Abstract
La presente proporciona materiales y métodos para el tratamiento de la obesidad y del exceso de peso, diabetes, y otros trastornos metabólicos asociados. En particular, la presente proporciona péptidos análogos de glucagón acilados efectivos en tales métodos. Los péptidos pueden mediar su efecto al tener mayor selectividad para el receptor de GLP-1 en comparación con el glucagón humano. Reivindicación 1: Un compuesto que tiene la fórmula (1): R¹-P¹-P²-R² (1) donde R¹ es H, alquilo C₁₋₄, acetilo, formilo, benzoílo o trifluoroacetilo; R² es OH o NH₂; P¹ es un péptido que tiene la secuencia: H-X²-X³-GTFTSDYSKYLDSYAAHDFVEWLLSA donde: X² se selecciona de Aib, Ala, D-Ala, Ser, N-Me-Ser, Ac3c, Ac4c y Ac5c; X³ se selecciona de Gln y His; P² está ausente o es una secuencia de 1 - 20 unidades de aminoácidos seleccionados de modo independiente del grupo que consiste en Ala, Leu, Ser, Thr, Tyr, Cys, Glu, Lys, Arg, Dbu, Dpr y Orn; o una de sus sales o solvatos farmacéuticamente aceptables; Y es un residuo de Lys, Arg, Orn o Cys en el que la cadena lateral se conjuga a un sustituyente que tiene la fórmula -Z²-Z¹; -Z¹ es una cadena grasa que tiene un grupo polar en un extremo de la cadena y una conexión a Z², -X- en el extremo de la cadena distal del grupo polar, donde el grupo polar comprende un grupo ácido carboxílico o un bioisóstero del ácido carboxílico, un ácido fosfónico o un ácido sulfónico; y -X- es un enlace, -CO-, -SO--, o -SO₂-; -Z²- es un espaciador de fórmula (2), donde: cada Y es de modo independiente -NH, -NR, -S o -O, donde R es alquilo, un grupo protector o forma una unión con otra parte del espaciador Z²; cada X es de modo independiente un enlace, CO-, SO-, o SO₂-; con la condición de que cuando Y es -S, X es un enlace; cada V es de modo independiente un resto orgánico bivalente que une Y y X; y n es 1 - 10; o una de sus sales o solvatos farmacéuticamente aceptables.
