Description
プのDPI製品は、併用薬剤、即ち、両方の有効成分を含む単一薬剤に重点を置いてきた。別の方法としては、国際公開第01/39823号に開示されているような装置を使用する方法がある。このような装置では、2種類の有効成分に対して別々のリザーバが設けられており、これらの有効成分は別々の空気路を介して送給される。この方法は、いくつかの利点をもたらす一方、空気路性能に関して特定の課題を提示する。これは主に、2種類の薬剤それぞれをエアロゾル化するのに空気流全体の半分しか利用できず、形状寸法が同様の単一空気路に比べて空気流の運動エネルギーも著しく減少することによる。従って、空気路設計の最適化が、このような装置には特に重要である。
使用時には空気が上流の入口端部から下流の出口端部へ引き込まれる空気路を備え、該空気路が、使用時には1用量の薬剤が該空気路に提供される薬剤提供領域と、第1空気入口と、前記薬剤提供領域から該空気路の前記出口端部まで延びるバレルとを含み、
(i)前記バレルの前記入口端部が、前記薬剤提供領域及び前記バレルの前記出口端部よりも内側寸法が小さく、前記空気路の狭窄を構成しており、
(ii)少なくとも1つの第2空気入口が設けられており、該第2空気入口が、使用時には空気が該第2空気入口から前記空気路へ前記バレルに沿った空気流の方向に対してほぼ直角の方向に入るように配置されている、

ラクソ・スミスクライン(GlaxoSmithKline)社から入手可能)のようないくつかの従来技
術の装置では、空気路の出口部分に2つの対称的な通気口が設けられている。しかしながら、例えばディスクへラー(登録商標)では、このような通気口は、空気路出口に非常に近い位置にあるため、使用時には患者の口で覆われてしまう可能性がある。従って、本発明では、前記空気入口が、通常の使用時に、使用者の唇によって又は実に使用者の身体のその他いずれの部分(例えば、装置を保持する手の指)によっても塞がれないように配置されることが好ましい。
例えば、フェノテロール、フォルモテロール、ピルブテロール、レプロテロール、リミテロール、サルブタモール、サルメテロール、テルブタリン)、非選択的β作用薬(例えば、イソプレナリン)、キサンチン気管支拡張薬(例えば、テオフィリン、アミノフィリン、テオフィリン酸コリン)、抗コリン剤(例えば、臭化イプラトロピウム、オキシトロピウム、チオトロピウム)、肥満細胞安定剤(例えば、クロモグリク酸ナトリウム、ケトチフェン)、気管支抗炎症薬(例えば、ネドクロミル・ナトリウム)、ステロイド(例えば、ジプロピオン酸ベクロメタゾン、フルチカゾン、ブデソニド、フルニソリド、トリアムシノロン、モメタゾン、シクレソニド、及び、塩若しくはその誘導体)が挙げられるが、これらに限定されない。

2つの製剤試験方法を用いて、空気路設計の製剤性能を調べた。様々な空気入口及び空気路タイプの初期開発段階における迅速スクリーニング試験には、ツイン・ステージ・インピンジャー(TSI:twin stage impinger)に基づいた粉末模倣試験を用いた。この
TSI試験によれば、微粒子用量(FPD:fine particle dose)及び微粒子割合(FPF)を測定するのに、薬剤含有開発混合物を用いるアンダーセン・カスケード・インパクタ(ACI:Andersen cascade impactor)試験をさらに最初に選択した。
・計量用量重量及び送給用量
3つの流速それぞれにおいて、2つの装置を5回作動させることにより(測定数10)、TSI薬剤模倣試験を行った。図8は、両方の用量計量部材に対する送給用量を示している。これら2つの曲線の勾配は0.016及び0.0143mg/l/分であり、30〜60l/分の範囲にわたる流速への依存度は弱いことを示している。
ACI分析により、流速60l/分における切捨て微粒子径が5μmであるという、装置の解凝集性能が実証された。ステロイド薬剤混合物の平均FPFは、7mm3用量計量
部材では38.5%、14mm3用量計量部材では33.5%であった。気管支拡張薬剤
混合物に関しては、平均FPFは42.3%であった。
この空気路は、低流速(<30l/分)において乱空気流を生成することが分かった。装置にわたる2、4及び6kPaの圧力降下及び比較的遅い流速において、この空気路は効率的で且つ流速に依存しない製剤性能をもたらすことができた。
Claims (23)
- 使用時に空気が上流の入口端部から下流の出口端部へ引き込まれる空気路を備えるドライパウダー吸引器であって、該空気路は、薬剤提供領域を含む密閉されたチャンバを備え、前記ドライパウダー吸引器が、更に、1用量の薬剤粉末を、該薬剤粉末が空気流に取り込まれる前に前記チャンバの壁部にある凹部又は第3の開口部に送給する機構を備えることにより、使用時に1用量の薬剤粉末が薬剤提供領域の該空気路に提供され、次に、前記1用量の薬剤粉末がこの空気路を通過する空気流に取り込まれ、空気路は、前記チャンバの壁が保持し第1の開口部となる第1空気入口を有し、該空気路は密閉されたチャンバの壁、及び該薬剤提供領域から該空気路の前記出口端部まで延びるバレルを含み、該バレルの前記入口端部が、前記薬剤提供領域及び該バレルの前記出口端部よりも内側寸法が小さく、前記空気路の狭窄を構成しており、
前記空気路に前記チャンバの壁が保持し第2の開口部となる少なくとも1つの第2空気入口がさらに設けられており、使用時には空気が該第2空気入口から前記薬剤提供領域へと入り、前記第2空気入口から前記薬剤提供領域へと流れる空気流は、前記第1空気入口から前記薬剤提供領域へと流れる空気流及び前記バレルに沿って流れる空気流の各方向に対して直角の方向となる、

