Claims (1)
- 【特許請求の範囲】 1. アミノ酸配列AQQQDQH(SEQ ID NO:17)を含むアミノ酸数約7〜約60の 単離ペプチド。 2. 前記ペプチドがアミノ酸数約10の長さである請求項1に記載の単離ペプ チド。 3. 前記ペプチドがアミノ酸数約20の長さである請求項1に記載の単離ペプ チド。 4. 前記ペプチドがアミノ酸数約30の長さである請求項1に記載の単離ペプ チド。 5. 前記ペプチドがアミノ酸数約40の長さである請求項1に記載の単離ペプ チド。 6. 前記ペプチドがアミノ酸数約50の長さである請求項1に記載の単離ペプ チド。 7. 前記ペプチドがアミノ酸数約60の長さである請求項1に記載の単離ペプ チド。 8. 前記ペプチドがアミノ酸配列YELAQQQDQH(SEQ ID NO:18)の少なくとも 連続約16個の残基である請求項1に記載の単離ペプチド。 9. (a)アミノ酸配列AQQQDQH(SEQ ID NO:17)を含むアミノ酸数約7〜 約60の単離ペプチド及び(b)製剤学的に容認できる緩衝液又は希釈液を含有す る抗原性合成物。 10. 前記抗原性合成物がさらに前記ペプチドに複合された担体を含む請求項 9に記載の抗原性合成物。 11. 前記担体がKLH(鍵穴付着ヘモシアニン)、ジフテリアトキソイド、破傷 風トキソイド又はCRM197である請求項10に記載の抗原性合成物。 12. さらにアジュバントを含む請求項9に記載の抗原性合成物。 13. 前記アジュバントが脂質を含む請求項12に記載の抗原性合成物。 14. 前記ペプチドが第2抗原に共有的に結合している請求項9に記載の抗原 性合成物。 15. 前記第2抗原がペプチド抗原である請求項14に記載の抗原性合成物。 16. 前記第2抗原が非ペプチド抗原である請求項14に記載の抗原性合成物。 17. 前記単離ペプチドがアミノ酸配列YELAQQQDQH(SEQ ID NO:18)の少なく とも連続約16個の残基を含む請求項9に記載の抗原性合成物。 18. アミノ酸配列AQQQDQH(SEQ ID NO:17)を含むアミノ酸数約7〜約60の 単離ペプチド及び製剤学的に容認できる緩衝液又は希釈液を含有するワクチン合 成物。 19. 前記単離ペプチドがさらにアミノ酸配列YELAQQQDQH(SEQ ID NO:18)の 少なくとも連続約16個の残基を含むと定義されている請求項18に記載のワクチン 合成物。 20. 哺乳類に(a)アミノ酸配列AQQQDQH(SEQ ID NO:17)を含むアミノ酸 数約7〜約60の単離ペプチド及び(b)製剤学的に容認できる緩衝液又は希釈液 を含有する抗原性合成物を提供するステップを含む前記哺乳類において免疫反応 を誘発するための方法。 21. 前記単離ペプチドがアミノ酸配列YELAQQQDQH(SEQ ID NO:18)の少なく とも連続約16個の残基を含む請求項20に記載の方法。 22. M.Catarrhalis単離菌O35EのUspA1抗原(SEQ ID NO:1)をコードする核 酸。 23. M.Catarrhalis単離菌O35EのuspA1 DNA配列(SEQ ID NO:2)を有する核 酸。 24. M.Catarrhalis単離菌O35EのUspA2抗原(SEQ ID NO:3)をコードする核 酸。 25. M.Catanhalis単離菌O35EのuspA2 DNA配列(SEQ ID NO:4)を有する核 酸。 26. M.Catarrhalis単離菌O46EのUspA1抗原(SEQ ID NO:5)をコードする核 酸。 27. M.Catarrhalis単離菌O46EのuspA1 DNA配列(SEQ ID NO:6)を有する核 酸。 28. M.Catarrhalis単離菌TTA24のuspA1 DNA配列順序(SEQ ID NO:10)を有 する核酸。 32. M.Catarrhalis単離菌TTA24のUspA2抗原(SEQ ID NO:11)をコードする 核酸。 33. M.Catarrhalis単離菌TTA24のuspA2 DNA配列(SEQ ID NO:12)を有する 核酸。 34. M.Catarrhalis単離菌TTA37のUspA1抗原(SEQ ID NO:13)をコードする 核酸。 35. M.Catarrhalis単離菌TTA37のuspA1 DNA配列(SEQ ID NO:14)を有する 核酸。 36. M.Catarrhalis単離菌TTA37のUspA2抗原(SEQ ID NO:15)をコードする 核酸。 37. M.Catarrhalis単離菌TTA37のuspA2 DNA配列(SEQ ID NO:16)を有 する核酸。 38. サンプル中における前記UspA1又はUspA2抗原のエピトープのコア配列の 残基に相当するM.Catarrhalisアミノ酸配列の存在を測定するステップを含むM .Catarrhalis感染症を診断するための方法。 39. 前記測定がPCRを含む請求項38に記載の方法。 40. 前記測定がM.Catarrhalis抗原に対する抗体の免疫学的反応性を含む請 求項38に記載の方法。 41. アミノ酸配列AQQQDQH(SEQ ID NO:17)を含むアミノ酸数約7〜約60の 単離ペプチドを個人に提供することを含むM.Catarrhalis感染症に罹患した前記 個人を治療するための方法。 