背景技术
白桦茸,别名桦褐孔菌,学名(Inonotusobliquus,Fuscoporiaoblique),是一种生长于白桦树、杨树、榆树上的大型真菌,主要生长在高纬度地区,北纬40°~50°,生长期长达15年,桦褐孔菌含有大量的抗癌、降血压、降血糖、复活免疫作用的植物纤维类多糖体,可以提高免疫细胞的活力。
多年来,白桦茸一直作为纯中药在民间用于治疗疾病,白桦茸亦被认为是21世纪的功能性保健食品,桦褐孔菌是俄罗斯、波兰、日本等国的民间常用药物。白桦茸多糖是白桦茸的重要药理活性成分之一,白桦茸的菌丝和菌核中的多糖主要由β-葡聚糖、杂多糖和蛋白复合物组成。研究表明,白桦茸多糖(肽)主要由葡萄糖、半乳糖、甘露糖、鼠李糖、木糖和树胶醛糖等糖苷组成,木糖和鼠李糖是白桦茸多糖生物活性贡献最大的单糖。白桦茸多糖的功能特性主要表现在白桦茸的水溶性和非水溶性多糖对糖尿病小鼠具有降低血糖的作用,尤其是白桦茸多糖的水提取物,它的降血糖作用可持续48h。桦褐孔菌的抗病毒作用也为降低胰岛毒性,促进胰岛素分泌起到积极作用。同时,白桦茸还拥有极高的SOD活性,白桦茸子实体中富含β-D-葡聚糖和超氧化歧化酶,其含量分别是名贵菌类-红樟菌的30倍和23倍,超氧化歧化酶的含量是灵芝的50多倍。SOD活性物质不仅能够有效地提升人体清除多种反应性氧自由基的能力,而且还能还原人体内的抗氧化系统,增强机体抗氧化能力。特别是反应氧自由基将随血糖的增高而呈正比增高。因此桦褐孔菌在抗氧化、糖代谢、糖尿病及糖尿病并发症具有治疗作用。
一种不添加辅料降血糖植物提取物片剂的加工方法以白桦茸为主要原料经水提取、浓缩、干燥得到提取物,无需制粒、不添加任何辅料直接压制成片剂。目前生产片剂的主要方法有:直接压片法、湿法制粒、干法制粒。这些方法能适应不同物料的特性,添加一些赋型剂、粘合剂等辅料就能解决大部分片剂生产的问题。按传统方式生产植物提取物片剂,就必须要制粒、烘干,还要加入一定比例赋形剂、粘合剂等,如硬脂酸镁、维晶纤维素等,这势必会降低药物的纯度,影响糖尿病人的治疗效果。
发明内容
为了解决上述的技术问题,本发明提供了一种不添加任何辅料,纯度为100%的降血糖片剂的生产方法。
为了解决上述技术问题,本发明提供了如下的技术方案:
本发明提供一种不添加辅料降血糖植物提取物片剂的加工方法,包括如下步骤:
(1)粉碎:将干燥的白桦茸子实体分别进行粉碎,得到碎末;
(2)称量、浸泡:称取白桦茸碎末投入多功能提取罐内,加入白桦茸碎末质量12倍的清水,开启搅拌器浸泡2小时;
(3)一次提取(料水比1:12):物料蒸煮过程,持续开启搅拌器,当温度升至80℃时,开启冷却水控制阀和冷却水循环泵,使蒸煮罐的汽水冷凝装置开始运行,当温度升至100℃时开始计时,让物料在罐内继续蒙蒸2hr;
(4)提取液出罐:当蒸煮时间结束时,关闭搅拌器,开启提取罐排空阀,开启真空泵,提取罐内的提取液在真空的作用下,经双联过滤器缓慢转移至贮液罐中;
(5)二次提取(第二遍料水比10:1),重复3-4步骤;
(6)提取液浓缩:浓缩过程的参数控制:负压:0.05~0.08mpa,温度:55~80℃;提取液浓缩至浓度23%,得稠浓缩液放入贮液罐,提取完成,料渣弃置;
(7)真空低温连续干燥:设定真空度-0.95~-0.99Mpa、加浆泵频率16Hz,主动轮频率32Hz,摆动布料装置设定,一区温度95℃,缷料时间15min,切断时间15min,粉碎仓筛片目数为40目;得灵芝白桦茸提取物;
(8)压片:灵芝白桦茸提取物过40目筛,投入压片机,设定压力与转盘转速,压制成0.35克/粒片剂。