- 前記薬剤が、前記チャンバの前記壁部にある凹部又は開口部に送給されることによって前記空気路に提供される、請求項2に記載の吸入器。
- 前記第1空気入口が、前記チャンバの前記壁部にあるスロットである、請求項2又は請求項3に記載の吸入器。
- 前記スロットが、前記バレルの長手方向軸を横断するように配置されている、請求項4に記載の吸入器。
- 前記スロットが、前記チャンバの、前記1用量の薬剤が提供される壁部とは反対側の壁部に形成されることにより、空気が、前記チャンバの前記薬剤が提供される前記壁部に向けた該空気の運動成分によって前記チャンバに引き込まれる、請求項4又は請求項5に記載の吸入器。
- 前記第2空気入口がスロットである、請求項2〜請求項6のいずれか一項に記載の吸入器。
- 前記第2空気入口が、前記チャンバの、前記薬剤が提供される前記壁部に対して直角である壁部に設けられている、請求項2〜請求項7のいずれか一項に記載の吸入器。
- 前記バレルの前記入口端部の内径が2mm〜4mmであり、前記バレルの前記出口端部の内径が4mm〜8mmである、請求項1〜請求項8のいずれか一項に記載の吸入器。
- 前記バレルの内壁に溝又は羽根が設けられている、請求項1〜請求項9のいずれか一項に記載の吸入器。
- 前記第1空気入口の断面積と前記第2空気入口の断面積の比が、10:1〜1:1である、請求項1〜請求項10のいずれか一項に記載の吸入器。
- 前記第1空気入口の断面積と前記第2空気入口の断面積の比が、5:1〜2:1である、請求項11に記載の吸入器。
- 使用時に空気が上流の入口端部から下流の出口端部へ引き込まれる空気路を備えるドライパウダー吸引器であって、該空気路は、薬剤提供領域を含む密閉されたチャンバを備え、前記ドライパウダー吸引器が、更に、1用量の薬剤粉末を、該薬剤粉末が空気流に取り込まれる前に前記チャンバの壁部にある凹部又は第3の開口部に送給する機構を備えることにより、使用時に1用量の薬剤粉末が薬剤提供領域の該空気路に提供され、次に、前記1用量の薬剤粉末がこの空気路を通過する空気流に取り込まれ、空気路は、前記チャンバの壁が保持し第1の開口部となる第1空気入口を有し、該空気路は密閉されたチャンバの壁、及び該薬剤提供領域から該空気路の前記出口端部まで延びるバレルを含み、該バレルの前記入口端部が、前記薬剤提供領域及び該バレルの前記出口端部よりも内側寸法が小さく、前記空気路の狭窄を構成しており、

- 前記第1空気入口及び前記第2空気入口が、通常の使用時に使用者によって塞がれないように配置されている、請求項1〜請求項17のいずれか一項に記載の吸入器。
- 前記バレルの内径が、前記バレルの前記入口端部から前記出口端部へ向けて徐々に大きくなっている、請求項1〜請求項18のいずれか一項に記載の吸入器。
- β2作用薬、非選択的β作用薬、キサンチン気管支拡張薬、抗コリン剤、肥満細胞安定剤、気管支抗炎症薬、ステロイドから選択される薬剤が充填された、請求項1〜請求項19のいずれか一項に記載の吸入器。
