Abstract
提供了将线性测序文库转换为环状测序文库的方法。其中,该方法包括以下步骤:利用正向接头引物和反向接头引物将线性测序文库进行PCR扩增,以便获得扩增产物;将所述扩增产物进行热变性处理,以便获得单链DNA混合物;将所述单链DNA混合物进行环化处理,以便获得环状DNA产物,所述环状DNA产物构成所述环状测序文库,其中,正向接头引物与所述线性测序文库的正向接头的至少部分序列互补配对,反向接头引物与所述线性测序文库的反向接头的至少部分序列互补配对,正向接头引物和所述反向接头引物的至少之一的5’端核苷酸经过磷酸化修饰。