作为本发明的一种优选技术方案,在步骤(1)中将干燥的白桦茸子实体分别经不锈钢多功能粉碎,得到3-5mm碎末。
与现有技术相比,本发明的创新性表现为:
(1)本发明采用真空低温连续干燥方法,所得到固体粉末未粉碎前目数为10目左右,经粉碎后可得到40目白桦茸提取物,细度适合于压制。提取物较传统高温干燥方式得到的提取物的密度大。白桦茸高温离心式喷雾干燥提取物密度为0.3-0.45g/ml,而真空低温干燥所得到的提取物密度为0.55-0.65g/ml,比重大易压制,也省去了制粒工序。
(2)本发明采用真空低温连续干燥方法,因在真空环境中干燥,游离水份能迅速排出,白桦茸提取物水分含量低,呈疏松的粉末状,所得到提取物在显微镜下观察,有较多孔隙,可不添加赋型剂和粘合剂等,在一定压力下粉末易粘合压制成型,崩解时限完全符合片剂生产要求。
(3)采用本发明对白桦茸进行提取、真空低温浓缩、低温干燥,若采用离心式高温喷雾干燥,需添加一定比例的糊精,才能完成干燥。真空低温连续干燥技术的应用,因干燥温度低,多糖成份不会因高温而产生熔融、焦化,可不添加辅料而完成干燥,具有药物成份不易被破坏、生产环节少、物料纯度为100%的特点,对于开发食药用菌的产品具有重要意义。

采用本发明对白桦茸进行提取、真空低温浓缩、低温干燥,若采用离心式高温喷雾干燥,需添加一定比例的糊精,才能完成干燥。真空低温连续干燥技术的应用,因干燥温度低,多糖成份不会因高温而产生熔融、焦化,可不添加辅料而完成干燥,具有药物成份不易被破坏、生产环节少、物料纯度为100%的特点,对于开发食药用菌的产品具有重要意义。
最后应说明的是:以上所述仅为本发明的优选实施例而已,并不用于限制本发明,尽管参照前述实施例对本发明进行了详细的说明,对于本领域的技术人员来说,其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分技术特征进行等同替换。凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (2)
1.一种不添加辅料降血糖植物提取物片剂的加工方法,其特征在于,包括以下步骤:
(1)粉碎:将干燥的白桦茸子实体分别进行粉碎,得到碎末;
(2)称量、浸泡:称取白桦茸碎末投入多功能提取罐内,加入白桦茸碎末质量12倍的清水,开启搅拌器浸泡2小时;
(3)一次提取(料水比1:12):物料蒸煮过程,持续开启搅拌器,当温度升至80℃时,开启冷却水控制阀和冷却水循环泵,使蒸煮罐的汽水冷凝装置开始运行,当温度升至100℃时开始计时,让物料在罐内继续蒙蒸2hr;
(4)提取液出罐:当蒸煮时间结束时,关闭搅拌器,开启提取罐排空阀,开启真空泵,提取罐内的提取液在真空的作用下,经双联过滤器缓慢转移至贮液罐中;
(5)二次提取(第二遍料水比10:1),重复3-4步骤;
(6)提取液浓缩:浓缩过程的参数控制:负压:0.05~0.08mpa,温度:55~80℃;提取液浓缩至浓度23%,得稠浓缩液放入贮液罐,提取完成,料渣弃置;
(7)真空低温连续干燥:设定真空度-0.95~-0.99Mpa、加浆泵频率16Hz,主动轮频率32Hz,摆动布料装置设定,一区温度95℃,缷料时间15min,切断时间15min,粉碎仓筛片目数为40目;得灵芝白桦茸提取物;
(8)压片:灵芝白桦茸提取物过40目筛,投入压片机,设定压力与转盘转速,压制成0.35克/粒片剂。
2.根据权利要求1所述的一种不添加辅料降血糖植物提取物片剂的加工方法,其特征在于,在步骤(1)中将干燥的白桦茸子实体分别经不锈钢多功能粉碎,得到3-5mm碎末。
