Abstract
La invención es concerniente con pares ortogonales de tARN y aminoacil-tARN sintetasas que pueden incorporar el aminoácido no natural fenilselenocisteína a proteínas producidas en células huésped eubacterianas tales como E. coli. La invención proporciona, por ejemplo pero no limitado a, nuevas aminoacil-tARN sintetasas ortogonales, polinucleótidos que codifican las nuevas moléculas de sintetasa, métodos para identificar y enmascarar las nuevas sintetasas, métodos para producir proteínas que contienen el aminoácido no natural fenilselenocisteína y sistemas de traducción. La invención proporciona además métodos para producir proteínas modificadas (por ejemplo, proteínas lapidadas) por medio de la modificación apuntada del residuo de fenilselenocisteína en una proteína.
