実施例1:Chrysanthellum indicumの乾燥抽出物
生または乾燥植物の地上部を、粗粉末が得られるまで機械的粉砕に供する。次いで、この粉末を、70/10の水/エタノール混合物の中で、室温で10~24時間、浸軟工程に供し、次いで、得られた混合物を、70/10の水/エタノール混合物を用いるパーコレーター中で、50℃での連続浸出に供する(植物/抽出物比は3/1である)。次いで、得られた抽出物を、ジクロロメタンまたはトリクロロメタンのような非極性の有機溶媒を用いる液体/液体洗浄に供する。35℃の低圧でのエバポレーションによる濃縮後、液体を得て、これを、24時間凍結乾燥して、水/アルコールの混合物中に溶解可能であるベージュ色の粉末を得る。この粉末(乾燥抽出物)は直接用いてもよいし、または使用前に適切な溶媒中に混合してもよい。

実施例9の組成物は、経口的に投与され得る錠剤型である。これは、組成物の総重量に対して重量パーセンテージで表した場合、15.6%のChrysanthellum americanumの地上部の乾燥抽出物、15.6%のCynara scolymusの葉の乾燥抽出物、及び23.5%のLycium barbarumの果実の乾燥抽出物を含む。これは、ビタミンB1、B2、B3、PP、B5、B6、H、B9、B12及びD3、同様に亜鉛及びクロムを含む。これはまた、賦形剤として、微結晶性セルロース及びステアリン酸マグネシウムも含む。

実施例10の組成物は、経口的に投与され得るゲルカプセルの形態である。これは、組成物の総重量に対して重量パーセンテージで表した場合、16.6%のChrysanthellum americanumの植物全体の乾燥抽出物及び16.6%のCynara scolymusの葉の乾燥抽出物、33.3%のOlea europaeaの果実の乾燥抽出物、25%のLycium barbarumの果実の乾燥抽出物を含む。この組成物はまた、Lactobacillus delbrueckii ssp Bulgaricusのような乳酸発酵物を含む。乳化剤として、非GMOチャネルからの大豆レクチン、増粘剤として、コロイド状シリカ及び脂肪酸モノグリセリド及びジグリセリドを含む。カプセルは、魚ゼラチンであり、グリセリン及び染料、赤色酸化鉄を含む。
実施例11の組成物は、経口的に投与され得るゲルカプセルの形態である。これは、Chrysanthellum americanumの地上部から、Cynara scolymusの葉から、及びLycium barbarumの果実から得られる粉末混合物の単一含水性アルコール抽出物を含む。3つの植物の間の比は、1/1/1.5である。植物の混合物の単一抽出物は、以下の工程を含むプロセスを介して得る:

実施例12の組成物は、経口的に投与され得る錠剤型である。これは、Chrysanthellum americanumの地上部、Cynara scolymusの葉、及びLycium barbarumの果実から得られた粉末混合物の単一含水性アルコール抽出物を含む。その比は1/1/1.5である。これは、また、ピぺリンを含むPiper nigrumの果実の含水性アルコール抽出物を含む。この組成物は、安定化剤として、オルトリン酸二カルシウム;固結防止剤として、脂肪酸マグネシウム塩、二酸化ケイ素及び脂肪酸;コーティング剤として、ヒドロキシプロピルメチルセルロース;染料として、Allura red AC及び酸化チタンを含むことがある。
マウスに対するインビボ実験を行って、特に総コレステロール及びLDLコレステロールならびに血漿トリグリセリドレベル、インスリン感受性及び空腹時血糖に対する、本発明による組成物の効果を実証した。
実験時間は、9週であって、1週の「ランイン(run-in)」があり、続いて、8週の植物抽出物及び組成物Xの補充があった。雄性マウスは、治療開始の時点で10週齢であった。
■C1:Chrysanthellum indicum(植物全体、餌の1%);
■C2:Cynara scolymus(葉、餌の1%)及び0.1%);
■C3:Chrysanthellum indicum(植物全体)+Cynara scolymus(葉)(それぞれ、餌の1%及び1%);
-血漿脂質(総コレステロール、LDLコレステロール、トリグリセリド)の測定;
-インスリン経口感受性試験の間の血糖の変化。この試験は、絶食状態におけるインスリン(2U/kg)の腹腔内注射から構成された。インスリン注射に応答する血糖の変化を、この注射の直前(t0)、及びその後、30、60、90及び120分後に生検によって尾で測定した。曲線下面積(AUC)を計算した。AUCの低下は、インスリン注射に対する応答がよいこと、従って、インスリン感受性の改善を反映している。逆に、AUCの増大は、インスリン感受性が劣ること、従ってインスリン耐性を反映している;
-空腹時血糖の測定。
得られた結果を以下の表4に示した。

インスリン抵抗性は、メタボリックシンドロームおよびアテローム性動脈硬化症を含む多くの病状の発症の最も重要なメカニズムの1つである(Samuel VTら、Cell、(米)、2012、Vol.148、p.852-871+Kim Bら、Experimental And Molecular Medicine、(米)、2015、Vol.47(3)、el49)。 本発明による組成物C3は、インスリン感受性を有意に改善する(図1に示す結果)。 従って、循環脂質およびインスリン感受性への作用によって、メタボリックシンドロームおよびアテローム性動脈硬化症の予防および治療のための非常に興味深い組成物となる。さらに、本発明による組成物C3で改善されたインスリン感受性は、2型糖尿病およびその合併症の予防および治療にとって特に興味深い。実際、インスリン抵抗性は、2型糖尿病の進行の主要なメカニズムの一つである(Samuel VTら、Cell、(米)、2012、Vol.148、p.852-871)。最後に、このパラメータに作用することにより、本発明の組成物C3は、一つの主な原因がインスリン抵抗性である他の病態に特に適している。これは特に、2型糖尿病の合併症の一つ、非アルコール性脂肪性肝炎(NASH;Samuel VTら、Cell、(米)、2012、Vol.148、p.852-871)およびアルツハイマー病(Bedse Gら、Frontiers Neuroscience、(米)、2015、Vol.9、p.204)の場合である。
組成物C3は、空腹時血糖を10%(対照群、537mg/dL対C3群、488mg/dL)減少させるが、2型糖尿病の治療における基準治療薬であるメトホルミンは、同じモデルにおいて、空腹時血糖を11%増大させるが、しかしマウスがより若く、その結果、上記対照マウス(対照群、414mg/dL対メトホルミン463mg/dL)と比較して空腹時血糖値が低下した実験においてである。
さらに、相乗効果は、「除草剤組合せの相乗的および拮抗的応答の計算」(Weeds、1967、15:20-22)に記載されているColby SR法に従って評価されている。この方法は、特に欧州特許第03812880号に使用されていた。各組み合わせについて、相乗効果係数を計算した。係数>1は、相乗効果の存在を示す。1未満の係数は、拮抗薬の存在を示す。実行される計算は下記のとおりである:
