Abstract

According to the invention the artificial fabrication method for fabricating simultaneously or separately bone and nacre is implemented by the activity of osteonacrigenic factors contained in nacre. The method according to the invention consists in using fragments of nacre or powder of nacre resulting from the test of bivalve aquatic Mollusca Gasteropoda or Cephalopoda in the "in vitro" or "in vivo" presence of bone cells or bone-forming cells or cells susceptible of becoming bone-forming cells, associated to one or a plurality of bone fragments. The method according to the invention may be implemented in molds of various shapes and sizes, whereafter the bone parts thus obtained may be used as grafts in bone surgery and the nacre may be used to produce pearls, jewels or objects.

Description

PRODUCTION D'OS ET DE PERLE A PARTIR DE CULTURE DE
CELLULES FORMATRICES D'OS
BONE AND PEARL PRODUCTION FROM CULTURE OF
BONE FORMAT CELLS
The present invention relates to a process for producing natural bone and pearl in the form of a pearl from bone-forming cells and biocrystals originating from the pearly test for aquatic molluscs.

Until now, it has never been possible to artificially produce a sufficient quantity of bone tissue of animal or human origin intended to serve as a graft. The surgical circumstances which can give rise to the need to perform bone grafts are numerous: orthopedic surgery - restorative surgery - plastic surgery - carcinology etc ... In all cases the grafts come either from the patient himself and in this case we performs an autograft, that is to say a bone bank made up of bone pieces treated so as to rid them of their organic part which can cause immune reactions, sterilized by ionizing radiation and frozen, a heterograft is then practiced.

These bone pieces, once grafted, constitute a mineral framework in which a process of bone neoformation will develop.

In this way, the cultured pearl is obtained from fragments of mother-of-pearl which are introduced into a specific organ of the living mollusk. The inclusion of such a fragment produces an irritant phenomenon which itself induces a mineralization process by affixing biocrystals around the fragment of mother-of-pearl, the pearls thus obtained are necessarily of rounded shape.

The disadvantages linked to bone grafts are of several orders, however the most important are the difficulties of supply and storage as well as the immune reactions of rejection in cases of heterografts. In the case of autografts the major drawback lies in the fact that one cannot have a bone fragment of sufficient volume when it comes to replacing a great loss of substance.

This is the reason why certain inventors have proposed bone pieces produced in biologically compatible natural biomaterials such as coral. These biomaterials are in fact only calcium reserves occupying the volume left empty by the loss of substance and which little by little will be replaced by newly formed bone.

The major problem comes from the fact that the osteogenic power of the bone cells entering the mineralization process is exhausted, thereby limiting the total colonization of the biomaterial graft.

Other inventors have proposed, as a substitute for bone parts, elements machined from the pearly test of certain molluscs, but here again the limited size and volume of these machined parts makes them unsuitable for use in replacing losses of substances from the most of the long bones of the skeleton.

It is for these reasons that we have experimented with a simple artificial process for the simultaneous or separate production of bone tissue and cultured pearl.

The method according to the invention consists in implementing a histochemical process intended to produce artificially at the same time or separately on the one hand from bone of animal or human origin and on the other hand from cultured pearl.

The method according to the invention consists in bringing into a suitable culture medium a layer of bone-forming cells (osteoblasts), particles or powder of mother-of-pearl biocrystals. The bone-forming cells will preferably be derived from explants of young mammals or from fragments of human bone without this example being limiting. The biocrystals come from the pearly test of bivalve aquatic molluscs, gastropods, or cephalopods.

The method according to the invention is implemented in modules of variable size and shape, these modules will serve as molds in which the method according to the invention will develop in order to obtain replicas of bone or pearl.

According to a preferred embodiment, the method according to the invention can be used for the manufacture of bone pieces intended to serve as grafts in bone surgery; and the production of objects or pearl jewelry of variable shape and dimension without this being limiting.

This embodiment being presented by way of non-exhaustive example. It is up to the person skilled in the art to implement the cultures of bone-forming cells derived from bone explants and of biocrystals of nacreous test of aquatic mollusk whenever it will need to produce artificially or bone or cultured pearl.
