Claims (11)

  1. XYLAN ASE V ARI ANTER
    1. Fremgangsmåde til ændring af sensitiviteten for et Familie-ll-xylanasepolypeptid over for en hæmmer, hvilken fremgangsmåde omfatter: (a) modificering af én eller flere af polypeptidets aminosyrerester; hvor modifikationerne er ved en hvilken som helst af positionerne 11, 12, 13, 15, 17, 29, 31, 32, 34, 113, 114, 119, 120, 121, 122, 123, 124 eller 175 af B. sntiif/fs-aminosyresekvensen ifølge SEQ. ID. NO. 1. eller ved én eller flere ækvivalente positioner i andre homologe xylanaser, hvor mindst én af aminosyremodifikationerne er mindst ved position 11 af B. sntiif/fs-aminosyresekvensen ifølge SEQ. ID. NO. 1 eller ved én eller flere ækvivalente positioner i andre homologe xylanaser; (b) måling af xylanasepolypeptidets sensitivitet over for en xylanasehæmmer og (c) måling af xylanasepolypeptidets effekt på en melopslæmnings viskositet; hvor peptidet eller fragmentet deraf har reduceret sensitivitet over for en xylanasehæmmer sammenlignet med moderxylanaseenzymet; hvor peptidet eller fragmentet deraf endvidere sænker en melopslæmnings viskositet mere end moderxylanaseenzymet.
  2. 2. Fremgangsmåde til ændring af sensitiviteten for et Familie-ll-xylanasepolypeptid over for en hæmmer, hvilken fremgangsmåde omfatter: (a) modificering af to eller flere polypeptidets aminosyrerester; hvor modifikationerne er af to eller flere af positionerne 11, 12, 13, 15, 17, 29, 31, 32, 34, 113, 114, 119, 120, 121, 122, 123, 124 og 175 af B. subtilis-aminosyresekvensen vist som SEQ. ID. NO. 1, eller ved én eller flere ækvivalente positioner i andre homologe xylanaser; hvor én af aminosyremodifikationerne er ved position 11; (b) måling af xylanasepolypeptidets sensitivitet over for en xylanasehæmmer og (c) måling af xylanasepolypeptidets effekt på en melopslæmnings viskositet; hvor polypeptidet eller fragmentet deraf har reduceret sensitivitet over for en xylanasehæmmer sammenlignet med moderxylanaseenzymet; endvidere hvor peptidet eller fragmentet deraf sænker en melopslæmnings viskositet mere end moderxylanaseenzymet.
  3. 3. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor aminosyremodifikationen sker ved målrettet mutagenese.
  4. 4. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor én af aminosyremodifikationerne er en hvilken som helst af: D11Y, DUN, D11F, D11K, DIIS, D11W, G12F, G13F, I15K, N17K, N17Y, N17D, N29K, N29Y, N29D, S31K, S31Y, S31D, N32K, G34D, G34F, G34T, Y113A, Y113D, Y113K, N114A, N114D, N114F, N114K, D119K, D119Y, D119N, G120K, G120D, G120F, G120Y, G120N, D121N, D121K, D121F, D121A, R122D, R122F, R122A, T123K, T123Y, T123D, T124K, T124Y, T124D, Q175E, Q175S og Q175L.
  5. 5. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor én af aminosyremodifikationerne er en hvilken som helst af: D11Y, DUN, D11F, D11K, DIIS eller D11W.
  6. 6. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor én af aminosyremodifikationeme erDllF.
  7. 7. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor to af aminosyremodifikationeme er D11F og R122D.
  8. 8. Anvendelse af en xylanasepolypeptidvariant tilvejebragt ved hjælp af fremgangsmåden ifølge et hvilket som helst af kravene 1 til 7 i en fremgangsmåde til modificering af plantematerialer.
  9. 9. Anvendelse af en xylanasepolypeptidvariant tilvejebragt ved hjælp af fremgangsmåden ifølge et hvilket som helst af kravene 1 til 7 i én eller flere af: bageri, dyrefoder, stivelsesproduktion, melseparation (vådformaling) og papir- og pulpproduktion.
  10. 10. Anvendelse af en xylanasepolypeptidvariant tilvejebragt ved hjælp af fremgangsmåden ifølge et hvilket som helst af kravene 1 til 7 i én eller flere af: forarbejdning af korn, stivelsesproduktion, dyrefoder, i forarbejdning af træ, forbedring af processen til blegning af træpulp.
  11. 11. Anvendelse af en xylanasepolypeptidvariant tilvejebragt ved hjælp af fremgangsmåden ifølge et hvilket som helst af kravene 1 til 7 til at ændre viskositeten, der er afledt af tilstedeværelsen af hemicellulose eller arabinoxylan i en opløsning eller et system, der omfatter plantecellevægmateriale.
