
를 포함하는, 하나 이상의 폴리에스테르를 포함하는 플라스틱 제품을 분해하는 방법. - 제14항에 있어서,
(b) 모노머 및 올리고머를 회수하는 단계,
를 더 포함하는, 방법. - 제14항에 있어서,
상기 플라스틱 제품이 폴리에틸렌테레프탈레이트(PET), 폴리트리메틸렌테레프탈레이트(PTT), 폴리부틸렌테레프탈레이트(PBT), 폴리에티렌이소소르비드테레프탈레이트(PEIT), 폴리유산(PLA), 폴리하이드록시알카노에이트(PHA), 폴리부틸렌석시네이트(PBS), 폴리부틸렌식시네이트아디페이트(PBSA), 폴리부틸렌아디페이트테레프탈레이트(PBAT), 폴리에틸렌푸라노에이트(PEF), 폴리카프로락톤(PCL), 폴리(에틸렌아디페이트)(PEA), 폴리에틸렌나프탈레이트(PEN) 및 이들 재료의 블렌드/혼합물로부터 선택되는 하나 이상의 폴리에스테르를 포함하는, 방법. - 제16항에 있어서,
상기 플라스틱 제품이 폴리에틸렌테레프탈레이트(PET)를 포함하는, 방법. - 제14항에 있어서,
