Abstract
Un método y una sonda de oligonucleótidos se describen para determinar la presencia o ausencia de alelos mutantes en un sitio genómico. La zona se une a diferentes alelos de una secuencia objetivo con diferentes temperaturas de fusión (Tm). El método determina la Tm de la sonda cuando se hibridiza a la secuencia objetivo para establecer si un ácido nucleico de variante tal un alelo mutante está presente o ausente en la secuencia objetivo. Puede haber variantes en una secuencia objetivo que no son de interés, por ejemplo, mutaciones fenotípicamente silenciosas. Para asegurar que estas variantes no influencian la Tm de la sonda, la sonda contiene sitios de base universal en donde se presentan variantes que no son de interés.