42. 前記ペプチドがアミノ酸配列YELAQQQDQH(SEQ ID NO:18)の少なくとも 約16個の連続残基を含む請求項41に記載の単離ペプチド。 43. 被験者にアミノ酸配列AQQQDQH(SEQ ID NO:17)によって形成されたエ ピトープと免疫学的に反応する抗体を提供することを含むM.Catarrhalis感染症 を予防又は制限するための方法。 44. 前記エピトープがさらにアミノ酸配列YELAQQQDQH(SEQ ID NO:18)の少 なくとも連続約16個の残基を含んでいると定義されている請求項42に記載の方法 。 45. 下記のステップを含むUspA1又はUspA2に免疫学的に結合する抗体との反 応性についてペプチドをスクリーニングする方法: a)前記ペプチドを提供すること; b)前記ペプチドを前記抗体と接触させること;及び c)前記ペプチドへの前記抗体の結合を測定すること。 46. 前記抗体が17C7、45-2、13-1、29-31、16A7、17B1又は5C12である、請 求項45に記載の方法。 47. 前記抗体が17C7である請求項46に記載の方法。 48. 前記抗体が45-2である請求項46に記載の方法。 49. 前記抗体が13-1である請求項46に記載の方法。 50. 前記抗体が29-31である請求項46に記載の方法。 51. 前記抗体が16A7である請求項46に記載の方法。 52. 前記抗体が5C12である請求項46に記載の方法。 53. 前記抗体が17B1である請求項46に記載の方法。 54. 前記決定がウェスタンブロット、ELISA、さらにRIA及び免疫親和性分離 からなる群から選択されるイムノアッセイを含む請求項45に記載の方法。 55. 下記のステップを含むM.Catarrhalisに対する防御免疫反応を誘発する 能力についてUspA1又はUspA2ペプチドをスクリーニングする方法: a)前記ペプチドを提供すること; b)ペプチドを適切な形で実験動物に投与すること; c)前記動物にM.Catarrhalisを用いて抗原投与すること;及び d)M.Catarrhalisを用いて前記動物の感染をアッセイすること。 56. 前記動物がマウスであり、前記抗原投与が肺抗原投与であり、さらに前 記アッセイが前記マウスによる肺クリアランスの程度を評価することを含む請求 項56に記載の方法。 57. 前記UspA1ペプチドがM.Catarrhalisの残基約582〜604(SEQ ID NO:1) 又はM.Catarrhalis菌株O35Eと比較して類似する位置を含む請求項56に記載の方 法。 58. 前記UspA2ペプチドがM.Catarrhalisの残基約355〜377(SEQ ID NO:3) 又はM.Catarrhalis菌株O35Eと比較して類似する位置を含む請求項56に記載の方 法。 59. 前記UspA1ペプチドがM.Catarrhalisの残基約452〜642(SEQ ID NO:1) 又はM.Catarrhalis菌株O35Eと比較して類似する位置を含む請求項57に記載の方 法。 60. 前記UspA2ペプチドがM.Catarrhalisの残基約242〜415(SEQ ID NO:3) 又はM.Catarrhalis菌株O35Eと比較して類似する位置を含む請求項58に記載の方 法。 61. 単離ペプチドであって、前記ペプチドが、前記UspA1タンパク質(SEQ I D NO:1)の残基約582〜604、又はUspA2タンパク質(SEQ ID NO:3)の残基約355 〜377、又はM.Catarrhalis菌株O35Eと比較して類似する位置によって定義され た領域内に位置する残基を含むM.CatarrhalisのUspA1又はUspA2タンパク質から の少なくとも連続7個のアミノ酸を有する単離ペプチド。 62. 前記ペプチドがアミノ酸数7〜60の長さである請求項61に記載の単離ペ プチド。 63. 前記ペプチドが非UspA1又は非UspA2配列順序を含む請求項61に記載の単 離ペプチド。 64. 前記ペプチドが非M.Catarrhalis配列順序を含む請求項61に記載の単離 ペプチド。 65. 下記を含む抗原性合成物: a)単離ペプチドであって、前記ペプチドが、前記UspA1タンパク質(SEQ ID NO:1)の残基約582〜604、又はUspA2タンパク質(SEQ ID NO:3)の残基約355〜3 77、又はM.Catarrhalis菌株O35Eと比較して類似する位置によって定義された領 域内に位置する残基を含むM.CatarrhalisのUspA1又はUspA2タンパク質からの少 なくとも連続7個のアミノ酸を有する単離ペプチド。 b)製剤学的に容認できる緩衝液又は希釈液。 66. 下記を含む抗原性合成物: a)単離ペプチドであって、前記単離ペプチドが第2抗原に共有的に結合され た担 体として機能するUspA1又はUspA2のアミノ酸配列の少なくとも連続7個の残基を 含む約7〜約60個のアミノ酸からなる単離ペプチド;及び b)製剤学的に容認できる緩衝液又は希釈液。 67. 前記第2抗原がペプチド抗原である請求項66に記載の抗原性合成物。 68. 前記第2抗原が非ペプチド抗原である請求項66に記載の抗原性合成物。
