Description
本出願は、参照によりその全体が組み込まれる、2017年7月12日に提出された英国(GB)出願第1711219.4号に対する優先権の利益を主張する。
本開示は、新しいヌクレオチドおよびオリゴヌクレオチド化合物、並びに核酸シーケンスアプリケーション(配列決定用途)におけるそれらの使用に関する。

R’は、H、一リン酸塩、二リン酸塩、三リン酸塩、チオリン酸塩、リン酸エステル類似体、反応性リン含有基に結合した-O-、またはブロッキング基によりブロッキングされた-O-であり得る。
R’’はH、OH、ホスホラミダイトまたは3’OHブロッキング基であり得、
R’’’はHまたはOHである。
ここで、R’’がホスホルアミダイトである場合、
R’は酸切断可能なヒドロキシルブロッキング基であり、自動合成条件下で後続のモノマーカップリングを可能にする。
(a)本開示の少なくとも1つの修飾ヌクレオチドをポリヌクレオチドに組み込むこと、および
(b)前記修飾ヌクレオチドに結合した色素化合物からの蛍光シグナルを検出することにより、ポリヌクレオチドに組み込まれた修飾ヌクレオチドを検出すること。

- Bは7-デアザプリン塩基である、請求項2に記載の化合物。
- 前記フルオロフォアがアルキル鎖リンカーを含む、請求項1から6のいずれか一項に記載の化合物。
- 前記アルキル鎖リンカーは、1つもしくは複数のヘテロ原子置換を有する、および/または、アミドもしくはエステル基を含む、および/またはアリール基、任意にトリアゾール基を含む、請求項7に記載の化合物。
- 前記化合物がオリゴヌクレオチドに結合している、請求項2に記載の化合物。
- 請求項2に記載の化合物で標識されたオリゴヌクレオチド。
- Bはピリミジン塩基である、請求項11に記載の化合物。
- Bは7-デアザプリン塩基である、請求項11に記載の化合物。
- 請求項11から15のいずれか一項に記載の化合物を含むオリゴヌクレオチド。
- 請求項11から15のいずれか一項に記載の化合物の2以上のコピーを含むオリゴヌクレオチド。
- 請求項11から15のいずれか一項に記載の化合物の10以上のコピーを含むオリゴヌクレオチド。
- 請求項11から15のいずれか一項に記載の化合物の100以上のコピーを含むオリゴヌクレオチド。
- 少なくとも1つのヌクレオチドが、請求項2から8のいずれか一項に記載の化合物である標識されたヌクレオチドである、2以上のヌクレオチドを含むキット。
- 少なくとも2つのヌクレオチドが、請求項2から8のいずれか一項に記載の化合物である標識されたヌクレオチドである、2以上のヌクレオチドを含む、請求項20に記載のキット。
- 少なくとも2つのヌクレオチドが、請求項2から8のいずれか一項に記載の化合物である標識されたヌクレオチドである、4つのヌクレオチドを含む、請求項20に記載のキット。
- 配列決定、発現分析、ハイブリダイゼーション分析、遺伝子分析、RNA分析、もしくはタンパク質結合アッセイ、またはそれらの組み合わせにおける請求項2から8のいずれか一項に記載の化合物の使用。
- 配列決定、発現分析、ハイブリダイゼーション分析、遺伝子分析、RNA分析、もしくはタンパク質結合アッセイ、またはそれらの組み合わせにおける請求項10に記載のオリゴヌクレオチドの使用。
