Abstract

La presente invención proporciona composiciones, métodos y kits para discriminar variación de secuencia entre diferentes alelos. Más específicamente, en algunas modalidades, la presente invención proporciona composiciones, métodos y kits para determinar la presencia y/o nivel (por ejemplo, cuantificación) de variantes alélicas raras (por ejemplo, mutantes), tales como polimorfismos de un solo nucleótido (SNP) o inserciones o supresiones de nucleótidos, en muestras que comprendan abundantes variantes alélicas (por ejemplo, tipo silvestre) con alta sensibilidad y/o especificidad. De esta manera, en ciertas modalidades, la presente invención proporciona un método altamente selectivo para la detección de mutaciones somáticas, por ejemplo, en muestras que contengan niveles abundantes de un alelo de tipo silvestre en comparación con niveles muy bajos de un alelo mutante.
